非热杀菌能更好地提供产品质量,保留较好的营养成分,比传统杀菌技术更能节省能源,为开拓新的市场创造更大机遇。利用自行设计的介质阻挡放电反应器(DBDR),以鲜榨果汁为杀菌介质,研究DBDR所产生的低温等离子体(LTP)对鲜榨果汁的S.aureus的杀菌规律。选取循环次数、pH、温度和电压4个因素进行中心组合设计,利用响应面对其杀菌规律进行研究,建立了低温等离子体(LTP)杀灭橙汁中S.aureus的二次多项数学模型,验证了模型的有效性,利用Design Expert软件对其进行分析表明:循环7次,pH 2.5,温度50 ℃,电压25 kV时,杀菌率log(N0/N)最大,其预测值为5.19,与实测值相符。  相似文献

Abstract— Antagonism against Staphylococcus intermedius was investigated amongst bacteria isolated from skin and mucosae of (1) 15 dogs with pyoderma and (2) 42 healthy dogs, including household pets, laboratory beagles and guide dogs. Fifteen isolates of S. intermedius, four coagulase-negative staphylococci and a Micrococcus were inhibitory in vitro against at least one of the three indicator strains using direct and deferred inhibition tests. More dogs with antagonists were found in the pyoderma group and amongst laboratory beagles (p < 0.05) compared to healthy household pets. The frequency of all antagonists and of antagonistic S. intermedius from mucosae was higher in the pyoderma group compared to healthy household pets (p < 0.01 for both) and all healthy animals (p < 0.05 and p < 0.01, respectively). Increased competition at mucosae of infected dogs caused by higher populations of bacteria may have favoured the selection of antagonistic strains. Further studies, including tests for adherence and virulence are required to determine whether these isolates could be used as interfering strains against canine pyoderma. Résumé— L'antagonisme contre Staphylococcus intermedius a étéétudié parmi des bactéries isolées à partir de la peau et des muqueuses de 15 chiens présentant des pyodermites et de 42 chiens sains, effectif comprenant des chiens de particuliers, des beagles de laboratoire et des chiens guides. Parmi les souches isolées, 15 S. intermedius, 4 staphylocoques coagulase négatifs et un micrococcus se sont révélés inhibiteurs in vitro vis à vis d'au moins une des 3 souches indicatrices, à la fois dans des test d'inhibition directs et indirects. La plupart des chiens portcurs d'antagonistes appartenait au groupe pyodermite et au groupe beagles de laboratoire (p < 0,05) plutôt qu'au groupe chiens de particuliers sains. La fréquence de tous les antagonistes et du S. intermedius antagoniste isolés à partir des muqueuses était supérieure dans le groupe pyodermite par rapport au groupe chiens de particuliers sains (p < 0,01) pour les 2) et au groupe incluant la totalité des animaux sains (p <0,05 et p < 0,01 respectivement). Une compétition accrue au niveau des muqueuses de chiens infectés causée par un plus grand nombre de bactéries peut favoriser la sélection de souches antagonistes. D'autres études incluant des tests d'adhérence et de virulence sont nécessaires pour déterminer si ces souches isolées peuvent être utilisées comme antagonistes dans les infections cutanées. OBJECTIVE: To study the pharmacokinetics of vancomycin in plasma obtained from the medullary sinusoids of the proximal phalanx (P1) after intravenous (IV) and intraosseous (IO) regional limb perfusion (RLP). STUDY DESIGN: Experimental study. ANIMALS: Twelve horses. METHODS: IV and IO RLP were performed in 2 groups (n = 6) of horses. Vancomycin hydrochloride (300 mg in 60 mL 0.9% NaCl) was randomly infused in 1 front limb, with the contralateral limb as control (60 mL 0.9% NaCl). A hole was drilled in the lateral cortex of P1, and blood samples from the medullary sinusoids of P1 were collected before infusion, and 15, 30, 45, 65, and 90 minutes after beginning infusion. Samples were centrifuged and plasma vancomycin concentrations determined. Vancomycin concentrations were compared over time and between routes using a 2-way repeated measures ANOVA. Pharmacokinetic variables were compared with a Kolmogorov-Smirnov test. Significance was set at P<.05. RESULTS: No vancomycin-induced clinical side effects such as lameness or swelling were observed. Both techniques, IV and IO, produced high vancomycin concentrations in the sinusoidal plasma of the P1, which remained above the minimum inhibitory concentration value for methicillin-resistant Staphylococcus aureus (MRSA) during the 90 minutes study. Concentrations and pharmacokinetic variables were not significantly different comparing both routes. CONCLUSIONS: IV and IO routes produced similar antimicrobial perfusion of the medullary cavity of P1. CLINICAL RELEVANCE: Either IV or IO routes for perfusion are likely to be equally selected when planning RLP with vancomycin.  相似文献

The distribution of copper and zinc among soluble proteins in liver from normal slaughter cattle was examined after gel filtration of the proteins. Gopper- and zinc-binding proteins were mainly separated into three fractions. Varying amounts of zinc were eluted in a fourth fraction of molecular weight less than 2,000. A clear relationship was noted between the amount of copper bound to the low molecular weight fraction (m.w. ~ 10,000) and the total liver zinc concentration. The high molecular weight protein fraction (m.w. > 65,000) dominated in liver with zinc concentrations below 40 µg/g wet weight and total copper concentrations from 16 to 240 µg/g, while in liver with zinc concentrations above 40 µg/g and copper concentrations ranging from 20 to 107 µg/g, the low molecular weight metallothionein-like fraction dominated.  相似文献

A small plasmid of 2.5 kB mediating constitutive resistance to macrolide-lincosamide-(ML)antibiotics could be detected in a “canine” Staphylococcus epidermidis-culture. This plasmid, designated as pSES 1, was identified by interspecies protoplast transformation into Staphylococcus aureus RN 4220. A detailed restriction map of pSES 1 could be constructed using the restriction endonucleases Acc I, Bcl I, Cfo I, Cla I, Hind III, Hinf I, Mbo I, Sst I and Taq I. This map allowed structural comparisons of pSES 1 with plasmids from “human” Staphylococcus- and Bacillus-species, also mediating macrolide-lincosamide resistance (MLR). On the basis of its restriction map, pSES 1 proved to be similar to the plasmids pNE 131 from “human” S. epidermidis, pE 194 from “human” S. aureus and pIM 13 from B. subtilis.  相似文献

The accumulation and distribution of cadmium (Cd) between cellular subfractions were studied in Cd-resistant Staphylococcus 3719 + and its plasmid-negative Cd-sensitive variant 3719–, grown in media containing 0.01, 0.1 or 50 mg of Cd/litre. In the media containing 0.01 and 0.1 mg of Cd/1 the uptake of Cd by the Gd-resistant S. aureus was significantly less than that by Cd-sensitive S. aureus, both in toto and in terms of the cell wall fractions. Only variant 3719 + grew in the medium containing 50 mg of Cd/1. The finding of a lag phase in the growth of both variants is interpreted to suggest that the resistance is of inducible character, even in the sensitive S. aureus. The mechanism of resistance to Cd could consist of both barrier and clearance functions.  相似文献

On the addition of small concentrations of deoxyribonuclease, produced by Staphylococcus aureus, to Toluidine Blue DNA agar, a medium is produced on which antibodies against S. aureus deoxyribonuclease may be detected. When samples of milk, or blood serum, containing antibodies against S. aureus are applied into wells in the agar, the deoxyribonuclease activity is inhibited by the antibodies diffusing into the agar. As a result of this inhibition, blue zones are produced around the wells in the otherwise bluish-red agar. The diameters of the zones correspond to the concentrations of antibodies, and the method may consequently be used for qualitative and quantitative examinations of antibodies against S. aureus deoxyribonuclease in milk and serum. The procedure and certain limitations of the method are described.  相似文献

The effects of pretreatment of Staphylococcus aureus with subMIC or 10 × MIC amoxycillin, cloxacillin and nafcillin on subsequent killing by bovine neutrophils or sensitivity to lysostaphin were examined in vitro, using penicillinase positive and negative strains. All penicillins increased the susceptibility of S. aureus to bovine neutrophils; with amoxycillin this was most marked with the penicillinase negative strain. Cloxacillin pretreatment was significantly more effective than nafcillin or amoxycillin in some tests and equally effective in others. The sensitivity of S. aureus to lysostaphin was increased following cloxacillin or nafcillin exposure. In a mouse model of mastitis, more cloxacillin and nafcillin-treated S. aureus (at 1/4 MIC) were killed than untreated bacteria 1 hr after infection but by 4 hr no differences were found. No differences were seen after intramammary infection with 10 × MIC penicillin-treated S. aureus. In glands with established chronic mastitis both cloxacillin and nafcillin at high doses failed to kill staphylococci. These findings indicate that prior exposure to penicillins increases staphylococcal susceptibility to killing by neutrophils but following phagocytosis, intracellular bacteria are protected from the lethal action of penicillins.  相似文献

To determine the effect of bacteria on the development of the ripening flavor of cured meat, pork loins were cured with pickles containing 8 or 16% sodium chloride at 0 and 8°C. The bacterial flora, total free amino acid content and total free fatty acid content in the cured loins, sensory properties of cooked pork loins and the relationships between the proteolytic and lipolytic activities of isolates and curing conditions were investigated. Desirable bacteria, including salt‐tolerant bacteria and lactic acid bacteria, were predominant under the condition of 8% sodium chloride in the pickles and a curing temperature of 8°C. The total free amino acid content and total free fatty acid content at a curing temperature of 8°C were higher than those at 0°C. Cooked pork loins cured in pickles containing 8% sodium chloride at a temperature of 8°C for more than 7 days were preferred in terms of color, flavor and taste. Before the curing procedure, gram‐negative bacteria (Vibrio, Acinetobacter, Pseudomonas and Enterobacteriaceae) were predominant in the pork loins. During the curing period, the numbers of viable gram‐positive bacteria (Micrococcus, Staphylococcus and Pediococcus) increased and the numbers of viable gram‐negative bacteria decreased. Strains of Micrococcus and Staphylococcus with proteolytic and/or lipolytic activities in cured meat also increased during the curing period and were more predominant in the pork loins cured in pickles containing 8% sodium chloride and at a curing temperature of 8°C than in pork loins cured in pickles containing 16% sodium chloride and at a curing temperature of 0°C. The actions of these bacteria were thought to be important factors affecting the flavor of the cured pork. Micrococcus and Staphylococcus with proteolytic activity might contribute to the development of the ripening flavor of ham. The results of the present study together with the results of further investigations on the relationships of the enzyme activities of Micrococcus and Staphylococcus in cured meat with a preferable flavor would be useful for establishing a novel effective method for using bacteria to produce ham of a high quality.  相似文献
