青藏高原海东鸡CAPN3基因第1外显子多态性研究

[目的]对青藏高原海东鸡CAPN3基因第1外显子的多态性进行研究。[方法]采用PCR-SSCP技术检测海东鸡CAPN3基因第1外显子的多态性。[结果]海东鸡CAPN3基因第1外显子存在c.223GA的错义突变,对应2种等位基因G和A,等位基因G为优势等位基因;该群体CAPN3基因检测区域0.25PIC≤0.5为中度多态,且偏离Hardy-Weinberg平衡状态(P0.01)。[结论]该研究为海东鸡功能基因的分子遗传研究提供了基础资料。     

Polymorphisms of the prolactin gene and their association with egg production traits in two Chinese domestic ducks

1. Prolactin (PRL) as a polypeptide hormone which plays a crucial role in egg production traits.

2. Polymorphisms of the PRL gene were analysed with DNA sequencing and polymerase chain reaction-single-strand conformation polymorphism methods in two Chinese domestic laying duck breeds (Jinding, n = 400, Youxian, n = 400, respectively).

3. The results showed that one polymorphism was detected (A-412G) in intron 1 of the PRL gene, with three genotypes: AA, AG and GG. Association analysis showed that the ducks with the GG genotype had significantly greater egg production and egg weight than those with AG and AA genotype (p < 0.05). Hence, the 412A > G polymorphism of the PRL gene in intron 1 is a potentially valuable genetic marker for laying duck breeding programmes.     

绵羊BMPR-IB基因第7外显子多态性与产羔性状的相关性研究

【目的】探索绵羊BMPR-IB基因第7外显子多态性与产羔数的相关性,寻找控制绵羊繁殖力的分子标记位点。【方法】以相同饲养管理条件下同龄已有产羔记录的蒙古羊(单胎母羊和双胎母羊)、无角陶赛特羊(单胎母羊和双胎母羊)、小尾寒羊(多胎母羊)为试验材料,采用PCR-SSCP结合DNA直接测序的方法,对BMPR-IB基因第7外显子进行多态性检测与分析。【结果】试验羊只存在AA型和AB型2种基因型;AB型个体在BMPR-IB基因第864位点发生C→T突变,出现T/C的杂合,所有试验样本的优势基因型均为AA型,优势等位基因均为A基因。χ2适合性检验表明,无角陶赛特羊、蒙古羊和小尾寒羊均处于Hardy-weinberg平衡状态,BMPR-IB基因第7外显子不同基因型在不同产羔类型绵羊中的分布差异显著,最小二乘法分析表明,绵羊BMPR-IB第7外显子多态性与产羔数有一定的相关性。【结论】BMPR-IB基因第7外显子第864位点的突变在不同繁殖力母羊群体中分布不同,表明绵羊BMPR-IB基因第7外显子多态性与绵羊产羔数相关,可以作为控制绵羊繁殖力的潜在遗传标记位点进行下一步研究。     

藏绵羊DRB3基因第2外显子多态性分析

为了研究藏绵羊(Ovis aries)DRB3基因的多态性,确定其等位基因数、等位基因间的核苷酸多态位点、变异类型,以及分析各等位基因间的遗传关系和进化意义,研究采用PCR-SSCP方法检测了500只藏绵羊DRB3基因第2外显子的多态性,并克隆、测序群体内变异产生的各等位基因.结果表明藏绵羊DRB3基因第2外显子表现了37个等位基因,37个单倍型序列分析发现82个核苷酸多态位点;37个单倍型序列与GenBank下载序列对比分析,结果表明35个DRB3的等位基因是本研究旨次发现;37个DRB3基因外显子2的单倍型序列NJ系统发育树呈2支分化趋势.研究认为藏绵羊DRB3基因第2外显子具有丰富的遗传多态性;藏绵羊DRB3基因最初是由2个等位基因突变分化成两大类等位基因的.     

固始-安卡鸡F_2资源群△-9脂肪酸脱氢酶基因第4、5、6外显子SNP及其与脂肪酸相关性研究

利用PCR-SSCP(single strand conformation polymorphism)方法,对固始-安卡鸡F2代资源群△-9脂肪酸脱氢酶基因第4、5、6对外显子多态性进行分析,发现存在一个单核苷酸多态(SNP,single nucleotide polymorphisms)位点。在△-9脂肪酸脱氢酶基因第4外显子编码区(89nt)发生了碱基的转换突变(G→A)。在整个F2代资源群中进行了该多态位点与脂肪酸性状的相关分析,结果表明:在8个脂肪酸性状中,AA型个体十六碳二烯酸(C16∶2)含量显著高于GG型和GA型个体(P<0.05);家系1中AA型的C16∶0、C16∶2、C18∶0含量均显著高于GG型和GA型(P<0.05);家系3中AA型的C14∶0含量显著高于GG型和GA型(P<0.05)。     

The Association Analysis Between the SNPs of Leptin Gene and Litter Size Trait in Pigs

The research was aimed to analyze the association of Leptingene and breed traits on pigs. Three pairs of primers were designed which based on DNA sequence of pig Leptin gene (GenBank accession No.U66254, AF026976),the single nucleotide polymorphisms (SNPs) of Leptin gene were detected by PCR-SSCP in Sujiang and Jiangquhai pigs. The results showed that three genotypes of AA,AB and BB were identified, and the mutation (T3469C) in CDS of Leptin gene was found,but amino acid change weren't happened. The Chi-square test results showed that the Leptin genotype distribution of Sujiang and Jiangquhai pigs was in Hardy-Weinberg equilibrium (P>0.05). The polymorphism information content (PIC) were moderate polymorphism in Sujiang pigs,and low polymorphism in Jiangquhai pigs. The statistic analysis results showed that there were no significant association between the SNP of Leptin genotype and litter size trait(P>0.05). But the average litter size of T3469C site AB genotype was higher than AA and BB genotypes, it was concluded that the T3469C polymorphism of Leptingene might be the genetic marker which potentially affected pig breed traits.
