不同有机物料对连作大豆根际土壤线虫的影响   总被引:1,自引:0,他引:1  
为探讨大豆连作体系下农业废弃物为主的不同有机物料对根际土壤线虫的影响,选用6种不同有机物料,设正茬和连作两种处理进行对比分析根际土壤线虫群落差异。结果表明:土壤线虫中重要的优势属和关键属种cp值均为3,说明其繁殖和抗干扰能力比较强。连作7年后根际土壤线虫营养类群中杂食-捕食性线虫相对丰度下降明显,植物寄生线虫相对丰度显著提高,食细菌线虫、食真菌线虫丰度略有提高;相较单施化肥(CK)处理,添加有机物料明显抑制连作大豆根际土壤中植物寄生线虫的大量增长,不同施肥处理效果分别为:猪粪秸秆菌渣牧草鸡粪黄腐酸生物有机肥CK,与正茬相比,各连作处理依次增长6.48%、15.34%、15.98%、18.51%、24.35%、26.33%和27.4%。土壤线虫的生态指数较CK相比也有明显改善。土壤线虫对肥料的响应取决于作物及肥料的质量和养分含量;土壤线虫优势营养类群、优势属种和关键属种可以有效地指示农业土壤健康状况和大豆田间线虫病害的发生程度。  相似文献

The commercially available parasitic nematode Phasmarhabditis hermaphrodita is an effective biocontrol agent for slugs and particularly Deroceras reticulatum, a widespread pest species. Use of the nematode is currently limited by cost and it may be that by developing a fuller understanding of the ecology and behaviour of this nematode, more cost effective application strategies can be developed. We investigated the ability of two strains of P. hermaphrodita (one newly isolated and one that had been maintained in vitro for >15 years) to move through mineral soils and organic media. Active dispersal of both strains was found to be greatest in organic media (bark chips and leaf litter, and to a lesser extent peat) and the nematode was capable of growth and reproduction in leaf litter. Conversely, active dispersal was poor in mineral soils. Nematodes moved further in a clay loam compared with a sandy loam, and moved more at a bulk density of 1.0 vs. 1.2 Mg m−3. However, P. hermaphrodita was capable of moving greater distances in mineral soils by using the earthworm Lumbricus terrestris as a phoretic host. Our data suggest that P. hermaphrodita is a facultative parasite that is adapted to living in leaf litter and organic material where slugs frequently rest. The implications of these findings for using the nematode as a biological control agent for slugs are discussed.  相似文献

为了研究小麦孢囊线虫病经济阈值,制定防治对策。采用室内盆栽试验和田间试验的方法,测定了小麦孢囊线虫(cereal cyst nematode,CCN)在不同群体密度下对小麦的危害损失,建立了CCN群体密度与产量损失的回归方程。结果表明:小麦孢囊线虫为害小麦根部,影响小麦根系发育、株高、分蘖、穗粒数、千粒重等农艺性状;每克土壤中CCN卵量≤(15±2.5)时,各项指标下降不显著,产量损失<10%;每克土壤中CCN卵量≥(30±5)时,各项指标下降显著,产量损失高达15.52%~55.01%。因此,CCN群体密度大于30卵/g土的地区,应加强防治。  相似文献

[目的]摸清宁波地区盆景线虫种类,以有针对性地进行线虫防控。[方法]2005~2008年,对宁波市3家主要盆景出口企业及周边的29种主要出口盆景及盆栽植物的根际土壤、栽培介质和根组织进行线虫种类调查,共采集样品142份。[结果]分离并鉴定线虫12属,发现了剑线虫属(Xiphinema sp.)、长针属(Longidorus sp.)、短体属(Pratylenehus sp.)、毛刺属(Trichodorus sp.)和拟毛刺属(Paratri-chodorus sp.)等检疫性线虫,未发现根结线虫属。提出了线虫防控措施。[结论]宁波盆景线虫种类较多,盆景出口前需要综合治理,以确保顺利出口。  相似文献

The false root-knot nematode, Nacobbus aberrans, causes severe damage to field and greenhouse pepper crops in several localities of the American continent. No commercial peppers (Capsicum annuum) resistant to this nematode are available up to the present. Host suitability of 6 experimental and 3 commercial peppers (some of them carrying Me1 and Me7 resistance genes to Meloidogyne spp.) to two N. aberrans populations were evaluated under greenhouse experiments. None of the peppers was found to be resistant to the nematode. The evaluated parameters exhibited significant differences among some peppers tested within a single population and between populations for a single plant material. Some peppers carrying resistance showed higher nematode reproduction than some lines that did not possess resistance genes. These results confirmed that the genes conferring resistance to Meloidogyne spp. do not provide protection against this species of root-galling nematode. Host suitability of pepper lines carrying Me1 or Me7 resistance genes against N. aberrans is evaluated for the first time. Search for resistant genes against this nematode in wild peppers growing in areas where this nematode is indigenous should be promoted.  相似文献

Pine wilt disease, caused by the pine wood nematode (PWN, Bursaphelenchus xylophilus), is a major threat to pine forests throughout East Asia. Nonetheless, its mechanism of invasion has not yet been described in detail. To better understand the pathology of this disease, it is important to examine the distribution of PWNs within pine tissue during the course of disease development. We attempted to stain nematodes with fluorescein-conjugated wheat germ agglutinin (F-WGA) as a means to locate and track the spread of PWNs. Although PWNs proliferated on Botrytis cinerea fungus were successfully stained only on their vulvas and spicule holes, PWNs extracted from inoculated Pinus thunbergii seedlings were stained on their surface. Stainability, or the percentage of PWNs stained with F-WGA over more than half of their surface, was about 20% by 1 day after inoculation, but increased to 80% at 10 days. The stainability of PWNs extracted from a 5-cm main stem segment that included the inoculation site was less than that of PWNs extracted from other parts of the main stem farther away (i.e., those that had dispersed). These results suggest that stainability is related to dispersal activity in time. Thus, to raise the stainability of PWNs at shorter timeframes after inoculation, PWNs with more than 80% stainability were re-inoculated into pine seedlings. This resulted in more than 70% stainability from 1 to 6 days after inoculation. In F-WGA stained thin paraffin sections of pine tissue of re-inoculated seedlings, PWNs brightly fluoresced under epifluorescence and were easily detected against the dark background of pine tissue. This staining technique with F-WGA is an excellent tool for detecting PWNs in pine tissue.  相似文献

 本文对5个属14个种共33株捕食线虫真菌采用聚丙烯酰胺凝胶电泳技术进行了酯酶同工酶谱和可溶性蛋白质谱分析。结果表明,酯酶同工酶谱带少,但各属的特征谱带是明显的,在同属不同种之间,多数种都有自已的特征谱带与其它种相区别,同种不同菌株间没有明显差异;蛋白质谱带较多,在每个属中均有1~2条特征谱带,属间的谱带差异明显,同属不同种和同种不同菌株间蛋白质谱带都表现出多型性。  相似文献

施用有机物料能够显著影响土壤肥力和生物群落结构,然而有机物料在土壤中分布不均会加剧土壤空间异质性,进而对土壤生物群落的结构和功能产生进一步的影响。选用水稻秸秆和白三叶草两类有机物料,按比例混合后装于网袋,置于预先装好土的培养容器中恒温培养,在培养的第14、35和70天分别对其中的不同微域(0~1 cm和1~5 cm)土样进行采集,分析其中的土壤原生动物和线虫群落结构的变化。结果表明,有机物料种类及培养时间对原生动物和线虫数量的影响更大,采样微域对二者的影响相对较小。水稻秸秆添加下的土壤植食性线虫、食真菌线虫和捕杂食线虫数量显著高于白三叶草添加;而白三叶草添加下的土壤变形虫、鞭毛虫和线虫总数、食细菌线虫数量显著高于水稻秸秆添加。从整个培养周期来看,鞭毛虫数量随培养时间的延长逐渐减少,而变形虫和线虫总数则随培养时间的延长逐渐增多。在同一有机物料下不同微域间土壤原生动物和线虫在培养初期会出现一定的数量差异,但这种差异随培养时间的延长逐渐消失。  相似文献

Apple replant disease (ARD) has been reported from all major fruit-growing regions of the world, and is often caused by a consortium of biological agents. The aim of this study was to investigate the etiology of ARD in South Africa in six orchard soils, using a multiphasic approach under glasshouse conditions. This approach first involved determining the ARD status of the soils by monitoring apple seedling growth responses in non-treated soil versus growth in pasteurized soil, as well as in 15% non-treated soil that was diluted into pasteurized soil. Subsequently, the potential for specific organisms to function as causal agents of ARD was investigated using (i) biocide applications, (ii) quantitative real-time PCR (qPCR) analyses of ARD ‘marker’ microbes (Pythium irregulare, Pythium sylvaticum, Pythium ultimum, Pythium vexans, Rhizoctonia solani AG-5 and the genera Cylindrocarpon and Phytophthora), (iii) nematode analyses, (iv) isolation of actinomycetes and (v) pathogenicity testing of actinomycetes individually, and when co-inoculated with P. irregulare or Cylindrocarpon macrodidymum. The analyses showed that the soils could be grouped into low, moderate and severe ARD soils, with each group containing two soils. Several lines of evidence suggested that actinomycetes are not involved in ARD in South Africa. Multiple biological agents were determined to contribute to ARD including (i) oomycetes (Phytophthora and Pythium) that are important based upon their widespread occurrence, and the fact that metalaxyl application improved seedling growth in four soils (ii) the genus Cylindrocarpon that was also widespread, and for which a synergistic interaction with P. irregulare was demonstrated and (iii) occasionally parasitic nematodes, mainly Pratylenchus penetrans, Pratylenchus scribneri and Pratylenchus delattrei, since fenamiphos application improved seedling growth in two orchards. qPCR analyses of the ARD marker microbes showed that R. solani AG-5 is absent from South African orchards, and that P. ultimum is widespread, even though the latter species could not be detected in previous isolation studies. The other marker microbes were also widespread, with the exception of P. sylvaticum. qPCR quantification of the marker microbes could not be correlated with the severity of ARD in any manner. qPCR analyses did, however, show that possible root pruning pathogens such as P. irregulare, P. sylvaticum and P. ultimum had much lower DNA concentrations in seedling roots than P. vexans and the genera Cylindrocarpon and Phytophthora.  相似文献
