The microbial activity and bacterial community structure were investigated in two types of peat soil in a temperate marsh. The first, a drained grassland fen soil, has a neutral pH with partially degraded peat in the upper oxic soil horizons (16% soil organic carbon). The second, a bog soil, was sampled in a swampy forest and has a very high soil organic carbon content (45%), a low pH (4.5), and has occasional anoxic conditions in the upper soil horizons due to the high water table level. The microbial activity in the two soils was measured as the basal and substrate-induced respiration (SIR). Unexpectedly, the SIR (μl CO2 g−1 dry soil) was higher in the bog than in the fen soil, but lower when CO2 production was expressed per volume of soil. This may be explained by the notable difference in the bulk densities of the two soils. The bacterial communities were assessed by terminal restriction fragment length polymorphism (T-RFLP) profiling of 16S rRNA genes and indicated differences between the two soils. The differences were determined by the soil characteristics rather than the season in which the soil was sampled. The 16S rRNA gene libraries, constructed from the two soils, revealed high proportions of sequences assigned to the Acidobacteria phylum. Each library contained a distinct set of phylogenetic subgroups of this important group of bacteria.  相似文献

Interactions between earthworms and microorganisms are essential for the functioning of soil ecosystems as they affect organic matter degradation and nutrient cycling. This is also true for the alpine region, where socio-economic changes lead to the increasing abandonment of pastures, which in turn, causes a considerable shift in the diet of saprotrophic invertebrates and thus impacts food web and decomposition processes. To enhance our understanding of how this diet shift influences earthworms and associated microorganisms, we studied the gut content and cast microbiota of Lumbricus rubellus (Lumbricidae, Oligochaeta), a key macrodecomposer on alpine pastureland in the Central Alps. A feeding experiment with L. rubellus and three different food sources that represent the vegetation shift from an alpine pasture to an abandoned site was set up. Earthworms were collected in the field, transferred to a climate chamber and fed with cow manure, dwarf shrub or grass litter for six weeks. PCR-DGGE (Polymerase chain reaction-denaturing gradient gel electrophoresis) analysis of the DNA extracted from the substrates, the earthworms' gut contents and casts revealed that the gut and cast microbiota was strongly influenced by the food source ingested. Cloning of bacterial 16S rRNA gene fragments demonstrated that the intestinal content was dominated by Proteobacteria, especially from the Gamma-subclass, followed by members of the phyla Bacteroidetes, Actinobacteria and Firmicutes. In contrast, Actinobacteria were detected abundantly in all samples types when a cultivation approach was used. In conclusion, the gut microbiota of L. rubellus was shown to be substantially affected by the food source ingested, suggesting that this essential macrodecomposer is exposed to the diet shift resulting from a land-use change in the alpine area.  相似文献

Soil physicochemical properties and microbial communities are highly heterogeneous and vary widely over spatial scales, necessitating careful consideration of sampling strategies to provide representative and reproducible soil samples across field sites. To achieve this, the study aimed to establish appropriate sampling methodology and to determine links between the variability of parameters, utilising two sampling strategies. The first (design 1) involved extracting 25 cores from random locations throughout the field and pooling them into five sets of five cores. The second (design 2) involved a further 25 cores within five 1 m2 sub-plots. Sub-samples from each sub-plot were pooled in order to determine between and within sub-plot variability. All samples were analysed independently and as pooled sub-samples. Results indicate that pooling spatially separated samples significantly reduced the variability in pH, compared to individual samples. Pooling samples from a small area resulted in lower within sub-plot variability than between sub-plots for pH and bacterial community composition assessed by terminal-restriction fragment length polymorphism analysis. Following multivariate statistical analysis, a large amount of variation in community composition was explained by soil pH, which is remarkable given the relatively small size of the sampling area and minor differences in pH. Moisture content was also important in determining bacterial communities in the random design (design 1). In the 1 m2 sub-plot design (design 2), the spatial location of the plots explained a large degree of the variation in bacterial community composition between plots, which was due to spatial autocorrelation of pH and possible additional environmental parameters. Dune systems are characterized by strong gradients of physical stress, with blowing sand and salt spray decreasing with distance from the ocean, and soil nutrients increasing. In this study we ask how soil microbial community composition and biomass change along transects away from the ocean, and whether these changes are regulated by abiotic stress or by resource availability. We collected bulk soil from under three plant species representative of the dune front, back, and flat: Ammophila breviligulata, Rosa rugosa, and Myrica pensylvanica. The biomass and composition of microbial communities were examined using phospholipid fatty acid (PLFA) analysis under patches of dominant vegetation, and in paired bare plots. We found that microbial biomass was strongly correlated with soil C, and thus the presence of vegetation. Community composition, on the other hand, varied with abiotic stresses, especially soil salinity. These variables in turn depended on distance from the shore, and were ameliorated in some cases by vegetation. These findings demonstrate that biomass and community composition are influenced by different environmental variables. Thus, relationships between biomass and composition are unlikely to be readily predicted on the basis of a single resource.  相似文献

The biodegradability of dissolved organic carbon (DOC) in different fractions from the forest floor was studied. Soil leachate (SL, the soil solution in macropores which is freely drained from forest floor after rainfall), the soil matrix solution (SMS, the soil solution in meso-/micropores of the soil matrix), and soil water extracts (SWE) from two different beech forest floors were compared. Zero-tension and tension lysimeters were used to collect SL and SMS, respectively. Loss of DOC (during 21 days) and respiration of CO2-C (during 7 days) were used as conventional measures of the availability of DOC. Bacterial production, measured using the leucine incorporation technique, and bacterial growth efficiency were also estimated. All methods were used to study differences in biodegradability between plots with and without ground flora (Deschampsia flexuosa or Anemone nemorosa) and different type of forest floor (with an organic (O) horizon or a mull (A) horizon). There were no differences in bioavailability of DOC from soil solutions extracted from plots with and without ground flora. The bioavailability of DOC in the different collected soil solutions varied, however. DOC in SWE was the most available, with a mean of 39% of DOC-loss in 21 days, and 18% of DOC being respired in 7 days. DOC in soil matrix solution was the least available of the soil solutions (7% respired), significantly less than DOC in soil leachate (11% respired). The methods measuring biodegradation of DOC, DOC-loss and CO2-C respiration gave similar results and were comparable to bacterial production and bacterial growth efficiency, with the exception of SWE from the O-horizon at the D. flexuosa site, which had low bacterial production and bacterial growth efficiency, indicating a limitation of the bacterial growth. This study is one of the first to use bacterial production and bacterial growth efficiency for measuring bioavailability in terrestrial environments, giving an extra dimension for the process of biodegradation of DOC.  相似文献

选取凡纳缤对虾(Litopenaeus vannamei)的4种组织(鳃、血淋巴、肝胰腺、肌肉),着重对膜蛋白的提取方法及其多肽组成进行分析。在对虾组织膜蛋白提取过程中,使用了5种蛋白酶抑制剂(苯甲基磺酰氟PMSF、胃酶抑制剂、亮抑酶肽、胰凝乳蛋白酶抑制剂、胰蛋白酶抑制剂),以减少组织自身的酶将蛋白降解的可能。电镜观察,可见纯度较高的、细胞膜结构卷曲形成的小泡样、封闭的结构。使用Gel-Pro软件对提取产物的SDSPAGE图谱分析,确定了这几种组织的特征多肽分子量分别为75.0kD、70.5kD、26.7kD和71.2kD。  相似文献

通过1988~1995年对安徽省竹类害虫的调查,查明现阶段竹类害虫223种,其中轻微发生的202种,中等发生的12种,严重发生的9种.灾害性害虫竹织叶野螟、竹笋禾夜蛾、江苏泉蝇、一字竹象在毛竹上的发生率分别为41.34%、2.52%、3.50%、0.51%,平均虫口分别为2.58头*株-1、0.05头*株-1、0.47头*株-1、0.01头*株-1,在刚杂竹上的发生率分别为43.60%、29.25%、34.32%、4.03%,平均虫口分别为2.04头*株-1、0.84头*株-1、9.86头*株-1、0.30头*株-1.测算竹类害虫的危害损失,提出综合治理对策.  相似文献

Through genetic selection and improvement of environment, litter size of sows increases. Increased energy requirement during lactation, increases the risk of excessive mobilization from body stores, with detrimental effects on reproductive performance. Feed intake capacity tends to decrease due to selection towards leaner pigs with a lower feed conversion ratio. However, to facilitate sows to wean large litters extra feed intake, or even better, a higher feed efficiency during lactation would be favourable. The objective of the present study was to describe the dynamics of body composition of sows and piglets during lactation, and to relate these traits to a newly introduced trait called “lactation efficiency”. Energy metabolism of lactating sows was described, based on on-farm observations of weight and backfat of sows before parturition and at weaning, weight of piglets at birth and at weaning and feed intake of sows during lactation. “Lactation efficiency” was defined as energy efficiency of sows, and calculated for individual sows at two different farms. The average lactation efficiency was 68% and 65% for both farms; meaning that 68 and 65% of the metabolisable energy through feed intake or mobilization from body stores, above maintenance of the sow (input), was used for piglet growth and piglet maintenance (output). The association between lactation efficiency and other reproductive traits was studied by estimating the correlations within farms. Sows with a higher lactation efficiency showed lower feed intake (r = − 0.27 and r = − 0.35 for both farms respectively) and smaller fat losses (r = − 0.34 and r = − 0.29, respectively). The energy output of efficient sows was slightly higher (r = 0.23 and r = 0.30). The more efficient sows were the better mothers, as mortality of their piglets was lower (r = − 0.12 and r = − 0.16), piglet growth rate was higher (r = 0.16 and r = 0.23), and at weaning their litters were less variable (r = − 0.08; only available at one farm). Results were remarkably similar for the two farms, despite different feeding strategies. Extra input, by means of feed intake or mobilization from body stores generated extra output by means of litter weight at weaning. This experiment demonstrated that an accurate recording of energy metabolism and relevant reproduction traits with little intervention is possible on commercial farms.  相似文献

Most studies on lactation curves only consider milk yield and describe a standard lactation curve of dairy cows, showing a peak or maximum daily yield occurring between 4 and 8 weeks after calving, followed by a daily decrease in milk yield until the cow is dried off. Wood's model is a widely used lactation curve function. Wood's model was fitted to test-day records of 95,405 lactations of parities lower than 5. Milk traits were milk yield (MY), fat percentage (F%), protein percentage (P%), fat yield (FY) and protein yield (PY), and the lactation curve was individually considered as a cluster of five linked curves. Milk trait and parity influence the goodness of fit of Wood's model. In 19.3% of the lactations, the shape of the MY, FY and PY curves follows the standard lactation curve while F% and P% have the reversed standard shape. The initial phase of lactation with the FY and PY curves contributes to the high variability of shapes.  相似文献
