分泌抗鸡Ig单抗杂交瘤细胞系的建立与单抗生物学特性的鉴定

从传染性法氏囊病病毒（ＩＢＤＶ）单克隆抗体（ＭｃＡｂ）杂交瘤细胞系１Ｄ１０、１Ｇ１中得到了３株能稳定分泌抗鸡免疫球蛋白（Ｉｇ）ＭｃＡｂ的杂交瘤细胞系１Ｂ７、１Ｄ７、３Ｇ６。其抗体类和亚类均属ＩｇＧ２ｂ，腹水的ＥＬＩＳＡ效价≥１∶２５６００，琼扩效价为１∶８～１∶１６。３株ＭｃＡｂ能与鸡血清中的Ｉｇ出现沉淀反应，琼扩在２４ｈ以内出现清晰的沉淀线，而不能和鸭、鸽、鹌鹑等禽类及异种动物血清出现沉淀线。纯化的鸡ＩｇＧ、ＩｇＭ经ＳＤＳ－ＰＡＧＥ后，分别用纯化并经辣根过氧化物酶（ＨＰＲ）标记的１Ｂ７、１Ｄ７、３Ｇ６单抗酶结合物进行免疫印迹试验证明，３株单抗识别的均为ＩｇＧ、ＩｇＭ分子的轻链     

Association of polymorphism of Fc receptor γ chain gene at position-29 in promoter with the susceptibility to systemic lupus erythematosus in southern Chinese

AIM:To detect the association between the polymorphism of Fc receptor γ chain gene at position-29 in promoter and systemic lupus erythematosus(SLE).METHODS:The genotypes at position -29 in promoter of Fc receptor γ chain gene were determined by polymerase chain reaction-restriction fragment length polymorphism (PCR-RFLP) method in 180 patients with SLE and 140 ethnically matched controls in southern China.RESULTS:The frequencies of TT genotype(33.3%) and T allele (54.4%) at position -29 in patients with SLE were significantly higher than those in controls (17.2% and 42.9%, respectively), whereas, the frequencies of GG genotype (24.4%) and G allele (45.6%) in patients with SLE were remarkably lower than those in controls (31.4% and 57.1%, respectively) (P＜0.05). The TT genotype and T allele at position -29 were not associated with lupus nephritis in SLE patients (P＞0.05).CONCLUSION:Our results indicate that the T allele at position -29 in promoter of Fc receptor gene probably contributes to the susceptibility to SLE, but does not play a role in the occurrence of lupus nephritis.     

Branched‐chain amino acid ratios in low‐protein diets regulate the free amino acid profile and the expression of hepatic fatty acid metabolism‐related genes in growing pigs

Liver metabolism is affected by nutrients. The aim of this study was to explore the effects of low‐protein diets (17% crude protein, CP) supplemented with branched‐chain amino acids (BCAAs), including leucine (Leu), isoleucine (Ile) and valine (Val), on hepatic amino acid profile and lipid metabolism in growing pigs. The ratio of Leu : Ile : Val in all groups was 1 : 0.51 : 0.63 (20% crude protein, CP), 1 : 1 : 1 (17% CP), 1 : 0.75 : 0.75 (17% CP), 1 : 0.51 : 0.63 (17% CP) and 1 : 0.25 : 0.25 (17% CP) respectively. Results revealed that compared to the positive control group (1 : 0.51 : 0.63, 20% CP), the low‐protein diets significantly augmented the concentrations of most essential amino acids and non‐essential amino acids (p < .05), with the greatest values observed in the 1 : 0.25 : 0.25 group. Moreover, relative to the control, the low‐protein diets with the Leu : Ile : Val ratio ranging from 1 : 0.75 : 0.75 to 1 : 0.25 : 0.25 markedly downregulated the mRNA abundance of acetyl‐CoA carboxylase (ACC), lipoprotein lipase (LPL) and fatty acid‐binding protein 4 (FABP‐4) (p < .05), and upregulated the mRNA expression of hormone‐sensitive lipase (HSL), peroxisome proliferator‐activated receptor‐g coactivator‐1α (PGC‐1α), uncoupling protein 3 (UCP3) and liver carnitine palmitoyltransferase 1 (L‐CPT‐1) (p < .05). Therefore, our data suggest that protein‐restricted diets supplemented with optimal BCAA ratio, that is, 1 : 0.75 : 0.75–1 : 0.25 : 0.25, induce a shift from fatty acid synthesis to fatty acid oxidation in the liver of growing pigs. These effects may be associated with increased mitochondrial biogenesis.

Mitochondrial DNA deletion of the brain tissue of aged rats with learning and memory deficit

AIM and METHODS: The ratio of mitochondrial DNA (mtDNA) deletion was measured to find the relationship between mtDNA deletion and aged learning and memory deficit. The aged rats were divided into two groups, aged learning and memory deficit group and aged learning and memory normal group. The ratio of mtDNA deletion was measured by dilution polymerase chain reaction. RESULTS: There are deleted mtDNA (about 4834 bp) in the cerebral cortex, hippocampus and cerebellum of both young and aged rats. The ratios of deleted mtDNA were similar in the cerebral cortex,hippocampus and cerebellum of young rats (about 0.00018%). The ratio mtDNA of aged learning and memory normal rats had increased by five-fold in the cerebral cortex and hippocampus, or one-fold in the cerebellum over young rats. The ratio of aged learning and memory dificit rats had increased by one-fold in the cerebral cortex or 0.8-fold in the hippocampus or two-fold in the cerebellum over aged learning and memory normal rats.CONCLUSIONS: There was really the increase of mtDNA in aging rat brain. And this increase was double in amount in aged learning and memory deficit rats compared to the normal learning and memory aged rats. It is suggested that the mtDNA deletions in the brain regions associated with learning and memory may be contributed to the cellular and molecular mechanism of learning and memory deicit with aged rats.     

外加盐对凉粉草胶溶液的耐盐性和分子链刚性度的影响

在有盐环境下贮藏和加工凉粉草胶必须了解其多糖分子对盐的敏感性,该文绘制了凉粉草胶多糖在不同离子强度NaCl、KCl、MgCl₂和CaCl₂溶液中的Huggins'图,分别计算其分子链在这4种离子溶液中的耐盐度S、链刚性度B等值,结果发现：凉粉草胶多糖在4种离子溶液中,其耐盐性由强到弱的顺序为:K⁺>Na⁺>Mg⁺⁺>Ca⁺⁺,其分子链的构象在4种离子溶液中由柔到刚的顺序为:K⁺>Na⁺>Mg⁺⁺>Ca⁺⁺,这说明凉粉草胶多糖适合在KCl溶液中贮藏和加工。与其他多糖分子链在NaCl溶液中的B值比较后发现,凉粉草胶多糖的分子链构象较海藻酸钠要刚,较黄原胶要柔,这说明凉粉草胶比黄原胶更适合在NaCl溶液中贮藏和加工。     

农产品冷链物流高质量发展水平评价及障碍度分析

推动冷链物流高质量发展,对减少农产品产后损失、扩大消费品优质供给、保障食品安全有重要作用。基于我国2012-2021年统计数据,构建了包括经济基础、物流发展、冷链运营、开放创新、绿色协调5个一级指标及21个二级指标的农产品冷链物流高质量发展评价指标体系,利用熵值法确定各指标的权重,综合评价指数确定得分水平,借助障碍度模型分析发展障碍因子。结果显示：我国农产品冷链物流高质量发展水平总体呈上升趋势且增速较快,到2021年水平值达到0.85,是初始状态的近13倍,其中开放创新水平的贡献度最高;阻碍高质量发展的障碍因素在2017年之前为开放创新水平,之后则是绿色协调水平。为进一步提高我国农产品冷链物流高质量发展水平,需要充分利用国内外两个市场、促进产业协同、发展绿色低碳物流。     

模糊加权马尔可夫链模型及其应用

在传统的用马尔可夫链模型预测的方法基础上,建立了模糊加权马尔可夫链模型。以某地区梅雨期长度的预测作为实例,介绍了使用这种模型的方法与步骤,提出了具有模糊状态的马尔可夫链的转移概率计算公式,并采取以规范化的各阶自相关系数为权重,用加权的马尔可夫模型预测的结果与实际梅雨期长度相吻合。     

北京郊区猪戊型肝炎流行初步调查

用间接ELISA试剂盒对北京市郊区所采集的174份猪血清样品进行了抗体检测,结果显示,其抗体总阳性率达62.1%。针对戊型肝炎病毒(HEV)ORF2/ORF3重叠区设计了简并引物,采用巢式RT-PCR对随即采取的少量猪粪便样品进行了抗原检测。结果表明,部分粪便样品中存在HEV。
