【目的】寻求梯形明渠临界水深的直接计算公式。【方法】针对梯形明渠临界水深传统计算方法计算过程复杂、误差大、适用范围小的缺点,以迭代法为基础,引入无量纲参数单位水面宽度,利用数值计算方法对梯形明渠临界水深的基本计算公式进行恒等变形,得到快速收敛的迭代公式,并对合适的迭代初值进行选取。【结果】提出了新的梯形明渠临界水深的直接计算公式,对其进行的误差分析结果表明,该公式计算临界水深的最大相对误差仅为0.010%。【结论】与现有公式相比,新建立的梯形明渠临界水深直接计算公式计算精度高、适用范围广,可供工程实际参考应用。  相似文献

电压门控钠离子通道对于脊椎动物脑神经起始、传播动作电位具有重要作用。为了解斑马鱼电压门控钠离子通道基因scn1Laa在脑神经中的作用,通过CRISPR/Cas9基因编辑技术,首次构建了可稳定遗传的生长没有受明显影响的scn1Laa缺陷型(scn1Laa-/-)斑马鱼家系。相比野生型,5 dpf(days post-fertilization,受精后5天)scn1Laa缺陷型斑马鱼兴奋抑制性神经元(氨基丁酸类神经元)表达相对增加,兴奋类神经元(谷氨酸能类神经元)和成熟神经元显著减少,脑部细胞增殖也显著减少。受精后5天和90 天的 scn1Laa缺陷型斑马鱼的运动较同时期野生型斑马鱼更为活跃,受精后90天的 scn1Laa缺陷型斑马鱼的运动具有明显的爆发性。以上结果表明,scn1laa缺失导致兴奋类神经元(谷氨酸能类神经元)以及神经细胞增殖减少,影响脑周围神经放电,导致运动神经调节障碍,出现运动行为异常活跃。即电压门控钠离子通道基因scn1Laa参与斑马鱼脑神经发育和生长,间接参与运动行为调节。同时本文也为进一步探究电压门控钠离子通道在脑神经中的作用奠定基础。  相似文献

种粮大户是新型农业经营主体的重要组成部分,其健康持续发展不仅关乎我国农业的发展方向,而且关乎亿万农户的切身利益。基于安徽省403户种粮大户的生产经营数据,构建粮食收购市场上买卖双方的议价模型,分析粮食销售渠道和收储设施,探讨种粮大户粮食销售价格的影响因素。结果表明,粮食销售渠道主要有政府(国家粮食收储企业)、合作社、农产品加工企业和个体商贩等,但种粮大户的粮食销售渠道过于单一,以个体商贩为主。晒场和仓库等收储设施情况和销售渠道对种粮大户粮食销售价格有显著影响。在粮食收购市场上,晒场和仓库等必要收储设施的缺乏,使得种粮大户在粮食收获后面临短时期大量售粮的压力,种粮大户在粮食销售时常处于被动地位,被迫接受低于公正基准价格的价格,平均而言最终的销售价格比公正的基准价格低了1.1%。因此,提出通过完善我国的粮食流通市场,完善种粮大户收储设施,解决种粮大户售粮难的问题。  相似文献

AIM: The purpose of this study was to investigate the effect of cyclosporin A on the isolated mitochondria from normal and ischemic rat brains and to observe the possible effect of mitochondrial ATP sensitive potassium channel on mitochondrial permeability transition. METHODS: The swelling of mitochondria isolated from normal and ischemic rat brain was evaluated by pectrophotometry. RESULTS: Cyclosporin A at concentrations of 0.5 μmol/L or 1 μmol/L and diazoxide at concentration of 30 μmol/L significantly decreased the swelling of the normal brain mitochondria induced by 200 μmol/L calcium, which was abolished by atractyloside at 100 μmol/L. However, cyclosporin A at concentration of 5 μmol/L did not affect the mitochondrial swelling. On the mitochondria isolated from ischemic brain, cyclosporin A at 0.5 μmol/L but not 1 μmol/L significantly decreased the mitochondrial swelling, which was cancelled by atractyloside at concentration of 100 μmol/L. Diazoxide at concentration of 30 mol/L had the similar effect with cyclosporin A at 0.5 μmol/L, which was locked by atractyloside at 100 μmol/L or 5-hydroxydecanoate at 100 μmol/L and 200 μmol/L. CONCLUSIONS: Compared with the mitochondria isolated from normal brain, mitochondria from ischemic brain are more sensitive to the inhibition of mitochondria permeability transition pore opening. Activation of mitochondrial ATP potassium channel may be one of the mechanisms by which the opening of mitochondrial permeability transition pore is inhibited.  相似文献

AIM: To investigate the effects of resveratrol on the L-type calcium current in isolated guinea pig ventricular myocytes. METHODS: The whole cell patch clamp method was used. RESULTS: (1)Resveratrol (1, 50, 100 μmol/L) reduced the ICa-L by 18.31%±3.15%, 56.20%±2.50% and 84.51%±4.01% in a concentration-dependent manner (n=5, P<0.05). But it has no change on I-V shape of ICa-L. (2) 8Br-cGMP (100 μmol/L), an activator of protein kinase G(PKG), deduced the density of ICa-L by 10.50%±1.11%. Applying resveratrol and 8Br-cGMP simultaneously decreased the ICa-L significantly by 87.58%±3.49% (n=6, P<0.05). (3) 5 μmol/L H8, a PKG inhibitor, inhibited the decrease in ICa-L caused by resveratrol. CONCLUSION: Resveratrol inhibits ICa-L in guinea pig ventricular myocytes, and this inhibitory effect involves the PKG pathway.  相似文献

AIM: To investigate the role of intracellular free Ca2+ concentration ([Ca2+i) in the regulation of calcium-activated chloride (ClCa) channels in pulmonary artery smooth muscle cells (PASMCs) of rats under normoxic, acute and chronic hypoxic conditions. METHODS: Acute hypoxia-induced contraction was observed in rat pulmonary artery by using routine blood vascular perfusion in vitro. The fluorescence Ca2+ indicator Fura-2/AM was used to observe [Ca2+i of rat PASMCs in normal and chronic hypoxic condition. The influences of ClCa channels on PASMCs proliferation were assessed by MTT assay. RESULTS: (1) The ClCa channel blockers niflumic acid (NFA) and indaryloxyacetic acid (IAA-94) produced inhibitory effects on acute hypoxia-evoked contractions in pulmonary artery. (2) Under chronic hypoxic condition, [Ca2+i was increased. In normoxic condition, [Ca2+i was (123.63±18.98) nmol/L, and in hypoxic condition, [Ca2+i was (281.75±16.48)nmol/L (P<0.01). (3) In normoxic condition, [Ca2+i had no significant change and no effect on ClCa channels was observed (P>0.05). (4) Chronic hypoxic increased [Ca2+i which opened ClCa channels. The NFA and IAA-94 blocked them and decreased [Ca2+i from (281.75±16.48)nmol/L to (117.66±15.36)nmol/L (P<0.01). (5) MTT assay showed that in chronic hypoxic condition NFA and IAA-94 decreased the value of absorbing light degree (A value) from 0.459±0.058 to 0.224±0.025 (P<0.01). CONCLUSION: Hypoxia increased [Ca2+i which opened ClCa channels and had a positive-feedback to [Ca2+i. This may play an important role in hypoxic pulmonary hypertension. In chronic hypoxic condition, ClCa channel may play a role in the regulation of PASMCs proliferation.  相似文献

AIM:To investigate whether calcium activated potassium channel (KCa) and mitochondrial permeability transition pore (mPTP) contribute to cardioprotective effect elicited by anoxic postconditioning.METHODS:The isolated perfused hearts of male Sprague-Dawley rats were subjected to 30 min global anoxic followed by 120 min reoxygenation. Formazan content of myocardium was measured at 490 nm spectrophotometrically,and the level of lactate dehydrogenase (LDH) in the coronary effluent was detected. The absorbance of isolated heart mitochondria at 520 nm was determined. RESULTS:Anoxic postconditioning increased formazan content,reduced LDH release,improved the hemodynamic parameters of the left ventricular developed pressure,maximal rise/fall rate of left ventricular pressure,left ventricular end-diastolic pressure and rate pressure product and attenuated the decrease of coronary flow during reperfusion. Pretreatment with paxilline (1 μmol/L) inhibited the effect of anoxic postconditioning. The opening of mPTP was suppressed in the mitochondria isolated from A/R hearts treated with anoxic postconditioning.CONCLUSION:The findings indicate that in the isolated rat heart,anoxic postconditioning protects myocardium against anoxic/reoxygenation injury via inhibiting KCa and the mPTP opening.  相似文献

AIM:To investigate the inhibitory effects of Cl- channel blocker, tamoxifen, on volume-activated Cl- currents of nasopharyngeal carcinoma cells (CNE-2Z cells) in G1 and S phases. METHODS:Highly synchronous cells in G1 phase and S phase were obtained by the serum starvation and the double-block techniques. The whole-cell patch clamp technique was used to observe the effects of tamoxifen on volume-activated Cl- currents and to analyze the anion permeability of volume-activated Cl- channels. RESULTS:47% hypotonic stimulation activated a Cl- current in the nasopharngeal carcinoma cells at the cell cycle stage G1 phase and S phase. Tamoxifen at concentration of 10 to 30 μmol/L completely inhibited the current. However, the time needed to completely inhibit the current was dose-dependent and was different between G1 phase and S phase. The time needed to completely inhibit the current was shorter in G1 cells than that in S phase cells. The anion permeability sequence of the volume-activated Cl- channel was I->Cl->gluconate in both G1 phase and S phase cells. The permeability of G1 phase cells to I- was higher than that in S phase cells, but to gluconate was lower than that in S phase cells. CONCLUSIONS:The density of the volume-activated Cl- current, the anion permeability of the channel and the sensitivity of the current to tamoxifen were different between the CNE-2Z cells in G1 phase and those in S phase. The results suggest that the expression of tamoxifen-sensitive, volume-activated chloride channels is differentiated at different stages of the cell cycle.  相似文献

AIM: To investigate the inhibitory effects of ATP on proliferation signaling in immortalized human fibroblasts. METHODS: Immortalized human fibroblasts were treated with ATP, ATP conbined with calcium or potassium channel antagonists, respectively. The intracelluar-free calcium ([Ca2+]i), inositol 1,4,5-trisphosphate(IP3) levels and cell viability were detected at different time points. RESULTS: ATP significantly increased the [Ca2+]i and decreased the IP3 level in immortalized human fibroblasts, especially at initial stage (P<0.01) . Compared to ATP alone, the proliferation rates remarkably increased when calcium or potassium channel antagonists were used (P<0.01, respectively) with ATP. CONCLUSION: The calcium and potassium channels and IP3 involved in the inhibitory effects of ATP on the proliferative signaling in immortalized human fibroblasts.  相似文献
