Ulcerative colitis (UC) is a lifelong illness with profound emotional and social impacts, and could cause serious damage to large intestine, especially in colon. However, the pathogenesis of UC remained unclear. The present study attempts to find out the role of matrix metalloproteinases-7 (MMP-7) and lysozyme in the pathogenesis of UC through a mice model induced by dextran sulfate sodium (DSS). The UC model was evaluated both by disease activity index (DAI) and the intestinal histopathology. The results show that there is a high correlation between the DAI score and the pathological changes of colon. Interleukin-6 (IL-6) serum levels and large intestinal fluids levels in UC mice are always higher than that of the control groups, which might be associated with the degree of the inflammation damage in the colon. The change tendency of the MMP-7 mRNA and protein expressions are both up-regulated firstly and then down-regulated from 1 to 5 d in the colon, but only the MMP-7 protein is up-regulated at 7 d again. The up-regulated MMP-7 levels in the early stage of UC may play a protective role through the activated defensins, while the down-regulated levels in the mid-later stage of UC may be connected with the severe lesions in the colon. However, the up-regulated MMP-7 levels in the later stage of UC in the colon may also contribute to the tissue repair or be served as a marker to CRC (colorectal cancer). The distribution of lysozyme protein indicates that there may be Paneth-like cells in the colon. Both the changes of MMP-7 and lysozyme in the small intestine may play a protective role for the safe environment of the whole gut, especially to the colon of UC. 相似文献

【目的】探讨罗丹明B(RB)在Fe(Ⅲ)/UV/空气下的催化降解过程及其机理。【方法】在间歇式光反应器中,研究了pH分别为1.0,3.0和4.3,Fe(Ⅲ)质量浓度为1.68~28mg/L,RB的初始质量浓度为4~150mg/L时RB的光降解过程,通过抑制剂NaN3和异丙醇研究了降解过程可能发生的机理,同时测定了降解过程中Fe2+质量浓度的变化。【结果】RB直接UV光解和Fe(Ⅲ)/UV/空气下的降解表观动力学均符合表观一级动力学规律。RB在Fe(Ⅲ)/UV/空气下降解时,随Fe(Ⅲ)质量浓度的增加,RB降解速率常数增大,但增加程度受到pH的影响,pH=3.0时增加程度最大,pH=1.0次之,pH=4.3时最小;RB初始质量浓度增加,降解速率减小,当RB初始质量浓度高于20mg/L时,继续增加RB质量浓度对其光解速率影响不明显;反应过程中,Fe2+质量浓度在反应初期快速增加,此后维持在一定的水平。【结论】Fe(Ⅲ)/UV/空气能够快速催化RB光解,但在不同pH条件下,Fe(Ⅲ)催化UV光解RB的机理并不相同。 相似文献

Abscisic acid (ABA), arginine and sucrose were evaluated for their effects on the morphology, germination rates and protein content of date palm somatic embryos (SE). Different concentrations of these supplements in the culture medium were used. The comparative study of SE length and thickness between treated and untreated SE revealed no differences, except for ABA (20 μM), which increased thickness. A decrease of water content (WC) in favor of an increase in dry weight (DW) was observed in all treated SE, especially with sucrose (90 g l−1) and ABA (20 μM). Only ABA (20 and 4 μM) caused a proliferation rate of the cultures higher than those in the control. Although all the tested compounds increased protein content, ABA (20 μM) was more effective in protein enrichment than arginine and sucrose treatments. The SDS-PAGE protein profiles showed a significant difference between treated and untreated SE. A protein band of 22 kDa, identified as glutelin in a previous work, was accumulated after treatment with 20 μM ABA or 3 mM arginine. These findings may contribute to further understanding of the mechanisms involved in the accumulation of specific storage proteins in several plants. 相似文献

The present study aimed to determine the most efficient experimental conditions of iron sulfate use leading to optimal inhibition in the development of fungal pathogens. Assays have been focused on fungal species inducing severe grapevine diseases. FeSO4 directly inhibited the in vitro mycelial growth of Botrytis cinerea, Eutypa lata, Phaeomoniella chlamydospora, Phaeoacremonium aleophilum, Diplodia seriata, and Neofusicoccum parvum with variable efficiency in the range of 0.5–10 mM. The development was always completely inhibited at 20 mM. This inhibitory effect was greatly increased at acidic pH values. The anionic moiety of the molecule was of importance since bromide, chloride and sulfate were highly active, whereas acetate and oxalate showed a small effect. Electron microscope observations on E. lata and B. cinerea showed that a treatment with FeSO4 induced dramatic changes in the hyphal organization leading to cell death. No toxicity was observed on grapevine leaves following repeated FeSO4 sprays in the antifungal concentration range. Therefore, FeSO4 may be proposed to effectively replace the long-term pollutant use of CuSO4 as an antifungal agent, with the additional advantage of iron being an important plant micronutrient. 相似文献

硫酸盐环境下,为模拟混凝土灌注桩施工时混入的硫酸盐在桩中的时变分布行为,基于Fick第二定律并结合初始条件与边界条件,应用分离变量法,建立硫酸盐在桩中的时变分布模型。通过算例将本文解与传统解进行比较,并进一步探讨了硫酸盐扩散分布的影响因素和影响规律。结果表明,传统解用于分析混凝土灌注桩桩中硫酸盐扩散有局限性,而本文解相对于传统解具有一定的优势。混凝土灌注桩半径越小,应用传统解分析灌注桩中硫酸盐时变分布误差较大,同时,误差随时间增大。硫酸盐在混凝土灌注桩中的时变分布呈沙漏形,沿半径方向x=0处和桩表面处扩散行为明显,在x=0处附近区域,初始第一年扩散速率较快,在后续的50 a内扩散速率显著减小,但每10 a的平均速率基本相同。扩散系数和初始浓度梯度对硫酸盐在混凝土灌注桩中的时变分布影响较大。 相似文献
