The Azospirillum genus comprises free-living, plant growth-promoting, nitrogen-fixing bacteria found in the rhizosphere of plant roots. Azospirilla are able to promote plant growth mainly through improvement of root development. Bacterial surface components, such as extracellular polysaccharides and proteins, are involved in root colonization. Phase variation – or phenotypic variation – is one of the mechanisms by which microorganisms adapt to environmental changes. This phenomenon is characterized by the presence of a sub-population of the bacteria presenting a different phenotype relative to the major population. In this study we characterized phenotypic variation of Azospirillum brasilense Sp7. When plated on solid media, some A. brasilense colonies were shown to possess a much more mucoid morphology, producing 7.5–8 times more exopolysaccharide with different monosaccharide composition than the parental strain Sp7. The rate of appearance of this kind of variant colonies was 1 in 5000, in agreement with the accepted rate for the phase/phenotypic variation phenomenon. The variants were significantly more resistant to heat and UV-exposure than the parental strain and displayed genomic changes as seen by the different band patterns following ERIC-PCR, BOX-PCR and RAPD analyses. In plant inoculation experiments under greenhouse conditions, with maize, wheat, soybean and peanuts, the EPS overproducing variants performed as similar as the parental strain. Therefore, EPS overproduction did not confer an apparent advantage to A. brasilense in terms of induction of plant growth promotion.  相似文献

A key issue when researching land degradation is the pollution of soils. For bioremediation of contaminated soil, Burkholderia sp. XTB‐5 cells were obtained from soil and grown on mineral salt medium with initial phenol concentrations of 650 mg L−1 and 850 mg L−1, which were found to degrade more than 98% of phenol content in less than 4 days. About 90% of phenol content (with initial concentration of 250 mg kg−1 soil) was removed from soil inoculated with XTB‐5 cells in 6 days. More than 90% of phenol content was removed within 20 days after co‐introduction of XTB‐5 cells and plants to sterilized soil in a greenhouse or to natural soil in field trials. But under the same conditions, individual introduction of plants to sterilized soil in the greenhouse reduced phenol content by about 50% and introduction to natural soil in field trials reduced phenol content by about 38%, suggesting that phytoremediation of phenol is often inefficient and microorganisms can efficiently degrade this pollutant. In addition, strain XTB‐5 was found to solubilize phosphate and produce 1‐Aminocyclopropane‐1‐Carboxylate (ACC) deaminase and siderophore. Strain XTB‐5 promoted plant growth in both phenol‐absent and phenol‐spiked soil under greenhouse and field conditions. Considering that ACC deaminase is beneficial to plant growth under adverse environmental conditions, plant growth promotion by XTB‐5 in phenol‐contaminated soil is not only due to XTB‐5 cell‐degradation of phenol and reduced phytotoxicity but also to production of ACC deaminase. 介绍了杨凌节水农业综合技术体系集成与示范区建设的设计构想。示范区设计以现有节水农业技术在西北半湿润区的适应性和可操作性研究为基础。通过技术筛选、技术集成及示范,形成规范化技术标准。在综合分析节水示范区自然和社会因素的基础上,对节水技术进行组装凝练,建设综合节水示范区,监测示范区的农业和生态用水变化状况,评估节水效益;探索渠灌区节水农业综合示范区管理体制和运行机制,形成一套相对完整的节水农业技术集成和发展模式,形成与市场经济接轨的示范区建设、运行与管理机制;建立示范区节水效益评估方法及监测指标体系。  相似文献

Three kinds of plant growth regulators, gibberellinA 4/7 (GA 4/7 ), 6-benzylaminopurine (BA), and chlormequat chloride (CCC), were evaluated for their ability to promote strobilus and cone production in a Chinese pine (Pinus tabuliformis Carr.) clonal seed orchard. Treatments (0, 250, 500, or 1000 mg L -1 ) were applied during three periods (June- July, July-August and August-September) in 2005. 该文系在完成“九五”国家农业高效用水科技产业示范工程项目“渠灌类型区农业高效用水模式与产业化示范”专题研究的基础之上,结合示范区建设及运行管理等情况总结而成。在分析研究渠灌类型区农业用水存在问题的基础上,首先提出了渠灌类型区实现农业高效用水的基本思路,依据上述思路确立了提高渠灌区输水过程用水效率、田间灌溉过程用水效率与生产效率,以及提高灌区工程运行管理水平的技术与措施;其次,根据以上研究结果进一步确立了渠灌类型区农业高效用水的模式,并建立了相应的实体模型进行工程示范,初步验证和确立了这种高效用水模式。  相似文献
