渗透胁迫下邻羟基苯甲酸对1年生杉木幼苗叶绿素含量的影响 总被引:3,自引:1,他引:2
为研究渗透胁迫下杉木自毒物质邻羟基苯甲酸(SA)对杉木幼苗的毒害作用,用不同浓度的聚乙二醇模拟干旱处理,通过水培方法研究在渗透胁迫下不同浓度(50,100,200 mg/L)杉木自毒物质SA对1年生杉木幼苗叶绿素含量的影响.结果表明:在轻度(-0.3 MPa)、中度(-0.5MPa)、重度(-0.7MPa)渗透胁迫下,杉木幼苗叶绿素含量总体呈下降趋势,且随着渗透胁迫程度的增加,叶绿素含量下降加速.在渗透胁迫下,当自毒物质浓度达200 mg/L时,与对照相比,在第7,14,21 d时,其叶绿素含量分别下降15.39%,25.78%,41.89%. 相似文献

【目的】近年来,RNA m6A甲基化修饰在肌肉发育中的作用不断被发现,通过探究m6A甲基化酶相关基因,包括METTL3,METTL14,WTAP,FTO和ALKBH5,在牛肌肉组织以及骨骼肌卫星细胞(skeletal muscle satellite cells, SMSCs)增殖和分化过程中的表达,同时分析体外成肌分化过程中m6A甲基化水平的变化,为阐明m6A修饰在骨骼肌发育中的作用及机制提供参考。【方法】使用实时荧光定量PCR(RT-qPCR)技术检测m6A甲基化酶相关基因在新生和成年牛不同部位骨骼肌中的表达。随后在秦川牛肉用新品系背最长肌中分离SMSCs,通过生长曲线、免疫荧光和RT-qPCR技术验证SMSCs的增殖和成肌分化功能,使用RT-qPCR检测m6A甲基化酶相关基因在SMSCs增殖期24、36、48、60、72 h和分化第0、2、4、6、8天中的时序表达谱。最后利用LC-MS/MS和Dot blot技术检测SMSCs分化过程中m6... 相似文献

黄土地表生物结皮对土壤贮水性能及水分入渗特征的影响 总被引:5,自引:1,他引:4
采用双环法对山西省偏关县3种结皮覆盖下(苔藓藻结皮、藻结皮和薄层藻结皮)土壤的贮水性能和渗透特征进行了对比研究.结果表明:不同结皮覆盖下土壤的贮水能力受结皮厚度和孔隙度状况的影响较大,0~10 cm土层饱和贮水量为502.69~525.80 t/hm2 ,滞留贮水量为169.71~198.29 t/hm2;初渗速率的变化范围为5. 19~11.10 mm/min,无结皮最高,苔藓藻结皮最低;稳渗速率变化范围为1.6 7~2.67 mm/min.采用的3种入渗模型(Kostiakov模型、Horton模型和Ph ilip模型)中Horton模型的拟合值更接近于实测值, 决定系数R2在0.98~0.99,更适用于描述本研究区具有生物结皮土壤的入渗特征. 相似文献

In this study,to investigate the regulatory role of methyltransferase-like protein 3 (METTL3) on milk protein and fat synthesis in bovine mammary epithelial cells (BMEC),different BMEC models were established, the expression of related genes and localization of METTL3 were detected,and the impact of METTL3 on milk protein and fat synthesis in BMEC after adding β-estrogen (E),methionine (Met),and overexpression or inhibition of METTL3 were measured using Western blotting and imunofluorescience methods. The results showed that when E and Met were added or the METTL3 expression was promoted,the expressions of mTOR,p-mTOR,GlyRS,p-GlyRS,CSN2 and SREBP-1c were increased,oppositely,the gene expression were decreased. Those results indicated that METTL3 could affect the milk fat and protein synthesis by adjusting the expression of related signaling pathway gene.METTL3 was one of the important regulating milk synthetic molecules and had an positively regulation on milk fat and protein synthesis in cells. 相似文献

The objective of this study was to investigate changes occurring in μ/m‐calpain in post mortem chicken muscles and to determine the origin of the unknown bands found in calpain casein zymography. The unknown bands were reported with slightly greater mobility compared to conventional μ/m‐calpain bands in casein zymography. Identification of these bands was accomplished using native polyacrylamide gel electrophoresis, liquid chromatography tandem mass spectrometry and with protein phosphatase treatment. Results showed that the unknown bands were corresponding to μ/m‐calpain, and dephosphorylation by protein phosphatase did not change their appearance. The calpain samples were then incubated with various concentrations of Ca2+ to determine the relationship between changes in μ/m‐calpain and the appearance of the unknown bands. The products of μ/m‐calpain partial autolysis were found to be consistent with the appearance of the unknown bands. Therefore, the appearance of these bands did not result from phosphorylation of μ/m‐calpain as previously hypothesized, but from partial autolysis of μ/m‐calpain. Also their presence suggests that μ/m‐calpain undergoes partial autolysis during aging which may play certain roles in meat quality improvement. 相似文献

To explore the mechanism of MHCⅠ molecule in immune response,chicken MHCⅠα and β2m genes were cloned by PCR.Then the fragments were inserted into the eukaryotic expression vector with fluorescent protein,and the recombinant plasmids pEGFP-MHCⅠα and pmCherry-MHCⅠβ2m were constructed.The recombinant plasmids were transfected into 293T cell with lipofectin reagent.The gene products of recombinant plasmids were mainly located to endomembrane system of the cells by fluorescence microscopy,and changed the intracellular localization of the fusion with the fluorescent protein.Moreover,the positive reactions were observed by the method of Western blotting,and the proteins had the molecular weight of 68.3 and 41.3 ku,respectively,in accord with the target proteins.The results showed that the recombinant plasmids were expressed in 293T cells with a good immunological activity,and the proteins had the binding reaction with specific antibodies. 相似文献

采用正交试验设计方法,研究有机肥比例、种植密度、磷酸二氢钾处理次数等主要栽培措施对杭菊产量、营养品质和药用品质的影响,旨在寻求杭菊在河南郑州中等肥力条件下的最优栽培措施组合。结果表明,3个因素对菊花各项指标综合加权评分值的影响由大到小排列依次为A有机肥比例B种植密度C磷酸二氢钾处理次数;最优组合为有机肥比例75%、种植密度52 500株/hm2、磷酸二氢钾处理3次;在此条件下,大洋菊的平均产量为2 185.65 kg/hm2,营养成分和药用成分含量较协调。这表明该栽培措施非常适宜于河南地区中等肥力土壤条件下杭菊的种植。 相似文献

通过对将乐县马尾松阔叶树混交林进行天然更新、人工促进天然更新和人工营造杉木林等3种更新方式的林分恢复调查,比较探讨其生长量、生物量及物种丰富度,结果表明:不同更新方式对林分生长、林下植物发育及林分物种丰富度影响较大,采用天然和人工促进天然更新形成的阔叶树林分树种多样,结构稳定,生长良好,是保护和发展阔叶林的一种有效途径。 相似文献
