Porcine reproductive and respiratory syndrome virus (PRRSV) has been endemic in Taiwan since 1991. This study aimed to present a highly virulent PRRSV in Taiwan based on farm data collection and both in vitro and in vivo evaluations in virus challenge studies. This virulent PRRSV strain was first noticed on Farm TSYM due to continuously high nursery mortality rate and severe PRRSV-associated pneumonia. In phylogenetic surveillance, the PRRSV TSYM-strain remained in the predominant position for years, even with several other PRRSV strain invasions. In laboratory challenge trials, the TSYM-strain led to prolonged pyrexia, growth retardation, high mortality rates and high viremia titer that similar to the highly pathogenic PRRSV. The TSYM-strain isolate also triggered early interleukin-10 up-regulation and significantly higher infection rates under in vitro experiments. This study provides information of a comparably virulent strain in Taiwan and its appearance in both farm and laboratory levels.  相似文献

This study was to investigate the effect of dietary thiamin on the immune response and intestinal microflora in juvenile Jian carp (Cyprinus carpio var. Jian). Seven diets (0.25, 0.48, 0.79, 1.06, 1.37, 1.63 and 2.65 mg thiamin kg?1) were fed to Jian carp (8.20 ± 0.02 g). At the end of feeding trial, red blood cell counts and white blood cell counts increased with increasing dietary thiamin levels up to 0.79 and 1.06 mg kg?1 diet, respectively (P < 0.05); Lactobacillus counts in intestine increased gradually, and maximum values obtained when the thiamin level was at 0.79 mg kg?1 diet (P < 0.05). After the feeding trial, fish were injected with Aeromonas hydrophila only one time on the first day of the challenge trial and fed the same diets as the growth trial for 17 days. The survival rate, leucocyte phagocytic activity, lectin potency, acid phosphatase activity, lysozyme activity, total iron‐binding capacity and immunoglobulin M content of fish after being injected with A. hydrophila were all improved with increasing dietary thiamin levels up to a point, respectively (P < 0.05). These results suggested that thiamin could improve immune response and the balance of intestinal microflora of juvenile Jian carp.  相似文献

We evaluated the effects of some dietary natural mineral materials as an antibiotic replacer based on growth performance, non‐specific immune responses and disease resistance in juvenile and subadult rainbow trout, Oncorhynchus mykiss. First experiment, juvenile rainbow trout averaging 2.7 ± 0.02 g (mean ± SD) were fed one of the six experimental diets; a basal commercial diet as a control (CON), CON with oxytetracycline (OTC), with yellow loess (YL), with Macsumsuk® (MS), with Song‐Gang® stone (SG) and with barley stone (BS) at 0.4% of each diet. At the end of 8‐week feeding trial, weight gain (WG), specific growth rate (SGR), feed efficiency (FE) and protein efficiency ratio (PER) of fish fed YL diet were significantly higher than those of fish fed CON diet. Non‐specific immune responses such as superoxide dismutase (SOD), myeloperoxidase (MPO), lysozyme (LYS) activity and oxidative radical production of fish fed YL diet were higher than those of fish fed CON diet. At the end of 15 days of challenge test with Aeromonas salmonicida, average cumulative survival rate of fish fed YL diet was significantly higher than that of fish fed BS and CON diets. However, there were no significant differences among fish fed YL, SG and OTC diets. Second experiment, subadult rainbow trout averaging 261.5 ± 3.5 g (mean ± SD) were fed one of the four experimental diets for 22 weeks: CON, and CON with OTC, YL or SG at 0.4% of each diet. At the end of feeding, growth performance of fish fed SG and YL diets was significantly higher than that of fish fed CON diet. Non‐specific immune responses in terms of SOD, MPO, LYS and NBT of fish fed SG and YL diets were significantly higher than those of fish fed CON diet. However, there were no significant differences among the fish fed YL, SG and OTC diets. The results indicate that dietary yellow loess or Song‐gang® stone at 0.4% of diet could replace oxytetracycline in juvenile and subadult rainbow trout.  相似文献

Pathogen interactions with cultured fish populations are well studied, but their effects on native fishes have not been characterized. In Chile, the disease caused by bacterial species Piscirickettsia salmonis represents one of the main issues and is considered to be one of the important pathogens in the field of aquaculture. They have been found to infect native fish. Therefore, it is necessary to understand the impact of P. salmonis on native species of local commercial value, as well as the potential impact associated with the emergence of antibiotic-resistant strains of P. salmonis. Due to this purpose, the native fish Eleginops maclovinus was used in our study. Fish were randomly distributed in tanks and intraperitoneally inoculated with two strains of P. salmonis. No mortality was recorded during the experiment. Cortisol, glucose and total α-amino acid levels increased in fish injected with AUSTRAL-005 strain compared to sham-injected and LF-89-inoculated fish. Moreover, results showed an increase in the activity of carbohydrates and lipids metabolism in liver; and an increase in the carbohydrates, lipids and total α-amino acid metabolism in muscle after injection with AUSTRAL-005. Our results suggest that P. salmonis modulates the physiology of E. maclovinus and the physiological impact increase in the presence of the antibiotic-resistant strain AUSTRAL-005.  相似文献

A bacterial infectivity challenge model of Edwardsiella ictaluri in striped catfish was developed. All experiments were conducted using a bacterial isolate of E. ictaluri that had been recovered during a natural outbreak of bacillary necrosis of Pangasianodon (BNP) in farmed striped catfish Pangasianodon hypophthalmus in Vietnam. Time of immersion in 107 CFU.ml−1 had a significant effect on mortality. The immersion bacterial dose of 107 CFU/ml for 30 s resulted in a cumulative percentage mortality of 63%. Three to four days post-bacterial challenge, fish showed gross clinical signs of natural BNP and E. ictaluri was recovered and identified from these fish. Moreover, a cohabitation challenge was evaluated as an alternative challenge method, although the mortalities among the infected fish were lower at around 15%–40%. This study confirmed the horizontal transmission of E. ictaluri in striped catfish and elucidated that cohabitation challenge could be used in reproducing the disease under controlled conditions.  相似文献

Piscirickettsiosis, caused by the intracellular Gram‐negative bacteria Piscirickettsia salmonis, is at present the most devastating disease in the Chilean salmon industry. The aim of this study was to analyse disease development after challenge with a P. salmonis strain (EM90‐like) under a controlled environment by comparing intraperitoneal challenge with cohabitation challenge. The P. salmonis EM90‐like isolate was cultured in a liquid medium for the challenge of 400 Atlantic salmon (Salmo salar) smolts. Cumulative mortality was registered, necropsy was performed, and bacterial distribution in the tissues and histopathological changes were analysed. The results revealed a similar progression of the disease for the two different challenge models. Pathological and histopathological changes became more visible during the development of the clinical phase of the disease. Bacterial DNA was identified in all the analysed tissues indicating a systemic infection. Bacterial tropism to visceral organs was demonstrated by real‐time quantitative PCR and immunohistochemistry. Better knowledge of disease development during P. salmonis infection may contribute to further development of challenge models that mimic the field situation during piscirickettsiosis outbreaks. The models can be used to develop and test future preventive measures against the disease.  相似文献

Infectious salmon anaemia virus (ISAV) surveillance in the Bay of Fundy has identified the existence of a large number of genetically distinct ISAV isolates which appear to be of variable virulence. Genetically distinct isolates are currently being designated based on sequencing of the hyper polymorphic region (HPR) of genomic segment 6, which encodes the haemagglutinin–esterase protein, but it has been difficult to elucidate a clear association between these molecular variations and variations in virulence. This has hampered the establishment of proactive management decisions regarding infected fish, and ISAV infections, regardless of type, must be treated as one. Field data of ISAV infections is difficult to collect and to compare between infections because of a wide range of confounding factors including time of year, fish stock, cage site location, mitigating factors and stressors. An important tool in determining the relationship between molecular differences and virulence comes from analysis of quarantine studies. The goal of this study was to compare the virulence, by co-habitation and intraperitoneal injection, of four regionally common and recent ISAV isolates in a controlled environment. We found significant differences in mortality between ISAV molecular isolates, and present data showing that survival of ISAV infection confers significant resistance to re-infection with a different ISAV isolate. These findings, if borne out in field studies, will significantly alter the way ISAV infections are managed in the Bay of Fundy and elsewhere.  相似文献

A challenge model for comparison of the virulence of epizootic haematopoietic necrosis virus (EHNV) to European stocks of redfin perch, Perca fluviatilis L., and rainbow trout, Oncorhynchus mykiss (Walbaum), was tested. The model investigated intraperitoneal (IP), bath and cohabitation routes at 10, 15 and 20 °C for 5–6 g fish and 15 °C for 20 g perch. In the IP challenges of perch, significant mortality occurred at 15 °C and 20 °C. In challenge trials for rainbow trout, significant mortalities were observed in IP and bath challenges at 20 °C. The mortality observed in IP challenged 20 g perch was not significantly different from that recorded for 6 g fish challenged IP. No significant mortality was observed in any other treatment groups. Re-isolation of ranavirus was confirmed by IFAT and was consistently associated with dead or moribund fish in the trial groups challenged with EHNV. The findings indicate that EHNV does not pose a high risk for wild perch and trout populations in Europe by natural exposure. Mortality appears to be primarily a function of environmental factors, with temperature playing an important role, and not just the presence of the virus in the fish.  相似文献

2018年,在中美贸易摩擦不断加剧的新形势下,保障国家粮食安全的任务更加艰巨。中国粮食依靠传统方式来实现增产、增效必将难以持续,需依靠科技创新走内涵式增产增效道路来提升粮食产业竞争力。通过对近几年粮食生产最新数据的统计分析,采用实地调研、文献梳理、定性分析等方法对新时期科技创新支撑粮食安全的新情况、新问题进行了深入研究。当前中国科技创新在保障粮食安全方面的突出问题包括:科技成果转化最后一公里的"诅咒"难以破解、科技创新体系不健全、科研经费投入结构有待优化和技术服务人才不足等。笔者提出深化粮食科研院所改革,构建上下协同、功能互补、配置高效的粮食科技协同创新体系,持续推进粮食品种与区域结构调整,加强精深加工与冷链物流等技术与设备研发,开展粮食绿色增产模式攻关,建立粮食作物全程机械化生产技术与装备体系等方法,促进粮食安全科技保障能力稳步提升。  相似文献

The aim of this study was to determine the best synbiotic combination (based on growth and short‐chain fatty acids production) between Enterococcus faecalis and eight prebiotics. Based on the results of in vitro studies, E. faecalis + galactooligosaccharide (EGOS) and E. faecalis + mannanoligosaccharide (EMOS) were selected as synbiotics. A 126‐day feeding trial was conducted to compare the effects of prebiotics, probiotic and synbiotics on the growth indices, In vivo ADC of nutrients, digestive enzymes, hemolymph indices and finally, biological responses against 48‐hr Aeromonas hydrophila exposure challenges of juvenile (4.13 ± 0.12 g) crayfish. The highest values of SGR (2.19% body weight day?1), VFI (2.75% body weight day?1), survival rate (96.67%) and the lowest FCR (2.33) were observed in the juvenile crayfish fed the EGOS‐ diet. The significantly (p < .05) highest means of in vivo ADCOM, in vivo ADCCP, in vivo ADCCF and in vivo ADCGE were measured in crayfish fed the EGOS‐ diet. The mean survival rate of Aeromonas hydrophila‐injected crayfish fed the EGOS‐ diet (56%) was significantly (< .05) higher than those of fed the control (8.67%) and other diets (22.67–35.32%). At the levels tested, 7.86 log CFU E. faecalis g?1 + 10 g kg?1 GOS in the diet was considered optimum.  相似文献
