AIM:To investigate the effects of siRNA targeting integrin-linked kinase (ILK) on the expression of glycogen synthase kinase 3β (GSK-3β) and β-catenin during epithelial-mesenchymal transition (EMT) in human kidney proximal tubular epithelial cell line HKC induced by high glucose. METHODS:HKC cells were divided into 4 groups:normal glucose (NG) group, high glucose (HG) group, HG+HK (a vector containing the non-specific siRNA designed as negative control) group and HG+ILK siRNA group. The inverted fluorescence microscope was used to examine the expression of green fluorescent protein (GFP). The expression of ILK at mRNA and protein levels was detected by RT-PCR and Western blotting. The expression of p-GSK-3β and β-catenin was observed by immunocytochemical staining. The protein expression of total GSK-3β, p-GSK-3β, nuclear β-catenin, total β-catenin, E-cadherin and α-smooth muscle actin (α-SMA) was measured by Western blotting. RESULTS:GFP was observed in HKC cells, indicating that the transfection was successful. Both the protein and mRNA of ILK were down-regulated in HG+ILK siRNA group compared with HG group and HG+HK group, but still higher than those in NG group. Silencing of ILK down-regulated the expression of p-GSK-3β and nuclear β-catenin. No difference of total GSK-3β or total β-catenin was observed among the 4 groups. CONCLUSION:These data support a functional role of ILK, GSK-3β and β-catenin in tubular EMT induced by high glucose. ILK may promote tubular EMT by regulating the activity of GSK-3β and β-catenin, the downstream effectors of the Wnt/β-catenin pathway. 相似文献

AIM:To study the effect of small interfering RNA (siRNA) on the expression of beta 2-microglo-bulin (β2M) in pre-differentiated bone marrow mesenchymal stem cells (BMSCs). METHODS:The β2M siRNA was transfected into the pre-differentiated BMSCs with Lipofectamine 2000. BMSCs were divided into transfection group, blank control group and negative control group. The expression of β2M at mRNA and protein levels was determined by real-time qPCR, Western blotting and laser confocal microscopy. The productions of aggrecan and type II collagen in pre-differentiated BMSCs were determined by toluidine blue staining and type Ⅱ collagen immunofluorescence. RESULTS:The results of real-time qPCR, Western blotting and laser confocal microscopy showed that siRNA successfully inhibited the expression of β2M at mRNA and protein levels in the pre-differentiated BMSCs. The results of toluidine blue and type Ⅱ collagen immunofluorescence staining showed that siRNA does not affect the productions of aggrecan and type Ⅱ collagen in the pre-differentiated BMSCs. CONCLUSION:siRNA targeting β2M reduces the expression of β2M in the pre-differentiated BMSCs and does not affect the chondrocyte characteristics of pre-differentiated BMSCs. 相似文献

Abstract – Parr of brown trout ( Salmo trutta L.) were exposed to constant or fluctuating water level and flow in artificial outdoor stream channels during 21 days in three consecutive runs, and their individual behaviour and social interactions were monitored. Despite the large variation in flow, no clear differences in growth, which was generally poor, and behaviour were found between treatments. The signs of a social structure in the stream channels were very subtle and there were no clear relation between growth rate and aggressive/dominant behaviour in any of the treatments in accordance with findings in studies of growth and dominance in the wild. Thus, in regulated rivers, daily fluctuations in flow and water level (hydropeaking) are not likely to alter the behaviour of juvenile brown trout or affect their energy turnover negatively. Several studies now indicate that if stranding can be avoided, hydropeaking has relatively small direct effects on stream salmonids. Therefore, future research should focus on cascading ecological effects that may influence the productivity of the ecosystems in regulated rivers. 相似文献

为了探究黄瓜苗期生长发育规律和育苗方式,以华南型黄瓜‘沪杂7号’和荷短型黄瓜‘春秋王2号’为试材,用卡尺对植株高度、茎粗度、子叶大小、叶片大小进行测量,用天平称量地上部、根鲜质量,电子秤称量产量,利用LI-6400型光合仪对气体交换参数净光合速率(Pn)、气孔导度(Gs)、胞间CO2浓度(Ci)与蒸腾速率(Tr)进行测量。结果表明,黄瓜植株高度、茎粗度增长均似线形,育苗后期生长速度更快,叶片长度、宽度增长速度前期较快,后期略慢;定植前大苗光合作用明显好于小苗,地上部、根鲜质量均显著高于小苗;华南型、荷短型黄瓜大苗定植,较小苗定植采收期均提前10 d以上,同时总产量明显增加。大苗定植对黄瓜提前上市及增加收入有明显效果。 相似文献

- The Pyrenean desman (Galemys pyrenaicus) is an endangered stream-dwelling insectivore endemic to the Iberian Peninsula and the north of the Pyrenees. It favours riffles over slow habitats such as runs or pools, yet it is still unclear whether habitat preference is based on prey availability or on other factors, such as mechanical constraints on its hunting habits.
- Desman diet and prey selection were analysed along the pristine Elama Stream and the slightly modified Leitzaran Stream in the Basque Country, Spain.
- In each stream the prey consumed were identified by metabarcoding 94 desman faeces, and prey availability characterized from 10 Surber samples taken in each habitat type (riffles, runs and pools). Invertebrates were sorted and identified, the biomass of each taxon was calculated, and their DNA was also extracted, amplified using PCR, and sequenced to build a reference database.
- Prey availability and diet varied between streams. Desmans positively selected shredders and invertebrates that live on the substrate, and selected against taxa that live in fine sediment, taxa with hard shells or those that are highly mobile. The diet of desmans was more selective in the Elama, the stream with better ecological status. Food availability did not differ among habitats, and therefore does not explain the preference of desmans for riffles; rather, mechanical constraints to deal with buoyancy and physical habitat heterogeneity were hypothesized to be the reason behind the habitat preference.
- The results suggest that promoting the formation of riffles in streams can improve the situation of some desman populations living in degraded habitats.

- The history of conservation of the Amazon can be viewed as a war involving many battles with interests in agribusiness on one side and in biodiversity conservation and sustainability on the other side. Trends in large-scale deforestation in the 1970s spurred a series of policies, stakeholder alliances and international and grass-roots movements, which decades later led to the establishment of protected areas and interventions in soy and beef supply chains of agribusiness. Together, these advances epitomized a conservation framework for the Amazon, which at one point nearly curbed deforestation in the Brazilian Amazon, although it included very few protections for freshwater ecosystems.
- While those conservation advances were taking place, however, a series of policy changes started to undermine them through expansions in deforestation, river regulation and mining activities. The election of Brazil's President Jair Bolsonaro in 2019 then hit the Amazon conservation framework much like a tsunami of policy setbacks and the re-establishment of the economic policies that sparked the Amazon war in the past.
- The current trajectory is one of large-scale degradation of Amazonian ecosystems and biodiversity with consequent impacts on local people. Because freshwater ecosystems are highly sensitive to human activities on water and on land, these growing impacts are particularly large.
- It is too early to know, but four decades of institutional and policy developments to conserve the Brazilian Amazon may soon be pushed past the point from which they will be able to recover. Four conditions will be pivotal to allowing the Amazon conservation framework to recoup: (a) the end of Bolsonaro’s mandate in 2022 or earlier; (b) remobilization of stakeholders; (c) investments in environmental research, policy and multiple collaborations; and (d) moving conservation beyond terrestrial landscapes to also encompass freshwater ecosystems and their people.
