本文以外周血淋巴细胞姐妹染色单体互换(SCE)频率和细胞周期动力学为指标,以较长期(4~13年)生活在砷污染区的黄牛和水牛为对象,对砷在动物体内慢性蓄积所诱发细胞遗传学效应进行了研究。其结果表明,砷污染区黄牛和水牛SCE频率均显著高于对照组,参照有关评价标准,可判为弱阳性,提示在当地污染水平下,黄牛和水牛都诱发了一定程度的细胞遗传学效应。对细胞周期动力学的观察结果表明,砷对黄牛和水牛没有明显的细胞毒性。本试验还报告了正常黄牛和水牛外周血液淋巴细胞SCE的自发频率以及培养72小时各期细胞的正常比率。  相似文献

A new assay for the detection of swine influenza virus (SIV) was developed with a novel nucleic acid probe——Molecular beacon in this study. The specific primers and molecular beacon probes were designed according to the conserved region of H3 and N2 genes of SIV H3N2 subtype. A digital RT-PCR assay was developed for detection of SIV H3N2 subtype. The results showed that SIV H3N2 subtype could be identified simultaneously on this microarry with high sensitivity and reproducibility,which could reach to 106 dilute viruse. The conclusion was that the digital RT-PCR method could analyze quantitatively the RNA templates.On the identification of H3N2 SIV,the digital RT-PCR method was much more scientific than Real-time quantitative PCR method.  相似文献

猪传染性胃肠炎病毒TaqMan荧光定量RT-PCR检测方法的建立   总被引:4,自引:0,他引:4  
根据猪传染性胃肠炎病毒和猪肌动蛋白的基因序列设计合成了引物和探针,通过对荧光定量RT-PCR反应条件的优化,建立了TaqMan荧光定量RT-PCR检测TGEV的方法。同时对37份现地病料进行检测并与常规RT-PCR方法、TGEV抗原快速检测试剂盒比较。结果,该方法的检测敏感性达到15.3拷贝/μL,且具有很好的特异性和重复性,而常规RT-PCR方法只能检测到1.53×103拷贝/μL。对37份现地病料的检测结果也表明该法(检出17份)比常规RT-PCR方法(检出12份)和TGEV抗原快速检测试剂盒(免疫层析法,检出10份)的敏感性高。  相似文献

采用液氮冷冻研磨法破碎吉陶单极虫(Thelohaneluskitauei)孢子,按常规法提取其基因组DNA。取DNAEcoRI消化产物,采用低熔点琼脂糖回收法制备大(3.0~15.0kb)、小(0.5~3.0kb)片段,将其克隆于pBluescriptksEcoRI位点,经α-互补现象、酶切鉴定和虫体DNA生物素标记探针筛选,构建了含23个克隆的基因文库,克隆片段介于0.9~6.8kb之间;经阳性克隆标记筛选及特性分析,制备了长度为0.9kb(19号质粒克隆片段)的探针,该探针具有较强的敏感性和特异性。对该探针两端DNA序列进行分析,设计出1对引物。上游引物序列为:5′TTTCGAACCCGCACAACAAG3′,下游引物序列为:5′TGAGTGGATAG-TATCGCTGC3′。应用该对引物对虫体进行了检测  相似文献

本研究以质粒pGEM-7Zf(+)为载体,对我国北方和南方疫区伊氏锥虫动基体株的kDNA小环进行了克隆,并以其中之pTK011-C_1为探针对不同地理区的动基体株和无动基体株的不同DNA进行杂交,结果表明,我国北、南两大疫区动基体株的kDNA小环大小均约为1kb,且均属A型,pTK011·C_1只与动基体株的kDNA及tDNA杂交,不与其nDNA杂交,也不与无动基体株的任何DNA及黄牛白细胞DNA杂交。说明具有特异性,可以用于诊断。tDNA的杂交信号与kDNA相当,诊断中可以代替kNDA作为检测对象。  相似文献

根据GenBank已公布的狂犬病病毒(rabies virus,RV)核蛋白(N)基因序列设计并合成一对特异性的引物和探针,建立基于TaqMan探针的荧光RT-PCR检测狂犬病病毒方法。对狂犬病疫苗提取核酸后进行RT-PCR扩增,将目的条带切胶回收,克隆测序,重组质粒作为标准阳性对照。对建立的TaqMan探针荧光RT-PCR方法做灵敏度、特异性、重复性及稳定性试验。结果显示,该方法可以达到10拷贝/μL的灵敏度,可以将狂犬病病毒与犬瘟热病毒、犬细小病毒、犬腺病毒、犬冠状病毒和犬副流感病毒分开,方法重复性好,稳定可靠。  相似文献

Objective To estimate mean Schirmer tear test (STT) and intraocular pressure (IOP) values in healthy koalas both conscious and anesthetized. Methods Data were gathered from koalas in Victoria, Australia. Conscious examinations were performed on captive koalas. Free‐ranging (wild) koalas were examined under anesthesia. Anesthesia was induced using alfaxalone, and animals were maintained on oxygen and isoflurane if required. All animals were healthy and had no surface ocular pathology detectable during slit lamp biomicroscopy. STT I tests were performed using commercial STT test strips placed in the lower fornix for 1 min. IOP was measured using an applanation tonometer after topical anesthesia. The higher value of the two eyes for both STT and IOP was analyzed. STT was measured in 53 koalas (34 conscious, 19 anesthetized) and IOP was measured in 43 koalas (30 conscious, 13 anesthetized). A two‐sample t‐test was used to compare means. A P‐value <0.05 was regarded as significant. Mean ± SD is presented. Results The mean higher STT in conscious koalas was 10.3 ± 3.6 mm wetting/min and in anesthetized koalas it decreased to 3.8 ± 4.0 mm wetting/min (P < 0.0001). The mean higher IOP in conscious koalas was 15.3 ± 5.1 mmHg, and in anesthetized koalas it was 13.8 ± 3.4 mmHg (P = 0.32). There was no effect of sex on either STT or IOP. Conclusions The mean and SD of STT and IOP values for koalas both conscious and anesthetized were reported. The mean STT was significantly reduced by alfaxalone anesthesia.  相似文献

High mean left atrial pressure (MLAP) due to canine degenerative mitral valve disease is associated with clinically relevant morbidity and mortality. The ability to noninvasively measure MLAP would assist in the diagnosis and treatment of disease. Doppler echocardiography allows measurement of early transmitral blood flow (E) and the velocity of the mitral valve annulus (Ea). The ratio of early mitral inflow velocity to early mitral annular velocity (E: Ea) correlates well with MLAP in human subjects. We sought to determine the ability of E: Ea to predict MLAP in dogs with experimentally induced mitral regurgitation. Nine anesthetized purpose-bred dogs underwent placement of a Swan-Ganz catheter into the left atrium and recording of MLAP. Simultaneous transthoracic echocardiographic and hemodynamic studies were performed after acute chordae tendineae rupture and during IV infusion with nitroprusside (2.5-5.0 microg x kg(-1) x min(-1)) or hydralazine (1-1.5 mg/kg). Mitral regurgitant fraction, measured by single-plane angiography and thermodilution, ranged from 17% to 81%. MLAP increased from 5.4 +/- 2.5 mm Hg to 17.4 +/- 9.4 mm Hg after creation of mitral valve regurgitation (MR; P = .018). Forty sets of echocardiographic measurements were obtained from 7 dogs, and E, as well as E: Ea, were linearly related to MLAP. The R2 value for the linear regression equation containing E: Ea as the dependent variable (0.83) was greater than that for E (0.73). The 95% confidence intervals were calculated for predicting MLAP = 20 mm Hg from E:Ea, and E:Ea >9.1 or <6.0 indicated a 95% probability that MLAP was >20 mm Hg or <20 mm Hg, respectively. Echocardiography can be used to predict MLAP in isoflurane-anesthetized dogs with experimentally induced acute mitral valve insufficiency.  相似文献
