科尔沁草甸湿地土壤碳氮剖面分布及生长季动态特征 总被引:1,自引:0,他引:1
采用定位观测的试验方法,于2016年5-10月及2017年8月对科尔沁草甸湿地0~100 cm土层土壤有机碳、全氮剖面分布及生长季动态特征进行了实验分析,旨在为科尔沁草甸湿地保护提供科学指导并为干旱半干旱地区湿地土壤碳氮储量估算提供借鉴。结果表明:1)科尔沁草甸湿地土壤有机碳、全氮含量整体随土层深度下降,0~20 cm土层间下降显著,20 cm以下趋于相对稳定,范围分别为11.9~23.5 g·kg-1和0.66~1.50 g·kg-1。2)各土层土壤碳氮含量月间差异显著(全氮40~60 cm土层除外),变化幅度随土层深度先减小后增大;土壤碳氮密度(100 cm)生长季变化大于年际变化,有机碳密度全生长季呈上升趋势,范围为15.44~20.82 kg·m-2,全氮密度生长初期明显下降,之后趋于相对稳定,范围为1.01~1.16 kg·m-2。3)土壤有机碳含量与全氮含量呈极显著正相关;植被和水文是影响其分布、变化的关键因子。科尔沁草甸湿地生长季土壤有机碳、全氮密度变化较大,且表现为潜在的碳汇和氮源,但年际间碳汇潜力未充分发挥,本研究建议禁牧力度应加大并增加氮肥投入以提高科尔沁草甸湿地生态系统功能。 相似文献

本研究旨在通过高效液相色谱-质谱联用(LC-MS)和高效液相色谱(HPLC)技术对沙冬青种子总黄酮主要活性成分进行分析及含量测定,并探索沙冬青种子总黄酮对小鼠免疫功能的影响。采用LC-MS分析沙冬青种子总黄酮主要活性成分,在HPLC优化条件下用标准品对主要活性成分的含量进行测定;采用MTT法和ELISA试剂盒检测沙冬青种子总黄酮对小鼠脾淋巴细胞增殖和IL-2、IL-4生成的影响,以及总黄酮对肝脏KCs细胞的增殖和NO、NOS含量的影响;运用定量溶血分光光度法和血清溶血素HC50测定法检测不同浓度沙冬青种子总黄酮对小鼠抗体生成细胞和血清溶血素的影响。结果显示,沙冬青种子总黄酮中主要活性成分为芒柄花素、7,3'-二羟基-4'-甲氧基黄酮、黄豆黄素和穗花杉双黄酮,其中,芒柄花素含量高达52.48%,其次为7,3'-二羟基-4'-甲氧基黄酮,含量为11.20%。在2.5~100 μg/mL总黄酮作用下,沙冬青种子总黄酮对脾淋巴细胞的增殖和细胞因子的产生具有明显的促进作用,尤其在20 μg/mL时脾淋巴细胞增殖率和IL-2的分泌量分别高达217.62%和159.661 pg/mL,与空白对照组相比差异极显著(P<0.01);在40 μg/mL时,脾淋巴细胞IL-4的分泌量高达149.274 pg/mL,极显著高于空白对照组(P<0.01)。沙冬青种子总黄酮对KCs细胞的增殖及NO和NOS分泌也具有明显促进作用,在60 μg/mL时,与空白对照组相比,KCs细胞增殖率达67.77%,NO和NOS的分泌量分别达180.106和29.942 μmol/L(P<0.01)。此外,沙冬青种子总黄酮能明显促进小鼠体内抗体生成细胞和血清溶血素含量的增加。综上表明,沙冬青种子总黄酮主要活性成分为芒柄花素和7,3'-二羟基-4'-甲氧基黄酮。沙冬青种子总黄酮对小鼠免疫活性细胞的增殖、相关免疫细胞因子和抗体的产生与释放等都具有显著的促进作用。 相似文献

Three rumen‐protected lysine (RPL) products (AjiPro®‐L, LysiPEARLTM, and Feedtech Bypass LysineTM: A, B, and C, respectively) were tested for stability in two forage‐based total mixed rations (TMR1, 41.3% dry matter (DM), and TMR2, 49.5% DM) (experiment 1) and for Brix value (experiment 2). In experiment 1, each RPL product (2 g each) and TMR diet (200 g) were mixed and stored in plastic bags at 20°C for 0, 1, 3, 6, 12, 24, and 48 hr. In experiment 2, each RPL product (2 g) was dispensed into ion‐exchanged water (20 ml) and kept at 20°C for 0, 0.5, 1, 3, 6, 12, 24, and 48 hr. At each time point, free lysine (Lys) content and Brix values of extracts were measured, and Lys release (LR, %) was calculated. All RPL products LR% varied with varying diets DM and increased with increasing of time exposed to diets; it was highest in C, followed by B, and then A. Water LR% positively correlated with that from diets and with Brix values of Lys dissociated in water. Our results indicated that Lys dissociation from RPL products is affected by diet DM content. Brix value may be used as a potential marker for RPL protection efficacy. 相似文献

为探索土地利用方式变化对土壤理化性状的影响,以青藏高原青海省海北州原生矮嵩草(Kobresia humil)草甸、原生灌丛草甸、退化矮嵩草草甸、退化灌丛草草甸、人工草地、农田地为研究对象,对各利用方式下土壤的基本理化性状进行研究.结果表明:退化导致0~10 cm土层土壤容重增大,而对下层土壤容重没有明显影响,2种退化草地0~10 cm土层的土壤容重均大于10~20 cm土层,其中退化灌丛草甸与原生灌丛草甸之间差异显著(P<0.05),人工种植也导致表层土壤容重有升高的趋势,人工种草和开垦均导致0~20 cm土层土壤容重的变异消失.草地退化和开垦均导致土壤有机碳和全氮含量降低,草地退化还导致土壤 相似文献

ObjectiveTo evaluate the effects of a constant rate infusion (CRI) of lidocaine alone or in combination with ketamine on the minimum infusion rate (MIR) of propofol in dogs and to compare the hemodynamic effects produced by propofol, propofol-lidocaine or propofol-lidocaine-ketamine anesthesia.Study designProspective, randomized cross-over experimental design.AnimalsFourteen adult mixed-breed dogs weighing 15.8 ± 3.5 kg.MethodsEight dogs were anesthetized on different occasions to determine the MIR of propofol alone and propofol in combination with lidocaine (loading dose [LD] 1.5 mg kg?1, CRI 0.25 mg kg?1 minute?1) or lidocaine (LD 1.5 mg kg?1, CRI 0.25 mg kg?1 minute?1) and ketamine (LD 1 mg kg?1, CRI 0.1 mg kg?1 minute?1). In six other dogs, the hemodynamic effects and bispectral index (BIS) were investigated. Each animal received each treatment (propofol, propofol-lidocaine or propofol-lidocaine-ketamine) on the basis of the MIR of propofol determined in the first set of experiments.ResultsMean ± SD MIR of propofol was 0.51 ± 0.08 mg kg?1 minute?1. Lidocaine-ketamine significantly decreased the MIR of propofol to 0.31 ± 0.07 mg kg?1 minute?1 (37 ± 18% reduction), although lidocaine alone did not (0.42 ± 0.08 mg kg?1 minute?1, 18 ± 7% reduction). Hemodynamic effects were similar in all treatments. Compared with the conscious state, in all treatments, heart rate, cardiac index, mean arterial blood pressure, stroke index and oxygen delivery index decreased significantly, whereas systemic vascular resistance index increased. Stroke index was lower in dogs treated with propofol-lidocaine-ketamine at 30 minutes compared with propofol alone. The BIS was lower during anesthesia with propofol-lidocaine-ketamine compared to propofol alone.Conclusions and clinical relevanceLidocaine-ketamine, but not lidocaine alone, reduced the MIR of propofol in dogs. Neither lidocaine nor lidocaine in combination with ketamine attenuated cardiovascular depression produced by a continuous rate infusion of propofol. 相似文献

Background: Direct colloid osmometry provides an objective assessment of the oncotic effects of crystalloid or colloidal fluid therapy, which is especially useful in monitoring fluid therapy of critically ill camelids due to their tendency toward nonspecific hypoproteinemia with increased risk of developing edema and ascites. Objectives: The aims of this study were to measure colloid osmotic pressure (COP) of alpacas and llamas, determine its correlation with concentrations of total protein (TP) and total solids (TS), as well as both albumin (A) and globulin (G) concentrations in the same model (A+G), and evaluate the effects of sample type and storage conditions on COP. Methods: Blood was collected from clinically healthy alpacas (n=23) and llamas (n=22) into heparin tubes. COP of fresh whole blood (COPFB) and plasma (COPFP) was determined using a membrane osmometer. For 20 alpacas, COP of refrigerated whole blood (COPRB) and frozen plasma (COPFrP) was also measured. Correlations between COPFB and TS, TP, and A+G concentrations were assessed by simple and multiple regression analysis to model potential predictors. Results: Median COPFB from alpacas (24.6 mmHg, range 19.3–28.1) was not significantly different from that of llamas (25.3 mmHg, range 22.5–33.7). Sample type or storage conditions did not affect COP. Measured COP had a strong positive linear correlation with TS, TP, and A+G concentrations in alpacas (r2=.7, .74, and .88, respectively). In llamas, COP correlated best with TS concentration (r2=.59), whereas correlation with TP and A+G concentrations was poor (r2=.19 and .25, respectively). Conclusion: COP can be measured using heparinized whole blood or plasma, either fresh or stored. Direct measurement is recommended whenever quantitative knowledge of COP is required in clinical or research setting. Further studies are needed to verify if the poor association of COP with TP found in this study can be generalized to llamas. 相似文献

为了建立能特异检测不同基因型猪瘟病毒(Classical swine fever virus,CSFV),同时又能区分其他瘟病毒的基因检测方法,本实验针对CSFV基因组5′端非编码区设计并合成了简并引物和TaqMan探针,在优化反应条件的基础上,成功地建立了特异检测CSFV的荧光定量RT-PCR检测方法。再以已知滴度的CSFV石门株血毒总RNA反转录产物建立标准品,该标准品可以用于定量临床样品中的CSFV滴度,所建立的荧光定量PCR方法可以灵敏地检测出10~(-0.82)个TCID_(50)病毒含量。最后用建立的方法对108份临床样品进行检测并同时进行病毒分离,荧光定量PCR方法检测出73份阳性样品且与病毒分离的符合率为100%,而常规RT-PCR只检测出54份阳性样品,表明本荧光定量RT-PCR法在检测猪瘟病料上具有潜在的应用价值。 相似文献

对15例临床上有明显低磷血症的奶牛血清磷、红细胞膜脂质成分进行了测定。结果表明,低磷血症奶牛血清磷、红细胞膜磷脂明显降低;红细胞膜胆固醇及膜胆固醇/磷脂摩尔比值(nch/npl)明显升高,与健康对照组牛相比差异极显著(P<0.01);直线相关分析表明,血清磷与红细胞膜磷脂之间呈极显著正相关(r=0.917,y=10.852x+3.196,P<0.01);与红细胞膜胆固醇以及nch/npl之间分别呈显著负相关(r=-0.940,y=2.850x-1.072,P<0.01;r=-0.920,y=1.968x-1.401,P<0.01)。此外,红细胞膜微粘度与膜磷脂之间呈显著负相关(r=-0.954,y=19.122x-4.384,P<0.01),而与膜胆固醇以及nch/npl之间呈显著正相关(r=0.988,y=0.927x+0.964,P<0.01;r=0.978,y=-0.293x+1.113,P<0.01)。结果表明,磷是红细胞膜脂质成分改变的先决因素,而后者又是红细胞膜流动性及膜结构、功能发生变化并导致膜损伤的一个重要因素。 相似文献
