The Effects of Selenium Source on Measures of Selenium Status of Mares and Selenium Status and Immune Function of Their Foals

The effect of organic and inorganic sources of selenium (Se) on measures of Se status of mares and their foals, Se concentrations of colostrum and milk, and indices of immune function of foals was studied. Twenty late-gestation Standardbred mares were randomly assigned to one of two groups. All mares received an identical balanced ration, except for the source of supplementary Se: one group received organic Se (Se yeast) and the other group received inorganic Se (sodium selenite), fed to deliver 0.3 mg/kg supplementary Se based on total ration dry matter. Mares received the experimental diet from 2 months before estimated due date until 1 month after foaling. Source of Se did not affect Se concentrations in maternal plasma, red blood cells, colostrum, or milk. At 1 month of age, foals from mares fed organic Se had higher red blood cell Se concentration than foals from mares fed inorganic Se (P < .05). Measures of immunity included serum immunoglobulin G concentration, lymphocyte proliferation in response to concanavalin A, and relative cytokine gene expression of stimulated lymphocytes (interferon gamma [IFNγ], interleukin [IL]-2, IL-5, IL-10, tumor necrosis factor alpha [TNFα]) and neutrophils (IL-1, IL-6, IL-8, IL-12, TNFα). Relative gene expression of IL-2, TNFα, and IFNγ by foal lymphocytes was associated with the source of Se supplementation provided to the mares. We conclude that the source of dietary Se provided to mares may influence the immune function of foals at 1 month of age through changes in relative gene expression of certain lymphocyte cytokines.     

犬IFN-α优化基因的真核表达及抗病毒活性的研究

本研究根据酵母系统密码子的偏嗜性,对成熟蛋白基因核苷酸序列进行密码子偏嗜性改造,利用毕赤酵母分泌型表达载体pPICZαC来表达犬IFN-α成熟蛋白基因。并采用非洲绿猴肾细胞(Vero) -水疱性口炎病毒(VSV)系统以细胞病变抑制法为基础来测定重组犬α干扰素的效价,同样方法测定了表达产物对PRV和CDV的抑制作用。本试验实现了犬α干扰素在酵母中的高表达,表达量高达58 mg/L,SDS-PAGE检测大小约为24 ku,比预测分子质量大,推测发生了糖基化。表达产物具有较强的种属特异性,在Vero细胞上对PRV具有较高的抗病毒活性,达到了3.6×10⁷ U/L,对CDV也有很好的抑制作用。本研究为犬IFN-α以后的临床应用研究奠定基础。     

BPV1贵州株L1基因序列分析及原核表达

为表达牛乳头瘤病毒1型贵州株(BPV1-GZLZ株)衣壳蛋白L1基因,采用PCR方法从贵州省六枝特区某规模化养牛场疑似病牛的皮肤肿瘤病料样品中扩增BPV1 L1基因,克隆至p MD19-T载体,测序后进行生物信息学分析;同时将BPV1-GZLZ株L1基因克隆于原核表达质粒p ColdⅠ中,经IPTG诱导L1蛋白表达,并对重组蛋白进行SDS-PAGE和Western blotting分析。生物信息学分析显示,BPV1-GZLZ株L1基因核苷酸序列长为1488 bp,编码495个氨基酸,分子结构式为C2484H3884N678O737S16,理论等电点(PI)为8.68,二级结构主要包括α-螺旋、无规则卷曲和β-折叠为主,无跨膜结构域和信号肽; BPV1-GZLZ L1基因序列与BPV1参考株的核苷酸和氨基酸同源性均为99.8%,处在同一遗传进化分支,且与BPV13和BPV2、BPV2-GZ01的亲缘关系较近; Western blotting结果显示,经IPTG诱导的重组表达蛋白能与His单抗产生特异性反应,条带为55.6 kD左右与SDS-PAGE结果一致。研究表明,试验构建了BPV1-GZLZ株L1基因原核表达载体并成功表达。     

Construction and Identification of PRRSV ORF5 Gene Prokaryotic Expression Vector

The present study was designed to construct recombinant plasmids,which could express porcine reproductive and respiratory syndrome virus (PRRSV) ORF5 gene.RNA was extracted from spleen and lung samples of the suspected pigs which were infected with PRRSV.According to PRRSV ORF5 gene,a pair of primers was designed for RT-PCR amplification.The ORF5 target gene was cloned into pMD19-T vector and then the recombinant pMD19-ORF5 was achieved.According to the sequencing results and the characteristics of expression vectors,a pair of primers with NcoⅠand XbaⅠenzyme cleavage sites was designed.Target fragment dORF5 was amplified and then connected to pProEXHTb and pNZ8149 vectors,respectively.And recombinant HTb-dORF5/DE3 and pNZ8149-dORF5/NZ3900 was induced by IPTG and Nisin,respectively,and analyzed by SDS-PAGE and Western blotting.Recombinant HTb-dORF5/DE3 induced by 1.5 mmol/L IPTG was expressed in the highest quantity.There were specific band at about 22 ku with reactionogenicity when it was tested by SDS-PAGE and Western blotting.Recombinant pNZ8149-dORF5/NZ3900 induced by 20 ng/mL Nisin was expressed in the highest quantity.There were specific band at about 19 ku with reactionogenicity when it was tested by SDS-PAGE and Western blotting.The IFA result showed specific green fluorescence.This study successfully constructed recombinant plasmids HTb-dORF5 and pNZ8149-dORF5 and expressed,the result laid a solid foundation for further development of PRRS vaccines.     

水牛骨形态发生蛋白1基因的克隆及其在不同组织中的表达研究

本研究克隆了水牛骨形态发生蛋白1(bone morphogenetic protein-1,BMP1)基因序列,并运用生物信息学方法对其核苷酸序列的保守性和氨基酸的理化性质、蛋白质结构进行了系统分析,此外还运用实时荧光定量PCR技术对BMP1基因在水牛不同组织中的表达差异进行了检测。结果表明,应用RT-PCR技术克隆得到水牛BMP1基因的cDNA序列长度为3 195 bp,其中包含完整的2 967 bp的开放读码框(ORF),编码988个氨基酸。经序列相似性分析显示,水牛BMP1基因与牛、猪、马、人和小鼠相应氨基酸序列相似性分别为99%、96%、96%、96%和95%,具有很强的保守性。结合系统进化树分析结果推测,BMP1基因在不同物种及进化的过程中具有高度的保守性。对水牛BMP1蛋白的结构域预测结果发现,其存在1个信号肽区、1个前肽区、1个金属蛋白酶区、5个CUB区和2个EGF-like功能区。定量表达分析结果显示,BMP1基因在水牛心脏组织中相对表达量最高,睾丸、卵巢和生殖嵴等性腺器官表达量次之,骨头等其他组织表达量较低,肝脏表达量最低。     

Divergence of a strain of Pseudomonas aeruginosa during an outbreak of ovine mastitis

Bacterial infections causing mastitis in sheep can result in severe economic losses for farmers. A large survey of milk samples from ewes with mastitis in Sardinia, Italy, indicated an increasing prevalence of Pseudomonas aeruginosa infections. It has been shown previously that during chronic, biofilm-associated infections P. aeruginosa populations diversify. We report the phenotypic and genomic characterisation of two clonal P. aeruginosa isolates (PSE305 and PSE306) from a mastitis infection outbreak, representing distinct colony morphology variants. In addition to pigment production, PSE305 and PSE306 differed in phenotypic characteristics including biofilm formation, utilisation of various carbon and nitrogen sources, twitching motility. We found higher levels of expression of genes associated with biofilm formation (pelB) and twitching motility (flgD) in PSE305, compared to the biofilm and twitching-defective PSE306. Comparative genomics analysis revealed single nucleotide polymorphisms (SNPs) and minor insertion/deletion variations between PSE305 and PSE306, including a SNP mutation in the pilP gene of PSE306. By introducing a wild-type pilP gene we were able to partially complement the defective twitching motility of PSE306. There were also three larger regions of difference between the two genomes, indicating genomic instability. Hence, we have demonstrated that P. aeruginosa population divergence can occur during an outbreak of mastitis, leading to significant variations in phenotype and genotype, and resembling the behaviour of P. aeruginosa during chronic biofilm-associated infections.     

凋亡素基因真核表达载体的构建及在人肿瘤细胞中的表达

为探讨凋亡素对肿瘤的基因治疗效果,构建了凋亡素基因的真核表达载体。将凋亡素基因重组入 ｐｃ Ｄ Ｎ Ａ３ 载体,并通过脂质体介导凋亡素在人喉癌、人肺癌细胞系中表达;通过 Ｒ Ｔ Ｐ Ｃ Ｒ 在转染细胞中检测到了凋亡素的 ｍ Ｒ Ｎ Ａ,这为应用凋亡素进行肿瘤的基因治疗奠定了基础。     

蚓激酶成熟肽的原核表达及纯化

应用PCR技术扩增获得蚓激酶成熟肽基因，并按相应的阅读框架克隆入表达载体pGEX-4T中谷胱甘肽S-转移酶（GST）的下游。重组质粒转化大肠杆菌Rosetta株，分别在1．0mmol／LIPTG、37℃和0．1mmol／L IPTG、28℃条件下诱导，蚓激酶成熟肽-GST融合蛋白获得了高效表达，且后者得到了可溶性蛋白。经SDS-PAGE证实所表达的融合蛋白产物相对分子量与预期的53kD相符。并且以等电点和亲和层析的方法对表达出的可溶性蛋白进行了纯化。     

柔嫩艾美耳球虫表面抗原SAG2基因的克隆与表达

根据生物信息学预测的基因序列设计引物,应用RT-PCR方法从柔嫩艾美耳球虫第二代裂殖子总RNA扩增获得了鸡柔嫩艾美耳球虫表面抗原2(surface antigen 2,SAG2)基因序列,将其与pGEM-T easy载体连接后转化E.coliDH5α,筛选阳性克隆,以带有限制酶切位点的特异性引物用PCR方法扩增不含SAG2 N端信号肽序列的ORF序列后克隆至表达载体pET-32 a(+),构建了重组表达质粒pET-32 a(+)-SAG2,并将其转化至E.coliBL21(DE3)。经IPTG诱导,获得了SAG2重组抗原在大肠杆菌的高效表达,重组蛋白的表达量约占菌体总蛋白的35%,融合蛋白的分子量约为47 ku。菌体经超声处理后进行SDS-PAGE分析表明,表达蛋白以包涵体的形式存在。     

猪戊型肝炎病毒ORF2片段克隆及原核表达

设计套式引物，内套引物含有BamHⅠ／XhoⅡ酶切位点，采用RT-PCR技术扩增猪戊型肝炎病毒（UEV）结构蛋白基因ORF2部分片段，长度为634bp，将其克隆于PGEX-T载体，测序结果表明插入的片段属于戊型肝炎病毒ORF2部分，将该片段插入PproEXHTb表达载体，经酶切鉴定证实获得了重组表达质粒。将重组质粒转化大肠杆菌BL21，经1mmol／L IPTG诱导，得到表达，表达蛋白分子量为27Ku，以包涵体形式存在。Western blot分析表明，该蛋白可以与猪戊型肝炎阳性血清反应，表明该蛋白具有良好的反应原性。
