The fast growth of the human population forces us to produce more food, but higher crop production also leads to the fast spread of diseases. Plant pathology deploys a wide range of methods that do not provide an adequate solution to all disease losses. In the case of viroids, therapeutic means of control are not available; therefore control strategies are more focused on the development of reliable detection methods to quickly exclude the infected plant material. Although viroids are the smallest and simplest plant pathogens, their identification and detection is not straightforward. Each viroid–host combination is specific, and for reliable identification, all steps from sampling to final detection must be performed accurately. In this review, several methods for viroid detection in various host plants are discussed, including their advantages and disadvantages. Even though relatively new molecular methods enable fast and sensitive detection of viroids, a combination of different methods gives the most reliable identification. Techniques based on nucleic acids may be the future for viroid detection but they still cannot replace biological indexing, which is usually essential in epidemiological and aetiological studies.  相似文献

Scientific interest in geophysical information about land surface temperature (LST) is ever increasing, as such information provides a base for a large number of applications, including environmental and agricultural monitoring. Therefore, the research of LST retrieval has become a hot topic. Recent availability of Landsat-8 satel- lite imagery provides a new data source for LST retrieval. Hence, exploring an adaptive method with reliable ac- curacy seems to be essential. In this study, basing on features of Landsat-8 TIRS thermal infrared channels, we re-calculated parameters in the atmospheric transmittance empirical models of the existing split-window algorithm, and estimated the ground emissivity with the help of the land cover classification map of the study area. Further- more, a split-window algorithm was rebuilt by virtual of the estimation model of the updated atmospheric transmit- tance and the ground emissivity, and then a remote sensing retrieval for the LST of Shihezi city in Xinjiang Uygur autonomous region of Northwest China was conducted on the basis of this modified algorithm. Finally, precision validation of the new model was implemented by using the MODIS LST products. The results showed that the LST retrieval from Landsat-8 TIRS data based on our algorithm has a higher credibility, and the retrieved LST is more consistent with the MODIS LST products. This indicated that the modified algorithm is suitable for retrieving LST with competitive accuracy. With higher resolutions, Landsat-8 TIRS data may provide more accurate observation for LST retrieval.  相似文献

This study examined the production and welfare effects of including a large self‐feeder competent rainbow trout, Oncorhynchus mykiss (Walbaum) (~665 g) in groups of smaller self‐feeder competent conspecifics (~234 g). Costs and benefits were examined for both welfare (aggression, fin damage, condition and mortality) and production (self‐feeder utilization and growth). The 8‐week experiment used six groups of small trout; three treatment groups containing a large trout and three control groups. After 4 weeks the large fish were removed from treatment groups and added to control groups, thus reversing the treatments. Whilst it was thought that the presence of a larger fish would suppress aggression in smaller conspecifics this did not occur. In fact aggression was significantly (= 0.036) higher when large trout were present during the first 4 weeks. No significant differences were found between other welfare indicators, self‐feeder utilization or production parameters. From a production and welfare perspective these results suggest that with the exception of initially increasing aggression larger fish do not represent a significant benefit or risk to smaller conspecifics being cultured in self‐feeder equipped tanks, when all fish are self‐feeder competent.  相似文献

The crucian carp (Carassius carassius, L.) is a cyprinid freshwater species, whose wild‐living populations have decreased in recent years due to an increasing competition of invasive species. Several initiatives were launched, attempting to reintroduce this fish back into its native open waters, which requires the use of crucian carp fry grown under controlled conditions. However, the use of popular commercial diets results in severe developmental abnormalities. The aim of this study was to analyse the impact of feeding natural feed (Chironomidae sp. larvae) or two popular commercial diets on the skeletal development of crucian carp juveniles. Whole‐mount staining and histological methods were used in the study. Skeletal tissues of fish fed both commercial diets were demineralized and displayed a variety of deformities, such as abnormal vertebral spines and vertebral fusion, as well as a new type of deformation, which involved the action of dorsal fin pterygiophores upon the spine. Finally, a novel analytical method was proposed, which utilizes LA‐ICP‐MS for the estimation of bone mineralization. The study proved that basic commercial diets are inadequate for the rearing of crucian carp juveniles.  相似文献

该文探讨了利用近红外光谱分析技术(NIRS)快速测定肉鸡粪便主要肥料成分含量的可行性。在饲养试验过程中采集了肉鸡粪便样品183个,利用常规实验室分析方法测定了其中的总氮(TN)、总磷(TP)、总钾(TK)、铵态氮(AN)、有效磷(EP)含量,利用近红外光谱仪取得了样品在1421.5~2572.2 nm波段的光谱,并使用偏最小二乘法(PLS)结合交互验证(CV)和留一检验法(LOO)建立了肉鸡粪便肥料成分的近红外光谱定标模型,同时利用该模型对20个样品的总氮、总磷、总钾、铵态氮、有效磷的含量进行预测,得出的5种主要肥料成分的预测值和真实值(实验室经典化学分析方法测定值)之间具有显著的相关性,其相关系数分别为0.9574,0.9590,0.9870,0.9572和0.9650。预测标准差分别为0.0014,0.0012,0.0012,2.3041×10-4和0.3814。结果表明,利用近红外光谱法对肉鸡粪便风干样品的主要肥料成分进行快速测定是可行的。  相似文献

Through his own research contributions on the modelling and genetic analysis of quantitative traits and through his former students and postdocs, Robin Thompson has indirectly left a major legacy in human genetics. In this short note, we highlight examples of the long‐lasting relevance and impact of Robin's work in human genetics. A lone early study of marker‐assisted selection developed many of the tools and approaches later exploited (often after reinvention) by the human genetics community in GWAS studies and for prediction. Furthermore, a particularly clear example of the pervasive impact of Robin's work is that REML has become the default method to estimate variance components and that genetic predictions exploiting linkage disequilibrium in the population are starting to become used in precision medicine applications.  相似文献

Screening for newer bioactive compounds from microbial metabolites resulted in the isolation of a novel antibiotic from the culture filtrate, Streptomyces sp 201, of a soil. The bioactive compound, with antifungal and antibacterial activity, was identified as 2-methylheptyl isonicotinate. The antifungal activity of live culture, culture broth and the isolated bioactive compound showed marked inhibition against dominant soil-borne phytopathogens such as Fusarium oxysporum Schlect, F moniliforme Sheldon, F semitectum Berkeley & Ravenel, F solani (Martius) Sacc and Rhizoctonia solani Kuehn. The compound had no effect on seed germination and seedling development as displayed by root and stem growth of the test plant species. In pot experiments with seedlings of cruciferous plants such as Raphanus sativus L (radish), Brassica campestris L (yellow mustard), Brassica oleracea var botrytis L (cauliflower), the antibiotic compound showed promising protective activity of 92% when seeds of the test plants were treated at a dose of 50 micrograms ml-1 prior to sowing. Seed treatment with a spore suspension (3 x 10(8) spores ml-1) of the Streptomyces sp 201 displayed protective activity in the range of 56-60%. Seeds coated with 2.5% methyl cellulose-amended spores of the antagonist showed protective activity in the range of 64-72%. Further, seed treatment with the culture filtrate of the antagonist also showed promising protective activity in the range of 64-84%.  相似文献

Background, aim, and scope  Vegetation stress diagnoses based on plant sampling and physiochemical analysis using traditional methods are commonly time-consuming, destructive and expensive. The measurement of field spectral reflectance is one basis of airborne or spaceborne remote sensing monitoring. Materials and methods  In this study, paddy plants were grown in the barrels evenly filled with 10.0 kg soil that was mixed respectively with 0, 2.5 × 207.2 and 5.0 × 207.2 mg Pb per 1,000 g soil. Rice canopy spectra were gathered by mobile hyper-spectral radiometer (ASD FieldSpec Pro FR, USA). Meanwhile, canopy leaves in the field-of-view (FOV) of spectroradiometer were collected and then prepared in the laboratory, (1) for chlorophyll measurement by Model 721 spectrophotometer, and (2) for Pb determination by atomic absorption spectrophotometer (SpectraAA-220FS). Results and discussion  Canopy spectral reflectance in the region of visible-to-near-infrared light (VNIR) increased, because ascended Pb concentration caused the decrease of canopy chlorophyll content. In the agro-ecosystem, however, heavy metal contamination is presented typically as mixture and their interactions strongly affect actually occurring effects. Normalized spectral absorption depth (D n), and shifting distance (DS) of red edge position (REPs) revealed the differences in Pb concentration for canopy leaves, especially at the early tillering stage. Due to insufficient biomass of rice plants, the 30th day was not reliable enough for the selection of crucial growth stages. Some special sensitive bands might be omitted at the same time because of limited sample sets. Conclusions  Our initial experiments are still too few in the amounts of both metals and plants neither to build accurate prediction models nor to discuss the transformation from ground to air/spaceborne remote sensing. However, we are pleased to communicate that ground remote sensing measurements would provide reliable information for the estimation of Pb concentration in rice plants at the early tillering stage when proper features (such as DS and D n) of reflectance spectra are applied. Recommendations and perspectives  Hyper-spectral remote sensing is a potential and promising technology for monitoring environmental stresses on agricultural vegetation. Further ground remote sensing experiments are necessary to evaluate the possibility of hyper-spectral reflectance spectroscopy in monitoring different kinds of metals’ stress on various plants.  相似文献
