牻牛儿基牻牛儿基焦磷酸合酶(Geranylgeranyl diphosphate synthase,GGPS)是萜类合成途径的结构酶,对植物生长发育具有重要意义。本研究通过RACE和RT-PCR方法克隆得到5条潜在的茶树GGPS序列,分别命名为CsGGPS1-4和CsGGPS9,其中CsGGPS9存在3条等位基因,分别是CsGGPS9-1、CsGGPS9-2和CsGGPS9-3,在系统进化树上与其他基因分成两支。蛋白质序列分析表明,茶树GGPS家族成员都具有polyprenyl_synt结构域,不存在信号肽序列。亚细胞定位预测结果显示,CsGGPS1、CsGGPS2和CsGGPS4定位在叶绿体上,CsGGPS3和CsGGPS9定位在线粒体上。通过Swiss Model进行三维建模,结合"three-floor"模型对茶树GGPS家族成员的功能进行预测,预测结果显示,CsGGPS1、CsGGPS2和CsGGPS4是GGPS;CsGGPS3是异源二聚体形式的牻牛儿基焦磷酸合酶的小亚基;CsGGPS9的催化主产物是碳链数大于30的异戊烯基焦磷酸。q RT-PCR分析表明,CsGGPS1整体表达丰度较低,仅在一芽二叶中表达量稍高;CsGGPS2在茶树各个组织中均有表达,在花中表达量最高,且花发育过程中表达量先上升后下降;CsGGPS3在叶和幼根中的表达量高于花,花发育过程中表达平稳;CsGGPS4在茶树各个组织中表达量数值相近,在花发育过程中表达量变化趋势与CsGGPS2相同;CsGGPS9的表达量在成熟叶中显著低于幼嫩叶片。  相似文献

根据目标性状有的放矢的选配杂交亲本是提高优异品质组成品种的选择效率的基本前提。本研究对东北三省102份大豆种质资源的蛋白、氨基酸组分、油份及脂肪酸组分进行测定,通过遗传多样性、主成分和聚类分析,对其进行表型鉴定及基因型分类以综合评价种质品质特性。结果表明:东北三省大豆种质油份及脂肪酸组分变异较丰富,遗传多样性程度较高。根据主成分分析筛选到9个主成分进行聚类分析,通过聚类分析将供试种质资源分为5类。第I类群蛋白含量较高、油份含量偏低,第II类群蛋白、油份含量均居中,第III类群油份含量较高、蛋白含量偏低,第IV类群高油,第V类群高蛋白,类群间的氨基酸、脂肪酸组分各有差异。需根据育种目标在群体间选配亲本,以提高品质育种的效率。  相似文献

为优化微波提取黑豆总黄酮化合物的工艺,并对其抗氧化性进行初步研究,在单因素试验的基础上,选取液料比、乙醇体积分数和微波功率为自变量,总黄酮得率为响应值的三因素三水平的响应曲面分析法,模拟得到二次多项式回归方程的预测模型。结果表明:提取工艺的最佳条件为功率600 W、液料比40∶1、乙醇体积分数60%,在此条件下,总黄酮得率达到22.1 mg·g~(-1)。抑制油脂过氧化作用的研究结果表明黑豆黄酮是一种天然的抗氧化剂。  相似文献

为明确主要气象因子对黑龙江省水稻主产区水稻鞘腐病发生的影响,从2007年开始连续10年进行定点调查,收集水稻鞘腐病发生时期的气象因子及病情指数,采用通径分析及逐步回归的方法,对影响水稻鞘腐病病情指数的气象因子进行了研究。结果表明,8月份的平均温度是对水稻鞘腐病影响最大的气象因子,其次为8月份的平均降雨量和9月份的平均日照时数。试验结果为适期施药防治水稻鞘腐病奠定了基础。  相似文献

为探讨抗旱节水小麦的定向培育方法,对抗旱节水型衡麦系列品种选育过程进行了总结和分析。多年来河北省农林科学院旱作农业研究所围绕"节水抗旱、抗逆高产、广适稳产、优质早熟"的育种目标,着眼解决地下水资源极度匮乏与农业生产矛盾突出的问题,为培育抗旱节水型广适小麦品种,利用常规育种技术与分子标记检测手段,广泛筛选遗传背景远缘且生态差异较大的优异亲本组配组合,创新采用不同世代水旱交替双向综合选择技术,先后育成16个抗旱节水型小麦品种,提高了选择效率和水资源利用率。在系谱上,衡观35等为代表的衡麦系列品种与蚂蚱麦、碧玉麦和洛夫林10号存在一定的血缘关系,且含有对光周期不敏感的Ppd-Dla等优异基因。这些品种目前已由冀中南冬麦区推广至黄淮冬麦区(南、北片)、北部冬麦区(天津)和长江中下游冬麦区(湖北襄阳)等区域种植,展示出生态广适、节水稳产的特点。  相似文献

Understanding the genetics of resistance to spot blotch disease, caused by Bipolaris sorokiniana (Sacc.) Shoem, is important to design an appropriate breeding strategy to improve the trait. The objective of this study was to determine the gene action and mode of inheritance of resistance to spot blotch in wheat. Eight genotypes with varying resistance to the disease were crossed in a full diallel mating design. Parents and their progenies were evaluated for spot blotch resistance. Data were analyzed using Hayman’s diallel analysis. The results suggested the importance of additive gene effects in controlling the resistance to spot blotch in the materials under study. No epistasis, maternal, or reciprocal effects were detected. Resistance to spot blotch exhibited partial dominance. Therefore, exercising selection for resistance in the early segregating generation should be an effective approach because of the predominance of additive gene effects. The Wr/Vr graph showed that the parents 30SAWSN5 (P3) and Coucal (P4) possessed more dominant genes, which makes them particularly suitable for inclusion in breeding for resistance to spot blotch.  相似文献

This study was carried out to better understand non-adoption of improved varieties of potato (Solanum tuberosum) and associated technologies by smallholder farmers in Chencha, Ethiopia. Data were collected through a survey (n = 47) and in-depth interviews (n = 20). It shows how wealth status was a factor of major importance. Most wealthy and some medium-wealthy farmers adopted improved potato varieties and many of the improved production practices; they had access to seed, associated knowledge and support, and sufficient resources that were necessary to apply the improved practices. Non-adoption was common among many medium-wealthy and most poor farmers: they lacked—next to access to the technologies and knowledge—cash, land, and labor. Results indicated the need to rethink research and intervention efforts. Next to paying attention to differences in the access to technology and the related knowledge, there is a need to consider the variation in technology needs, supporting microcredit services, and room to experiment. As a result, different combinations of improved production practices may be adopted.  相似文献

Soybean lodging can result in serious yield reduction. Detecting the quantitative trait loci (QTL) associated with lodging tolerance for their further application in marker‐assisted selection (MAS) has the potential to enhance soybean breeding efficiency. In this study, a genome‐wide association analysis (GWAS) was performed to identify soybean accessions that could potentially be used to produce lodging‐tolerant varieties, based on the comprehensive evaluation of lodging scores (LS) obtained for the parental cultivar “Tokachi nagaha” and its 137 derived cultivars. Results showed that genotype, environment and genotype × environment interaction significantly influenced LS. Of the 31 significant SNPs identified, 22 were consistently detected in two or more environments and 27 SNPs were located in or close to agronomically important QTL mapped by linkage analysis. Best linear unbiased predictors (BLUPs) of LS tend to decrease with the elite alleles contained by accessions increasing. Some excellent accessions, with lower BLUPs and Di (stability coefficients) values and more elite alleles, were selected. This study contributed to understand the genetic mechanism of lodging, providing genetic and phenotypic information for MAS.  相似文献

Plant height is closely related to seed yield of soybean. The goal of this study was to identify important loci affecting soybean plant height using meta‐analysis based on a reference physical map. Plant height related to QTLs was mapped across eight years with a RIL population by WinQTLCart v2.5. 182 QTLs related to plant height of soybean from database and our research were collected, and each QTL was projected onto the soybean physical map by software BioMercator v2.1. The confidence interval of meta‐QTL ranged from 0.09 to 5.07 Mb, and the mean phenotypic variance ranged from 4.9% to 73.0%. Furthermore, 4,259 candidate genes were located in these consensus QTLs, and 40 of them were involved in the plant growth and stem elongation and annotated as plant hormone signal transduction (pathway ID ko04075) in KEGG pathway. These results would lay a foundation for fine mapping of QTLs/genes related to plant height and marker‐assisted selection for breeding in soybean.  相似文献

Striga hermonthica is a major constraint to sorghum production and productivity in sub‐Saharan Africa, Ethiopia, in particular. The objectives of this study were to evaluate the performance of 49 sorghum genotypes based on their reaction to S. hermonthica and investigate the relationships among yield and Striga resistance traits, including the Striga emergence count, area under Striga number progress curve, area under Striga severity progress curve, grain yield under infested, grain yield under non‐infested conditions and relative yield loss. The genotypes differed significantly in all measured parameters under Striga‐infested and non‐infested conditions. The genotypes ‘Birhan’, ‘Gubiye’, Wolegie, Zegerie, Nechmashila I, Woftel, Tetron and Eyssa were identified as promising ones based on grain yield and Striga‐related traits. On the other hand, the genotypes Jamyo, Bobie, Gedido, Mankebar and Zengada had moderate Striga numbers with low relative yield loss as compared with susceptible checks. The most promising variety, Zegerie, Mankebar and Zengada, out yielded the standard resistant checks ‘Birhan’ and ‘Gubiye’ under Striga infestation. Ward cluster analysis grouped the 49 sorghum genotypes in to four distinct clusters under Striga‐infested conditions. All members of clusters II and III showed the highest yielding group with the lowest to moderate Striga number, while cluster IV supported the lowest yield with the highest Striga number. Significant negative correlations were observed between yield‐ and Striga‐related traits. Highly significant and strong correlations were observed among Striga resistance indices, indicating that any of the Striga resistance parameters can be used as an indicator of resistance. The wide variations in grain yield among genotypes under Striga‐infested conditions would be invaluable genetic resources for production in Striga endemic areas of Ethiopia.  相似文献
