Radiocesium is an extremely harmful radionuclide because of its long half-life; it is important to reduce its transfer from contaminated soil into crops. Here we surveyed genetic variation for seed cesium (Cs) concentration in soybean mini-core collections representing large genetic diversity. The collections grown over 3 years in rotational paddy fields exhibited varying seed Cs concentrations with significant year-to-year correlations, although the phenotypic stability of Cs concentration was lower than that of the congeners potassium (K) and rubidium (Rb). Although Cs is supposedly accumulated in plants via the K transport system, there was no apparent relationship between Cs and K concentrations, whereas a clear positive correlation was observed between Cs and Rb concentrations. Cs and K concentrations in seed showed slightly positive and negative correlations, respectively, with days to flowering. We selected several high or low Cs accumulator candidates on the basis of the 3 years of seed concentration data. These two groups showed significantly different seed Cs concentrations in another field. The differences could not be explained by flowering time alone. These results suggest that genetic variation for seed Cs concentration is present in soybean germplasm and would be useful for breeding low Cs-accumulating varieties.  相似文献

本研究利用L16(54)正交试验设计研究了影响冬瓜SSR-PCR的五个因子(10×Buffer(含Mg2+),d NTP,DNA,引物,r Taq),研究结果表明Buffer(含Mg2+)和r Taq对PCR影响最明显,最终筛选出最佳反应体系:10×Buffer 2.0μL(1×Buffer),d NTP 2.0μL(200μmol/μL),DNA 0.6μL(6 ng/μL),引物0.4μL(0.2μmol/L)。体系体积为20μL,循环数为35。引物筛选程序为:94℃预变性3 min,94℃变性45 s,58℃~68℃复性30 s,5个循环,每个循环退火温度降2℃,72℃延伸1 min,94℃变性30 s,50℃~58℃复性1 min,8个循环,每个循环退火温度降低1℃,72℃延伸1 min,94℃变性30 s,50℃复性30 s,72℃延伸1 min,共30个循环,72℃延伸7 min。本研究建立了适合冬瓜的SSR反应体系和引物多态性筛选程序。  相似文献

Summary The establishment of representative core collections and back-up reserve collections was proposed to facilitate effective management and to promote utilization of large base collections. The priorities of the developing national PGR programmes, in the organization of components of their base collections, are different from those of IARCs since the strengths as well as limitations of the two systems vary. Unlike most IARCs, national programmes have the option of networking their active collections, maintained at several eco-sites, and linking this network to the base collection kept under long-term storage. This keeps open the option to develop situation-specific subsets for an effective germplasm utilization. There is a pressing need for a scientific re-examination of the concept of germplasm core in order better to apply it in developing core subsets in the national PGR programmes. The formulation of situation-specific subsets is advocated, as the system would be directed to users' requirements or addressed to gene bank managers' resource constraints. Arguments given in favour of developing core subsets, rather than a single core are: (i) logical; (ii) population genetic; and (iii) germplasm usage considerations. Summary The use of random amplified DNA fragments as genetic markers in Coffea was investigated. Arbitrary oligonucleotides were used as primers to amplify genomic DNA of different coffee accessions representing major Coffea species by polymerase chain reaction. Intraspecific variation was easily detected in C. canephora and C. liberica whereas the primers assayed failed to reveal polymorphism between C. arabica accessions. Extensive interspecific variation was observed. Genetic relationships between Coffea species are deduced from the degrees of similarity in amplified product profiles. Random amplified DNA markers appeared to be of high value for characterization, analysis and utilization of coffee genetic resources.  相似文献

Genetic diversity of seven Rhus L. species was assessed using random amplified polymorphic DNA (RAPDs) markers. Initially, 90 primers were screened, of which 25 produced reproducible amplification products. These primers generated a total of 296 bands, with an average of 11.8 bands per primer. Out of 296 bands scored, 236 (80%) were polymorphic and 62 (20%) were monomorphic. Primers OPC-05 and OPD-05 generated 100% polymorphic bands. The resolving power of primers ranged from 9.4 to 26.8. Similarity matrix values ranged from 0.45 to 0.63. The dendrogram generated using Unweighted Pair Group Method using Arithmetic Averages (UPGMA) grouped all the species of Rhus in one major group with two sister groups, whilst R. pyroides Burch. and R. dentata Thunb. were outliers. R. gerrardii (Harv. ex Engl.) Diels, R. glauca Thunb. and R. pentheri Zahlbr. constituted one sister group, while R. natalensis Bernh. ex C. Krauss and R. gueinzii Sond. were included in the other. The degree of genetic diversity observed between seven species of Rhus with RAPD markers suggest that this approach could be used for studying the phylogeny of the genus.  相似文献

Gap formation is suggested as an alternative forest management approach to avoid extreme changes in the N cycle of forest ecosystems caused by traditional management practises. The present study aimed to investigate the effect of gap formation on N availability in beech litter and mineral soil on sites, which experienced only little soil disturbance during tree harvest. N pools, litter decomposition, and N mineralization rates in mineral soil were studied in two gaps (17 and 30 m in diameter) in a 75-year-old managed European beech (Fagus sylvatica L.) forest in Denmark and related to soil temperature (5 cm depth) and soil moisture (15 cm depth). Investigations were carried out during the first 2 years after gap formation in measurement plots located along the north-south transect running through the centre of each gap and into the surrounding forest.An effect of gap size was found only for soil temperatures and litter mass loss: soil temperatures were significantly increased in the northern part of the large gap during the first year after gap formation, and litter mass loss was significantly higher in the smaller gap. All other parameters investigated revealed no effect of gap size. Nitrification, net mineralization, and soil N concentrations tended to be increased in the gaps. Cumulative rates of net mineralization were two fold higher in the gaps during the growing season (June-October), but a statistically significant increase was found only for soil NH4-N concentrations during this period. Forest floor parameters (C:N ratios, mass loss, N release) were not significantly modified during the first year after gap formation, neither were the total C content nor the C:N ratio in mineral soil at 0-10 cm depth.  相似文献
