应用DNA—荧光染料法评定瘤胃细菌细胞计数的研究   总被引:1,自引:0,他引:1  
本研究应用瘤胃Succinogenes85细菌 ,用DNA荧光染料法探讨一种新的、简便而快速、无需厌氧操作的方法来评定瘤胃细菌计数。结果表明 ,加进DNA染料后 ,用超声波裂解细胞的时间不能低于30分钟 ,并且裂解后的样品在室温下平衡90分钟 ,对细菌的DNA含量没有影响。应用DNA荧光染料法测定的瘤胃细菌数与用细胞流动器测得的数值相比 ,从统计分析来看 ,没有显著差异(P<0.01)。  相似文献

分别采用组织块法和消化法2种接种方法进行银狐垂体细胞系构建研究。在消化法中使用不同浓度的胰蛋白酶来检验效果,同时在原代培养期使用反复差速贴壁法纯化,传代培养期结合使用反复差速贴壁以及两步消化法进行处理。结果表明:组织块法接种后成纤维细胞生长明显,原代培养后期成纤维细胞已为主要细胞;降低胰蛋白酶浓度能降低对细胞损伤,但消化时间会增长;采用消化法接种在抑制成纤维细胞生长方面效果好于组织块法。纯化效果上,结合反复差速贴壁法与两步消化法处理后获得了较好的效果,在原代培养时期能较好去除成纤维细胞并通过持续使用该方法可在传二代后获得较高纯度的细胞,纯度能达到80%以上,能够建立细胞系。  相似文献

The effectiveness of feeding barriers to protect seedlings against damage caused by pine weevil (Hylobius abietis) was evaluated in experiments established from 1995 to 1999 in southern Sweden. For this purpose the damage done to containerized seedlings protected by various types of barrier was evaluated, and compared with damage to untreated and insecticide (permethrin)-treated seedlings. Three types of feeding barrier that gave significant protection during the first 3 yrs were recognized: coating on the stem bark, shields with a collar at the top, and shields without a collar. The mean accumulated mortality caused by pine weevil for coatings (47%) and shields without a collar (59%) was significantly higher than the corresponding figure for permethrin treatment (13%), whereas shields with a collar (26%) did not differ significantly from the permethrin treatment in this respect. The highest mortality was found for untreated controls (87%).  相似文献

We histologically examined lectin binding patterns in the olfactory bulb of mallard ducks (Anas platyrhynchos) using 21 biotinylated lectins. Positive staining for the N‐acetylglucosamine‐specific lectins (Bandeiraea simplicifolia II, Datura stramonium, Lycopersicon esculentum, Solanum tuberosum, Triticum vulgare), galactose or N‐acetylgalactosamine‐specific lectins (Artocarpus intergrifolia, Phaseolus vulgaris erythroagglutinin, Phaseolus vulgaris leucoagglutinin, Ricus communis) and the mannose‐specific lectins (Lens culinaris and Pisum sativum) was observed in the olfactory nerve and glomerular layers. Canavalia ensiformis staining showed a similar pattern to that obtained with the lectins and there was also faint staining in the mitral cells. Olfactory nerve axons terminate in the glomeruli, where they make excitatory synapses with the dendrites of mitral cells. This finding indicates that glycoconjugates that bind Canavalia ensiformis play an important role in formation of glomeruli. No positive staining for the other lectins was seen in the olfactory bulb. Based on these results, we conclude that cell surface sugar moieties of the olfactory bulb in mallard ducks express N‐acetylglucosamine and mannose residues rather than N‐acetylgalactosamine residues. The carbohydrate composition of mallard duck olfactory bulb differed from that of other vertebrates found in previous studies.  相似文献

 本文以白菜幼苗为材料,研究了芸苔链格孢菌毒素(AB-毒素)对白菜叶片细胞膜透性及叶片内SOD酶、POD酶活性变化的影响。结果表明,白菜叶片用AB-毒素处理后,细胞膜透性增大,且随处理时间的延长和毒素浓度的增加而增高。不同白菜品种用毒素处理后膜透性的改变与病菌接种的发病程度呈正相关,即电导率大的白菜品种,叶片发病也重。试验结果还发现,芸苔链格孢菌及其毒素处理白菜后,体内SOD酶和POD酶活性的变化基本趋于一致,即处理后12 h内急剧下降,然后(12~48 h)活性上升。而对照处理基本变化不大。可见,芸苔链格孢菌毒素表现了与其病原菌一致的致病作用。  相似文献

The study was aimed to explore the antioxident function of sulforaphane (SFN) on the leydig cells of cadmium (Cd) exposured mice.Cd and SFN were added to the cell culture medium of TM3 cell, half inhibitory concentration (IC50) of Cd and SFN safe dose range were determined.The tested model was set up,and the relative survival rate of TM3 cells, lactate dehydrogenase (LDH) activity and cell antioxidant levels were determined to study the antioxident function of SFN to cadmium exposured mice.The results showed that:①With the increase of Cd concentration,the relative survival rate of TM3 cells was decreased,and the IC50 of Cd was 51.4 μmol/L;Within a certain concentration range,SFN could increased the cells survive rate,but there was toxicity when the SFN concentration was more than the scope,and the greater the concentration,the greater the toxicity. At experimental condition,SFN safety concentration were 2.5,5 and 10 μmol/L.②Compared with the control group,the GSH content,the T-SOD and GSH-Px activities in Cd group were significantly or extremely significantly decreased (P < 0.05;P < 0.01),and the LDH activity,the MDA content were increased.Additionally,the LDH activity and MDA content of SFN groups were decreased,while others three indexes were increased. Compared with Cd group,the GSH content,the T-SOD and GSH-Px activities of Cd+SFN groups were significantly or extremely significantly increased (P < 0.05;P < 0.01),however,the LDH activity,MDA content were decreased. The result indicated that SFN had the antagonism effect on toxicity of Cd in TM3 cell.  相似文献

Elevated ultraviolet‐B (UV‐B; between 290 and 320 nm) radiation, because of depletion of the stratospheric ozone layer, is one of the major environmental factors influencing plant metabolic processes and yield. The southern US rice cultivars contribute greatly towards US rice production, but the effects of elevated UV‐B radiation on these cultivars are not well known. The objective of this study was to determine the effects of elevated UV‐B radiation on leaf photosynthetic rate (Pn), membrane stability, pollen viability, phenolic concentration and yield of eight commercially popular southern US rice cultivars (five inbred cultivars and three hybrids). Plants were grown in a temperature‐controlled greenhouse in Beaumont, TX, USA, and were exposed to UV‐B radiation of 0, 8 or 16 kJ m?2 day?1 for 90 days. For most of the cultivars, plants grown under 8 or 16 kJ UV‐B radiation showed significant decreases in Pn, membrane stability, pollen viability, and yield compared with the plants grown under an UV‐B‐free environment, whereas there was a significant increase in leaf phenolic concentration under 16 kJ UV‐B radiation. The hybrid ‘Clearfield XL729’ performed best among the selected southern US rice cultivars under 16 kJ UV‐B radiation.  相似文献
