In vitro culturing of plant cells can cause changes in karyotype. Chromosome variations following long-term propagation in suspension culture of Triticum timopheevii (Zhuk.) Zhuk. were studied by routine staining and C-banding. The culture was highly heterogeneous with respect to the number and structure of chromosomes. The modal class cells had a lower chromosome number than T. timopheevii (2n= 28). This data was confirmed by cytophotometric analysis of nuclear-DNA content. Frequencies of chromosome loss varied for different homoeologous groups. At genome chromosomes tended to be preferentially eliminated in cells of different ploidy levels. Deletions, insertions, translocations, telocentric chromosomes, isochromosomes and dicentrics and their derivatives were observed in cultured cells. Chromosomes of various homoeologous groups differed in the frequencies and spectra of re—arrangements, but most aberrations occurred in the G-genome chromosomes. In vitro chromosome modifications did not correspond to in vivo variation. Presumably, this difference was caused by differences in the mechanisms of adaptation to the environment at the levels of the cell and the whole organism. G-genome chromosomes were more frequently involved in this process, both in vivo and in vitro.  相似文献

The objective of this study was to correlate root length of wheat (Triticum aestivum L.) genotypes grown in Al-containing, acidic hydroponic solutions, with root weights from acid-soil experiments and field scores from Brazilian acid-field trials. A total of 43 wheat genotypes, primarily from Brazil, were evaluated by growing seedlings for 4 days in hydroponic solutions containing 0.0–4.0 mg/l Al. The root growth rate of all the genotypes was reduced with the addition of Al to the solution and the Al-sensitive and Al-tolerant wheat genotypes were clearly identified. Genotypes with intermediate Al-tolerance levels showed variable root lengths in response to Al stress. Correlations between root length or a root tolerance index (RTI) in the Al solutions versus acid-soil experiments and acid-field trials were highly significant (r = 0.71–0.85, P < 0.01). The most significant correlation was observed among seedlings grown in 1 mg/1 Al. This study presents evidence that this short duration and simple screening technique provides a highly significant correlation with previous acid-soil Al-tolerance evaluations. Furthermore, the data obtained suggest that hydroponic screening of wheat seedlings for Al tolerance may be used in breeding programmes or in screening germplasm collections.  相似文献

Summary A reciprocal F1 monosomic analysis of chromosomal differences between Spica and Bersée was carried out under controlled environment conditions. Chromosomes associated with differences in days to ear emergence, number of leaves and number of spikelets were identified. The results indicated that chromosome 2B of Spica carries a photoperiod insensitivity allele at the Ppd 2 locus. Both Spica and Bersée appear to have a vernalization insensitity allele at the Vrn 2 locus on chromosome 5B. On chromosome 3A, 4B, 4D and 6B factors were found with major effects on earliness per se, diffeences in ear emergence and number of spikelets which were independent of photoperiod and vernalization. The possibility that these factors influence growth rate is discussed.  相似文献

Fifty-five spring bread wheat (Triticum aestivum L.) cultivars, mostly released between 1975 and 1991 in eight leaf rust-prone spring wheat growing regions of the former USSR, were tested in the seedling growth stage for reaction to 15 Mexican pathotypes of Puccinia recondita f. sp. tritici. In total, seven known and at least two unknown genes were identified, either singly or in combinations: Lr3 (7 cultivars), Lr10 (14), Lr13 (5), Lr14a (1), Lr16 (1), Lr23 (3); the unknown genes were identified in 14 cultivars. The first unknown gene could be either Lr9, Lr19, or Lr25; however, the second unknown gene in 9 cultivars was different from any named gene. Twelve of the 15 pathotypes are virulent for this gene, hence its use in breeding for resistance will be limited. The cultivars were also evaluated at two field locations in Mexico with two pathotypes in separate experiments. The area under the disease progress curve and the final disease rating of the cultivars indicated genetic diversity for genes conferring adult plant resistance. based on the symptoms of the leaf tip necrosis in adult plants, resistance gene Lr34 could be present in at least 20 cultivars. More than half of the cultivars carry high to moderate levels of adult plant resistance and were distributed in each region.  相似文献

Summary A study was undertaken to evaluate the breeding behaviour and to identify a spontaneously produced putative chromosomal deletion in the winter wheat (Triticum aestivum L. em. Thell.) cv Norstar. Male and female transmission studies of plants heterozygous for the deletion chromosome indicated 9.5% and 48.8% transmission through the pollen and the egg, respectively. Meiotic analyses of progeny from deletion heterozygotes indicated that the deletion chromosome was eliminated from half of the plants, which agreed with the male and female transmission frequencies. Testcrosses of the deletion chromosome with telocentrics and nullisomic-tetrasomic combinations suggested that the deletion involved the long arm of chromosome 5D. This was confirmed by restriction fragment length polymorphism (RFLP) analysis. Also, monosomic plants obtained in progeny of deletion heterozygotes were shown to be monosomic for 5D. Studies of plants homozygous for the deletion showed relatively normal pairing between the deletion chromosomes, and with the short arm (5DS), but not the long arm (5DL). Deletion homozygotes were self-sterile, and morphologically similar to plants nullisomic for 5D, but plants that also contained 5DL, or a homoeologous chromosome were self-fertile and had normal morphology. Studies of chromosome morphology indicated that the deletion chromosome was metacentric, and the length of the long arm was reduced by approximately 60%. RFLP studies showed that, in terms of genetic distance, 90% of the arm was missing.  相似文献

Summary Somatic embryogenesis was initiated from immature embryos on Murashige-Skoog (MS) medium plus 2 mg.l-1 2,4-dichlorophenoxyacetic acid, 2% sucrose and 0.6% agarose. Somatic embryos were isolated and regenerated into whole green plants on MS medium devoid of 2,4-D. These regenerants were previously demonstrated to differ in their mitochondrial DNA organization. In order to estimate their characteristics three progenies of short-term culture regenerants and three progenies of long-term culture regenerants were analyzed and compared to the parental line. These somaclones obtained from the wheat variety Chinese Spring were evaluated for variation of 13 agronomic and morphological quantitative characters in comparison to the parental line. Significant variation was observed for plant height, spike length, main tiller diameter, between the somaclones regenerated from long-term culture and their parent. Differences were observed to increase with the duration of culture, leading to a significant modification of the structure of the plants. Several changes occurred during the somatic tissue cultures, but to a lesser extent than has previously been described in the literature.  相似文献

The objective of this paper was to study the differences between some prolamin variants coded at the loci Glu-3/Gli-1, Glu-1 and Gli-A2 and their relative roles in durum-wheat quality. F3 lines from four durum wheat crosses (‘Abadia’בMexicali’. ‘Oscar’בArdente’, ‘Oscar × Mexicali’ and ‘Alaga’בC. of Balazote’) were analysed for gliadin and glutenin composition by electrophoresis. Whole-grain-derived samples were analysed for SDS sedimentation (SDSS) value, mixing properties, and contents of protein and vitreousness. The glutenin patterns LMW-2. LMW-2? and LMW-2 (CB) at Glu-B3/Gli-B1 were associated with better gluten quality than were LMW-1 and LMW-2*. The glutenin subunits LMW4 and LMW3 + 15 at Glu-A3/Gli-A1 and HMW-1 showed better mixing properties than LMW7 + 12, LMW5 and the null phenotype. respectively. The HMW glutenin subunits 20 + 8 at Glu-B1 showed a negative association with gluten quality, but the rest of the HMW glutenin subunits and α-gliadins did not show any influence on gluten quality. Correlations between the results of the SDSS test and the mixograph were highly significant, but no correlation was found between these results and protein and vitreousness contents. The results are discussed in relation to the development of durum wheat varieties with improved qualities.  相似文献

The mode of inheritance and allelic relationships among genes conferring resistance to Karnal bunt were studied in seven bread-wheat (six resistant and one susceptible) genotypes. The resistant genotypes originated in China (‘Shanghai#8’), Brazil (PF71131), the USA (‘Chris’), and Mexico (‘Amsel’, CMH77.308 and ‘Pigeon’). The susceptible line WL711 was from India. Evaluation of these wheat lines and all possible crosses among their F1 and F3 generations (about 100 progenies in each cross) revealed that two partially recessive genes conferred the resistance to Karnal bunt in ‘Pigeon’, whereas four partially dominant genes were present in the other genotypes. ‘Chris’, ‘Amsel’ and PF71131 carry one gene, whereas ‘Shanghai#8’ and CMH77.308 have two genes. ‘Chris’, ‘Amsel’, and PF71131 have different genes, whereas one gene was common to PF71131, CMH77.308 and ‘Shanghai#8’, and another to ‘Chris’ and CMH77.308. Gene symbols were formally designated to the resistant stocks. Resistance was incomplete and stable.  相似文献

Summary Populations of wild emmer wheat, Triticum dicoccoides, in Israel, originating from diverse habitats, and tested earlier for allozyme (Nevo et al., 1982) and disease resistance polymorphisms (Moseman et al., 1983a, 1983b; Nevo et al., 1984a, 1984b), were compared and contrasted for performance in agronomically important phenotypic traits. The traits compared involved 10 variables comparing germination, earliness, biomass and yield variables. The field experiments were conducted in 1980, 1981 and 1982 in two relatively standardized and contrasting environments: mesic (Mount Carmel, Haifa) and xeric (Acedat Farm, and Sede Boqer, in the northern Negev desert). The experimental design involved 12 population quadrangles at Avedat Farm in 1980, and rows of randomized genotypes of five populations in both Haifa and Sede Boqer in 1981 and 1982.The results indicate that the characters studied are partly genetically determined. Striking genetic variation was found between and, at least in some characters, also within populations in each site, whereas remarkable environmental variation including genetic-environmental interaction was found between the mesic and the xeric sites. We conclude that natural populations of wild emmer wheat in Israel vary not only in genetic polymorphisms of allozymes and disease resistance, but also in quantitative traits of agronomic importance. These traits are economically significant and should be conserved and utilized in wheat crop improvement.  相似文献

Summary Genetic and cytogenetic studies were done on a male sterile mutant of the wheat variety Probus. Association of the 4A chromosome carrying the ms gene was studied in the F1 of the male sterile Probus with Chinese Spring ditelo 4AS, with Transec and with line T4AS-DRS respectively. The presumption that the genetic male sterility of the mutant was due to a terminal deletion of the short arm of chromosome 4A could be confirmed.Linkage studies showed that the ms gene was at 17 map units from the dwarfing gene (Rht3) of Minister dwarf. This allows selection of short male sterile plants at the seedling stage.  相似文献
