Summary The effect of genotype, growing conditions for donor plants and type and concentration of carbohydrate in the culture medium was investigated for anther culture of head cabbage (white cabbage, savoy cabbage, pointed-headed cabbage). Strong genotypic effects on embryo formation from the cultured anthers were shown as well as superior embryo formation from anthers of field grown donor plants compared to plants grown in the greenhouse. When comparing 7, 10 and 13% sucrose in the medium, embryo response increased with increasing sucrose concentration. With maltose, which was generally inferior to sucrose as carbohydrate source for anther culture, the embryo response did not increase with maltose concentration above 10 per cent.  相似文献

Summary A spring wheat genotype which produces somatic embryos in vitro, after short and long-term culture, was tested for its ability to sexually transmit this embryogenic trait. Reciprocal crosses were performed between a embryogenic line and a nonembryogenic variety.Immature embryos were cultured on Murashige and Skoog medium plus 2 mg/l 2,4-dichlorophenoxyacetic acid, gelled with 5.5 g/l agarose. Somatic embryogenesis was not expressed in the F1's. In contrast, from several hundred immature embryos of the F2 generation of one cross, 10.7% and 1.6% expressed somatic embryogenesis in short and long-term cultures respectively. These percentages of embryogenic: non-embryogenic fits a model of a few complementary genes. The embryogenic capacity of the F2 genotypes depends on the presence of recessive alleles at these gene loci. 以未成熟子叶为外植体,研究25种基因型对大豆愈伤组织形成与体细胞胚胎发生的影响。结果表明,大豆不同基因型间愈伤组织诱导率变化较小,而基因型对体细胞胚胎发生的影响较大,依基因型不同体细胞胚诱导率变化在3.33%~83.33%。体细胞胚诱导率、平均胚数在基因型间的差异均达极显著水平,且两者间存在极显著相关关系。  相似文献

The aim of this study was to examine the embryogenic potential of floral material of the cassava cultivar MCOL 1505. Macerated immature inflorescences were found to be highly embryogenic, with almost 78% of the explants producing somatic embryos. Somatic embryos were also produced from whole male florets and half florets although at much lower rates. No regeneration was obtained from anther, microspore or floret wall tissue. Somatic embryos derived from immature inflorescences were regenerated via organogenesis and the plants derived from this process were assessed in terms of phenotype and ploidy level. If haploid plants could be produced by this method, this would have significant implications in assisting traditional cassava breeding, as this would allow homozygosity to be reached more rapidly. In a crop such as cassava, which is highly heterozygous in nature, the use of haploids in a breeding programme could considerably shorten the time taken to produce new desirable cultivars. This is the first report on plant regeneration through somatic embryogenesis from floral tissue of cassava. A single-step method for the induction and development of somatic embryoids from hypocotyls explains of Brassica juncea is reported. On modified MS medium containing 2 % sucrose, 0.25 mgl 1 2,4-D, 0.5 mgl 1 each of NAA and BaP-R, each explant calluses at both of and at its best, 31% of explants produce embryoids. In the variety RLM-198, the number of embryoids ranges from 8–21 per culture. Each embryoid, upon proliferation, developed up to the 25 shoots. The method is rapid; the time La ken from inoculation to the development of intact plantlets is 8–10 weeks. Regenerated plants have flowered normally and have set seed. The system can profitably be used for in vitro mutant selection and early bulking in mustard.  相似文献

Culture medium composition is critical for the successful induction of microspore division in vitro. The present experiments have focused on a relatively neglected area of cell and tissue culture research, namely the carbohydrate component used in the medium. Three spring barley genotypes were cultured on a medium which was modified by replacing sucrose with the following carbohydrates (6 % w/v): maltose, fructose, malt extract, galactose and a glucose (3 % w/v)/fructose (3 % w/v) mixture. Both maltose and malt extract were superior to sucrose in their capacity to induce green plantlet differentiation from microspores. The concentrations of both sucrose and maltose were also varied. Overall the response of anthers on maltose based media was higher than on sucrose based media. Furthermore, a concentration of maltose m the range 6—12 % w/v produced a higher frequency of green plants than a low concentration (1—3 % w/v). The effect of maltose based media on germplasm of direct relevance to barley breeders was also tested. The cultivar ‘Blenheim’ was shown to be very responsive and this genetic factor was transmitted to the F1 hybrid. The frequency of haploid to diploid regenerants was not consistent over genotypes, but in general there were more haploid than diploid regenerants. The implications of these results for barley breeding are discussed.  相似文献

Nickel (0.13, 0.25, 0.5, 1.0 and 1.5 mg/l) increased the efficiency of somatic embryogenesis in leaf base and mesocotyl derived calli of Setaria italica. A lower concentration of nickel in the culture media promoted long-term maintenance of embryogenic calli that regenerated into plantlets. The plants obtained from embryogenic calli grown on Ni-containing medium showed tolerance to nickel. The growth of embryogenic callus was the maximum at 1.5 mg/l as compared to a lower or a higher concentration of Ni. Use of nickel may help in the induction of high frequency somatic embryogenesis in Setaria italica. This revised version was published online in July 2006 with corrections to the Cover Date.  相似文献

在改良MS培养基上从5个大豆品种成熟种子来源的幼苗未展开叶片上诱导出愈伤组织,利用2个月月龄的这些愈伤组织在摇床上建立了细胞悬浮培养系,细胞悬浮培养系每1至2周转培1次,并用于原生质体分离,转培后3到5天的细胞经酶解后原生质体的产量最高,纯化后的原生质体用3种方法进行培养,即液体培养法、琼脂包埋法和琼脂底层法,在琼脂底层法中,培养皿底层先浇上无甘露醇的琼脂培养基,而原生质体则悬浮在2到4毫升含0.45 mol甘露醇的培养基中,3种方法以琼脂底层法最佳,原生质体在培养后2到3天内能见到第1次细胞分裂,在适当的条件下1周内高达80%的原生质体发生分裂,置板频率一般在40%到60%之间,5个供试品种中4个的原生质体愈伤组织在固体和液体培养中出现体细胞胚胎发生,在液体培养条件下得到了大量子叶和胚根发育良好的体细胞胚,但由于这些胚未进一步萌发,故未能得到完整的再生植株。  相似文献

Summary Seedlings of Trifolium repens showed considerable variation with regard to the morphology and growth of their calli, and their ability for in vitro differentiation of shoots. One of the lines selected for regeneration in primary callus cultures also showed shoot formation from protoplasts. Somatic embryogenesis in callus cultures of T. pratense and T. arvense occurred only in selected seedling lines. This paper highlights the importance of screening a large number of plants within a cultivar of outbreeding species to achieve reproducible plant regeneration from tissue culture.  相似文献

Summary Somatic embryogenesis was initiated from immature embryos on Murashige-Skoog (MS) medium plus 2 mg.l-1 2,4-dichlorophenoxyacetic acid, 2% sucrose and 0.6% agarose. Somatic embryos were isolated and regenerated into whole green plants on MS medium devoid of 2,4-D. These regenerants were previously demonstrated to differ in their mitochondrial DNA organization. In order to estimate their characteristics three progenies of short-term culture regenerants and three progenies of long-term culture regenerants were analyzed and compared to the parental line. These somaclones obtained from the wheat variety Chinese Spring were evaluated for variation of 13 agronomic and morphological quantitative characters in comparison to the parental line. Significant variation was observed for plant height, spike length, main tiller diameter, between the somaclones regenerated from long-term culture and their parent. Differences were observed to increase with the duration of culture, leading to a significant modification of the structure of the plants. Several changes occurred during the somatic tissue cultures, but to a lesser extent than has previously been described in the literature.  相似文献
