黄腐酸对雾培马铃薯幼苗抗旱性的影响   总被引:4,自引:0,他引:4  
试验以马铃薯(Solanum tuberosum)幼苗为研究对象,采用雾培装置进行不同黄腐酸用量和干旱胁迫处理(分别为CK,不加黄腐酸和PEG; 10% PEG;0.01% FA+ 10% PEG;0.03% FA+ 10% PEG;0.05% FA+ 10% PEG和0.10% FA+ 10% PEG),分析其对马铃薯生长发育及抗性生理的影响。结果表明,在干旱胁迫下,雾培马铃薯幼苗的成活率、叶绿素含量、膜稳定指数、根系活力、匍匐茎数、结薯率和产量呈下降趋势,而丙二醛(MDA)、过氧化氢(H2O2)、脯氨酸、可溶性糖含量和过氧化氢酶(CAT)活性及超氧阴离子(O-2)产生速率呈上升趋势,过氧化物歧化酶(SOD)和过氧化物酶(POD)活性先增加后下降,说明雾培马铃薯幼苗在干旱胁迫下生长发育受到显著抑制,正在遭受逆境胁迫。经黄腐酸处理后,幼苗的成活率、匍匐茎数、结薯率和产量则得到了提高,而叶绿素含量、膜稳定指数和根系活力仍呈下降趋势,但较单一干旱胁迫的下降趋势缓慢,而MDA含量、SOD和POD活性及产生速率呈先升高后下降的趋势,脯氨酸、可溶性糖、H2O2含量及CAT活性呈上升趋势,其中H2O2含量上升趋势较单一干旱胁迫缓慢,说明黄腐酸处理后使干旱胁迫下雾培马铃薯幼苗的生长发育得到了促进,减轻了干旱胁迫对幼苗造成的伤害,提高了植株的整体抗旱性,而这种抗性与黄腐酸浓度呈明显相关性,0.05% FA对植株的的保护效应最显著,而0.10% FA则协同PEG加剧了对幼苗的伤害作用。  相似文献

应用不同浓度的聚乙二醇(PEG10000)处理红地球葡萄生根试管苗,在造成一定程度水分亏缺的情况下,观察葡萄试管苗的生理反应和植株生长、叶片形态解剖结构的变化。结果表明:在PEG处理下,葡萄生长发育迟滞,植株节间变短,株高变矮,但发根数量增加,根变长;新生叶片与对照比,叶片小而皱缩,气孔指数降低,上皮细胞排列更紧凑;同时葡萄试管苗脯氨酸的积累和内源ABA迅速增加。说明在一定强度的水分胁迫下,葡萄试管苗叶片的发育向增强其耐旱能力的方向进行,其渗透调节物质的积累是葡萄试管苗对长期水分胁迫的生理反应。  相似文献

以1年生红鳞蒲桃盆栽苗木为研究对象,设置不同盐胁迫浓度及干旱程度,研究盐旱交互作用对红鳞蒲桃苗木的高、地径生长和各项生理指标的影响。结果表明:干旱和盐胁迫都抑制了红鳞蒲桃的生长,盐胁迫的影响要大于干旱胁迫,高生长受到的限制大于地径生长,2 g/kg盐分浓度在水分充足条件下促进了地径生长。相对电导率和丙二醛含量均随着干旱和盐胁迫的增强而增大。不同盐分浓度下,干旱胁迫对相对电导率的影响趋势一致,不同盐分胁迫的影响差异不显著。高盐浓度下丙二醛含量的增加显著大于低盐浓度,表明盐胁迫对红鳞蒲桃的伤害更大;红鳞蒲桃的渗透调节物质在干旱和盐交互胁迫下有一定程度的提高。水分充足时,盐分浓度的变化没有对红鳞蒲桃的游离脯氨酸含量产生影响。随着胁迫的增加,渗透调节能力显著提高。低盐浓度下,抗氧化酶活性在水分充足时没有受到显著影响,高盐浓度下,随着干旱胁迫的加剧则显著提高。  相似文献

Cadmium (Cd) is a widespread environmental toxicant that can cause severe hepatic injury. Oyster protein hydrolysates (OPs) have potential effects on preventing liver disease. In this study, thirty mice were randomly divided into five groups: the control, Cd, Cd + ethylenediaminetetraacetic acid (EDTA, 100 mg/kg), and low/high dose of OPs-treatment groups (100 mg/kg or 300 mg/kg). After continuous administration for 7 days, the ameliorative effect of OPs on Cd-induced acute hepatic injury in Cd-exposed mice was assessed. The results showed that OPs significantly improved the liver function profiles (serum ALT, AST, LDH, and ALP) in Cd-exposed mice. Histopathological analysis showed that OPs decreased apoptotic bodies, hemorrhage, lymphocyte accumulation, and inflammatory cell infiltration around central veins. OPs significantly retained the activities of SOD, CAT, and GSH-Px, and decreased the elevated hepatic MDA content in Cd-exposed mice. In addition, OPs exhibited a reductive effect on the inflammatory responses (IL-1β, IL-6, and TNF-α) and inhibitory effects on the expression of inflammation-related proteins (MIP-2 and COX-2) and the ERK/NF-κB signaling pathway. OPs suppressed the development of hepatocyte apoptosis (Bax, caspase-3, and Blc-2) and the activation of the PI3K/AKT signaling pathway in Cd-exposed mice. In conclusion, OPs ameliorated the Cd-induced hepatic injury by inhibiting oxidative damage and inflammatory responses, as well as the development of hepatocyte apoptosis via regulating the ERK/NF-κB and PI3K/AKT-related signaling pathways.  相似文献

AIM To investigate the effects of astragaloside on the levels of sex hormone and oxidative stress in rats with polycystic ovary syndrome (PCOS). METHODS Female SD rats (n=60) were randomly divided into normal control group, model group, Diane-35 (0.339 2 mg/kg) group, low dose astragaloside (12.5 mg/kg) group and high dose astragaloside (50 mg/kg) group, with 12 rats in each group. The PCOS model was induced by letrozole (1 mg/kg), which was administered by gavage once a day for 3 weeks. After administration, the estrus cycle of the rats was observed by vaginal smear, and the ovarian index was calculated. HE staining was used to observe the histopathological changes of the ovaries. Serum levels of the sex hormones testosterone (T), estradiol (E2), luteinizing hormone (LH) and follicle-stimulating hormone (FSH) were measured by ELISA. The levels of superoxide dismutase (SOD), malondialdehyde (MDA) and glutathione peroxidase (GSH-Px) in serum and ovarian tissue were detected by colorimetry, and the protein levels of steroidogenetic acute regulatory protein (StAR) and apoptosis-related proteins cleaved caspase-3, Bax and Bcl-2 in ovarian tissue were detected by Western blot. RESULT Compared with control group, the oestrous cycle of the rats in model group was disorder, and the ovarian index was increased, ovary was polycystic. The serum levels of T, LH and MDAwere significantly increased (P<0.05), while the contents of E2, FSH and the activities of GSH-Px and SOD were significantly decreased (P<0.05). The levels of MDA, StAR, cleaved caspase-3 and Bax proteins in ovarian tissue were significantly up-regulated (P<0.05). GSH-Px and SOD activities and Bcl-2 protein levels were significantly down-regulated (P<0.05). CONCLUSION Astragalosideeffectively balances the levels of sex hormone in PCOS rats and relieves the oxidative stress injury, the mechanism may be related to the inhibition of StAR expression.  相似文献

Summary

Retrotransposons are major components of the genomes of most eukaryotic organisms and have resulted in the introduction of desirable traits in many crops, including fruit trees. Here, we describe a Ty3-gypsy-like retrotransposon associated with a short-catkin mutant in Chinese chestnut (Castanea mollissima), resulting in catkins that are < 20% the length of normal staminate catkins. A partial sequence of the retrotransposon, named CmRT1, detected by amplified fragment length polymorphism (AFLP) analysis, and its complete sequence were determined from the genome of Chinese chestnut (Castanea mollissima) using improved Tail-PCR. CmRT1 was 10,067 bp in length and shared high homology in its predicted amino acid sequence and motifs with other Ty3/gypsy-like retrotransposons. The 5’ long terminal repeat (LTR) of CmRT1 contained a TATA box and several cis-elements that were predicted to be important for processes involving abscisic acid, gibberellic acid, and auxins and in stress-mediated responses. Further characterisation of the transposition event that led to the short-catkin phenotype was performed using two pairs of primers that aligned with the flanking region of the LTRs. The expected PCR bands were observed only in genomic DNA from plants that showed the mutation. Finally, cloning and real-time qPCR analysis of an NADP-dependent alkenal double-bond reductase (CmADBR) target gene that was adjacent to CmRT1, revealed that CmADBR expression was significantly down-regulated in the short-catkin mutant. Taken together, these results suggest that the CmRT1 retrotransposon is responsible for the short-catkin phenotype.  相似文献

以酿酒葡萄"赤霞殊"(Cabernet Sauvignon,CS)1年生扦插苗为试材,采用营养液水培系统,研究了根施外源硅(K2SiO3·9H2O,Si)对盐(50 mmol/L NaCl)胁迫下赤霞珠葡萄幼苗叶片生理生化特性的影响.结果表明:盐胁迫处理下,葡萄幼苗叶片超氧阴离子(O2-')产生速率加快,过氧化氢(H2O2)、丙二醛(MDA)含量和相对膜透性增加;超氧化物歧化酶(SOD),过氧化物酶(POD)和过氧化氢酶(CAT)活性增强;组织含水量降低;施用2.0mmol/L Si明显抑制了O2-、H2O2和MDA的积累,增加了组织含水量,并进一步提高了抗氧化酶SOD和POD的活性.说明外源Si可通过提高盐胁迫下葡萄幼苗体内的抗氧化酶活性,从而降低活性氧(ROS)水平,一定程度上减轻盐胁迫对植株所造成的细胞膜脂过氧化伤害,增强其耐盐性.  相似文献

The purpose of this study was to evaluate oxidative stress and neurotoxic potential of organophosphorus (OP) insecticide diazinon in the sentinel freshwater fish, Oreochromis niloticus. Antioxidant and acetylcholinesterase (AChE) enzyme activities and malondialdehyde (MDA) and protein levels were measured spectrophotometrically in gill, kidney, alimentary tract, and muscle tissues of fish treated with sub-lethal diazinon concentrations for 1, 7, 15, and 30 days. Dose-dependent inhibitions of AChE were observed in all the experimental fish. On the contrary of alimentary tract, MDA levels were elevated in kidney and muscle and gill was not affected. AChE and MDA levels intercorrelated in kidney and muscle tissues. Diazinon had increased superoxide dismutase (SOD) activities in all the tissues, while kidney was the most affected tissue. Tissue-specific alterations were observed on catalase (CAT) and glutathione peroxidase (GPx) activities; however, the activities were not changed in gill and muscle tissues for GPx and in gill, muscle, and kidney tissues for CAT. Protein levels decreased in kidney, muscle, and alimentary tract, while increased in gill and alimentary tract in 15 days. With respect to these results, diazinon has oxidative and neurotoxic potentials in O. niloticus. Observed changes with diazinon treatment were generally tissue-specific and dose-dependent.  相似文献
