[目的]建立生石花植株离体再生及组培快繁体系。[方法]以生石花成熟种子为外植体,研究生石花组培快繁技术。[结果]萌发后的幼苗在MS+0.50 mg/L 6-BA+0.05 mg/L NAA培养基上被成功诱导出愈伤组织;不添加6-BA的MS培养基有利于愈伤组织的分化,不定芽诱导率为4.65;在MS+0.05 mg/L 6-BA+0.01 mg/L NAA培养基上不定芽增殖率较高,可达5.60。[结论]建立了生石花植株离体再生及快繁体系,有利于生石花种质资源保护和工厂化生产。  相似文献

[目的]研究影响蝴蝶兰花梗芽增殖率的因素。[方法]在蝴蝶兰花芽产生期分别采集2、3、4、5和6 cm的花梗芽为试材,并把材料设成1/4、1/2、3/4的节段,研究花梗芽采摘时间、培养基配方和暗培养对蝴蝶兰花梗芽增殖率的影响。[结果]5~6 cm的花梗芽和3~4 cm的花梗芽在分化腋芽数量上有所差异,但在总增殖率上没有明显差异。暗培养能更好诱导不定芽的产生,但对1/4和1/2节段的花梗芽影响较大。从节位来看,3/4的花梗芽诱导效果最好。筛选培养基的正交试验发现,KC l及KT能有效提高增殖率,NAA对花梗芽增殖有抑制作用。[结论]试验筛选的培养基1/2 MS+KC l 1.0 g/L+BA 5.0 mg/L+NAA 0.6 mg/L+KT 1.0 mg/L可使蝴蝶兰花梗芽的增殖率达到8.5个,是试验初始增殖率的3~4倍。  相似文献

应用 ̄3H-TdR掺入法和ELISA测定了锌和其它微量元素对离休boPBMC增殖和分泌免疫球蛋白(Ig)的影响。结果表明,0.01mmol/L的锌能促进boPBMC增殖(P<0.05)和分泌Ig(IgG_1,p<0.05;IgM,P<0.01);其它微量元素,诸如镍,铜,铁,镉等均无这种作用。此外锌对培养2d后的细胞不表现出促进作用的结果表明,锌主要作用于免疫细胞被活化的初始阶段,提高了细胞的增殖和分化能力。  相似文献

AIM:To study the relationship between apoptosis, proliferation and expression,mutation of related genes in breast cancer.METHODS:Methods of TUNEL, immunohistochemical S-P and PCR-SSCP were used respectively to study apoptotic index (AI), mitotic index(MI), expression of Bcl-2,p53,c-erbB-2,PCNA,Ki67,TopoⅡ and mutation of p53 in 54 cases of breast cancer.RESULTS:AI and MI were 9.40±3.78 and 5.96±2.36, respectively. There was a significant direct correlation between them(r=0.46.P<0.01).High expression of Bcl-2,PCNA,Ki67,TopoⅡ coincided with high AI,MI(P<0.01). High expression of p53,c-erbB-2 and mutation of p53 coincided with high MI(P<0.01). Type of p53 mutation coincided with AI(P<0.05).CONCLUSION:Disturbance of gene control between apoptosis and proliferation is related with expression,mutation of related genes in breast cancer.  相似文献

AIM: To observe the effects of Yangxue qingnao-containing serum on rat vascular smooth muscle cell (VSMC) proliferation induced by lysophosphatidic acid (LPA). METHODS: The [3H]-TdR incorporation and mitogen-activated protein kinasc (MAPK) activity were measured in cultured VSMC. End product of lipid peroxidation-MDA levels were also detected. RESULTS: 1×10-9,1×10-8 and 1×10-7 mol/L LPA enhanced the cultured VSMC [3H]-TdR incorporation, increased MAPK activity and MDA content in a concentration-dependent manner. 5%, 10% and 15% Yangxue qingnao-containing serum concentration-dependently inhibited the increase in VSMC [3H]-TdR incorporation, MAPK activity and MDA content induced by LPA. CONCLUSIONS: LPA has a stimulating effect on VSMC proliferation. The LPA-induced intracellular signal transduction may be related to MAPK activity. Yangxue-qingnao can efficiently inhibit LPA -induced VSMC proliferation,MAPK activity and lipid peroxidation.  相似文献

AIM: To investigate the effects of testosterone on endothelial function and intimal proliferation after balloon injury in male rabbit abdominal aorta. METHODS: 24 male New Zealand white rabbits were divided randomly into three groups: control group (n=8, sham castration), hypotestosteronemia group (n=8,castration) and testosterone replacement group (n=8,castration +testosterone undecanoate intramuscular injection,14mg/kg). Abdominal aorta was injured with 3 mm PTCA balloon after testosterone undecanoate had been injected for three days. Two weeks later, blood samples were obtained for detection of plasma testosterone, lipids, metabolic product of nitric oxide (NO2-/NO3-), superoxide dismutase(SOD) and malondialdehyde (MDA),and all the abdominal aorta were excised to be analyzed by computer. RESULTS: The intimal area of hypotestosteronemia group were significantly larger than that of other two groups(P<0.01). plasma NO2-/NO3- and SOD levels were significantly decreased, while the total cholesterol(TC),triglycerides(TG), low density lipoprotein(LDL) and plasma MDA were significantly increased. No difference was observed between control group and testosterone replacement group in all parameters. CONCLUSION: Testosterone, at physiological level,had the effects of inhibiting the intimal proliferation and of protecting the endothelial function after balloon injury in male rabbit abdominal aorta.  相似文献

最近,由于干细胞研究普遍取得的令人鼓舞的进展以及干细胞分离、培养、移植等新技术方法的出现和发展,出生后雄性个体的生殖系干细胞———精原干细胞(spermatogonialstemcell,SSC)已成为一个愈加引人瞩目的课题。SSC位于睾丸曲细精管基膜上,既具有自增殖(self-renewal)潜能和定向分化潜能,又是自然状态下出生后个体内在整个生命期间能够进行自增殖并能将基因传递至子代的惟一成体干细胞。了解干细胞增殖与分化的调节机制,获得足够数量的种子细胞一直是所有干细胞研究共同面临的迫切问题之一。近年来,借助于SSCs移植技术这一功能性检测方法,国内外学者就SSCs体外培养的条件进行了深入探讨,取得了可喜进展。结合自己的前期工作,参考相关最新进展,就影响哺乳类SSCs体外存活、增殖、分化的因素及未来需要解决的问题进行了评述。  相似文献

With different parts of bulb scale as explants,the proliferation method of Guizhou Lilium brownii were studied with 3% sodium hypochlorite and MS medium with different concentrations of hormones.The results show that it is feasible to disinfect the bulbs of Lilium brownii with 3% sodium hypochlorite,moreover,the sodium hypochlorite is very cheap and harmless to researchers,experimental materials and environment.MS+NAA 0.3 mg/L+6-BA 1.5 mg/L is optimum for the induction of bulbs and,the basal part of Lilium brownie is the optimum explants.After culture for 25 d on the same medium,the tube bulbs could be obtained with the characteristics of high propagation coefficient,strong and new roots.The survival rate is over 90% for transplantation of tube bulbs with diameter between 1-2 cm.The method developed in the present study can proliferate abundant Lilium brownii seedling in short time.  相似文献

研究虎杖疏松愈伤组织诱导的最佳因素组合.通过单因素和正交试验研究不同外植体、不同浓度的非激素水平和激素水平对虎杖愈伤组织诱导率、疏松程度以及增殖率的影响.结果表明:以叶柄为外植体更容易诱导出疏松的愈伤组织,虎杖愈伤组织诱导的最佳非激素因素为pH值5.8,蔗糖5 g/L,水解酪蛋白300 mg/L,肌醇10 mg/L,继代增殖最佳的激素组合2,4-D 2.5 mg/L+NAA 0.3 mg/L.适宜的激素和非激素组合,能促进虎杖获得质地疏松、淡黄色、生长旺盛的愈伤组织.  相似文献

以对节白蜡带芽嫩茎为外植体,以改良的MS为基本培养基,附加不同种类和浓度的植物激素来诱导产生芽、根,进而培育出完整植株,结果表明:适宜的培养基为(1)诱导芽为改良MS 2mg/L6-BA 0.1mg/L NAA(2)芽增殖为改良MS 1.5mg/L6-BA 0.1mg/L NAA(3)生根培养基1/2MS 0.5mg/LVB1 0.3mg/L NAA。  相似文献
