Roots of Morinda citrifolia (Noni or Yor in Thai) have been used traditionally for thousands of years to treat chronic diseases such as cancer and heart disease. In this study, three new anthraquinones together with 15 known compounds were isolated from the roots of M. citrifolia (Rubiaceae). Their structures were established by spectroscopic methods, particularly 1D and 2D NMR techniques. Six known compounds, together with two new compounds (2 and 3) showed significant inhibitory effects on the proliferation of human lung and colon cancer cells. 相似文献

[目的]研究大花蕙兰茎尖外植体原球茎诱导及增殖培养、褐化污染的影响因素。[方法]以大花蕙兰茎尖作为外植体,研究无菌系建立、诱导培养的灭菌方法与褐化防治、原球茎诱导培养初始培养基的筛选、原球茎增殖培养的影响因素。[结果]结果表明:解决茎尖内生菌污染及褐化严重的问题,两段式灭菌(75%酒精30 s+0.1%升汞10 min+剥去幼叶+0.05%升汞5 min)优于单一式的灭菌;原球茎诱导培养基为1/2MS+2.0 mg/L 6-BA+0.2 mg/L NAA;原球茎增殖培养基为1/2MS+2.0 mg/L 6-BA+0.2 mg/L NAA+2.0 g/L活性炭+10%马铃薯。[结论]该研究可为大花蕙兰的茎尖快速繁殖及工厂化生产提供技术依据。 相似文献

AIM: To explore the effects of uremic serum on proliferation and trans-differentiation of human renal tubular epithelial cells. METHODS: Human renal tubular epithelial cell line (HK-2) was cultured in RPMI-1640 medium. The proliferation effects of uremic serum at different concentrations were evaluated by methylene blue assay (MTT method) and flow cytometry. The positive cells percentage of α-smooth muscle actin(α-SMA)in different concentration uremic serum medium was also measured by flow cytometry in vitro. RESULTS: Absorbance 490 (A490) was increased in 5%-20% uremic serum groups compared with that in normal controls with the use of MTT. Cells in G1 phase were decreased, but proliferation index (PI) was increased in 10%-20% uremic serum groups compared with that in normal controls with the use of flow cytometry. No significant difference of cell proliferation index was found among uremic serum groups. The positive percentage of α-SMA cells was increased significantly in uremic serum groups compared with that in normal controls, and increased in parallel with the increasing of uremic serum concentration. CONCLUSION:These data suggest that AGEs could induce a high expression of MIP-1αm RNA and protein in cultured HUVECs in a dose-dependent and time-dependent manner. 相似文献

目的:探讨survivin蛋白在胶质瘤中的表达情况及其与增殖指数、凋亡指数和微血管密度的相关性。方法:采用S-P法检测胶质瘤56例和正常脑组织8例中survivin蛋白、Ki67和FⅧRAg的表达,肿瘤细胞的凋亡采用TUNEL检测,并结合病理特点进行分析。结果:Ⅰ~Ⅱ、Ⅲ、Ⅳ级胶质瘤中,sur-vivin蛋白的表达率分别为25.3%、56.8%、83.2%,组织学分级之间差异有显著性(P〈0.05);survivin表达阳性者的Ki67、微血管密度明显高于survivin阴性者,而其凋亡率则低于survivin阴性者,差异有显著性(P〈0.05);正常脑组织中未见survivin表达。结论:Survivin在胶质瘤中高表达,与其恶性程度、凋亡、增殖活性及血管生成密切相关。 相似文献

[目的]建立红肉苹果的高效植株再生体系。【方法]以红肉苹果的茎尖和带腋芽的茎段为外植体,通过组织培养试验筛选出最佳的灭菌时间、基础培养基、增殖培养基和生根培养基。[结果]随着消毒时间的延长,外植体的污染率明显下降,但外植体受到的伤害显著增加。用0.1%HgCl2灭菌5rain的外植体生长正常,污染率仅为3.3%。在增殖培养基MS+6一BA0.5mg/L+NAA0.1mg/L、MS+6.BA0.5mg/L+NAA0.2mg/L和MS+6一BA0.5mg/L+NAA0.5mg/L中。每个外植体平均增殖不定芽4.1、6.8和3.3个。在生根培养基1/2MS+0.1mg/LIBA+0.1mg/LNAA、1/2MS+0.2mg/LIBA+0.2mg/LN从和1/2MS+0.5mg/LIBA+0.5mg/LNAA上幼苗的生根率分别为56.7%、96.7%和83.3%。[结论]该研究为红肉苹果的快速离体扩繁和工厂化育苗以及园林应用奠定了良好的基础。 相似文献

Interleukin-12 (IL-12) plays a pivotal role in regulating cellular immune responses involving autoimmunity, infectious disease, and cancer. Human recombinant (hr) IL-12 is being evaluated for therapy of human cancer. We investigated the potential of hrIL-12 to activate canine peripheral blood mononuclear cells (PBMC) using proliferation and cytotoxicity as readouts. Human rIL-12 caused increased proliferation of PBMC, and enhanced lysis of allogeneic canine tumor targets mediated by PBMC from normal dogs in vitro. In addition, antibody-dependent cellular cytotoxicity (ADCC) mediated by canine PBMC was enhanced by hrIL-12. These results indicate that hrIL-12 is recognized by canine immune cells, triggering a number of immune responses in canine PBMC, that may be important for immunotherapy of canine cancer. Information from this investigation provides impetus for evaluation of the effects of hrIL-12 on PBMC from tumor-bearing dogs and should be helpful in the development of hrIL-12 as an immune cell activator in vivo in the dog. 相似文献
