为阐释津田芜菁Br UBC11基因的表达特性,克隆了津田芜菁UBC11基因的全长c DNA序列,命名为Br UBC11,Gen Bank登录号为KM396887。其c DNA全长685 bp,ORF区全长447 bp,编码148个氨基酸的开放阅读框;亚细胞定位结果显示,Br UBC11-GFP定位于细胞核内,表明Br UBC11蛋白可能在细胞核中发挥其功能。荧光定量PCR检测Br UBC11在芜菁不同组织中的表达结果表明,该基因在花瓣中表达量最高,花蕾中次之,具有组织特异性。而且Br UBC11在芜菁白色根皮中的表达受长波紫外线(UV-A)诱导。研究结果显示,在芜菁中,UBC11属于多功能基因,具有潜在的参与花发育和UV-A信号转导相关途径的功能。 相似文献

Creation of a New Resistant Germplasm to Verticillium Wilt by Distant Hybridization in Upland Cotton 总被引:1,自引:1,他引:0
To create new germplasm lines resistant to Verticillium wilt in upland cotton, 65 distant hybridization germplasm lines (DHGLs) in upland cotton genetic background were cultivated by interspecific hybridization between Gossypium hirsutum and wild species including G. anomalum, G. armourianum, G. aridum, G. raimondii, G. mustelinum, interspecific F1 backcrossing with G. hirsutum for four generations, and selfing for four generations, followed by a conventional breeding program. The results of agronomic trait identification during 2011-2012 indicated that average plant heights of DHGLs were closely similar to commercial cultivars of upland cotton, while average fruit branches, fruit nodes, bolls of DHGLs individual plant were lower than those in commercial cultivars of upland cotton. Average single boll weight and lint percentage of DHGLs were lower than commercial cultivars of upland cotton. Fiber length, strength, fineness and maturity of DHGLs were reasonably collocated. Fiber of most lines was suitable for spinning extra high count yarn, but the main fiber quality indices of commercial cultivars of upland cotton were not well coordinated. Identification of resistance to Verticillium wilt in defoliation disease nursery during 2012-2013 indicated that five DHGLs resistant to Verticillium wilt . Suyuan 040, Suyuan 045 and Suyuan 061 were highly resistant to Verticillium wilt with a disease index of 8.33, 4.35 and 7.79, respectively. Suyuan 030 and Suyuan 034 were resistant to Verticillium wilt with a disease index of 12.35 and 13.70, respectively. The genetic relationship of new germplasm lines resistant to Verticillium wilt were traced and showed that Suyuan 040 and Suyuan 045 were DHGLs of G. raimondii, Suyuan 061 was DHGL of G.mustelinum, Suyuan 030 and Suyuan 034 were DHGLs of G. aridum. 相似文献

Summary Eight stem rust (Puccinia graminis tritici Eriks. and Henn.) resistant lines (designated TICENA lines) that had been selected by Veiga et al. (1981) following gamma radiation of BH-1146 wheat (Triticum aestivum L.) were studied. Six of the lines were resistant to race 15B-1 of stem rust and susceptible to race 56, and proved to carry the gene Sr7a. TICENA 4 carries two unidentified genes, each giving resistance to one of the two races. TICENA 10 carries Sr6, Sr7a and an unidentified gene giving resistance to race 56 but not 15B-1. ACC合成酶(ACS)是高等植物乙烯生物合成途径中的限速酶。在克隆到甘蔗ACC合成酶基因家族3个成员Sc-ACS1、Sc-ACS2和Sc-ACS3的基础上,分别以它们作为探针,检测了激素诱导和环境胁迫对甘蔗苗期该3成员表达的影响。结果表明,乙烯利诱导Sc-ACS1在叶中表达,在根中不表达;Sc-ACS2在根、叶中表达;Sc-ACS3主要在根部表达,在叶中不表达。在甘蔗叶片中,Sc-ACS1对冷胁迫、暗培养和LiCl胁迫均有应答,并受植物生长激素IAA诱导而表达;Sc-ACS2无论是在生长激素诱导还是环境胁迫下,其mRNA均维持较低水平。 相似文献

A total of 105 European wheat cultivars were assessed for seedling and adult plant resistance (APR) to stem rust using an
array of Australian isolates of Puccinia graminis f. sp. tritici. Twenty-seven cultivars were susceptible at both seedling and adult plant growth stages. Twelve catalogued seedling stem
rust resistance genes (Sr7b, Sr8a, Sr8b, Sr9b, Sr9g, Sr11, Sr15, Sr17, Sr29, Sr31, Sr36 and Sr38) were detected in the remaining cultivars, and 13 cultivars carried additional seedling resistance genes that could not be
postulated with the isolates used. Low levels of APR to stem rust were found in the cultivars Artaban, Forno, Mec, Mercia,
Pandas and Vlada. Although the genetic identity of this APR was not determined, it was clear that the only designated stem
rust APR gene Sr2 was not present in any of the cultivars tested based on the absence of the linked traits seedling chlorosis and pseudo black
chaff. One of these cultivars, Forno, is believed to carry the leaf rust APR gene Lr34, previously reported to be associated with improved resistance to stem rust. A detailed genetic characterisation of the APRs
in these cultivars will be needed to understand their modes of inheritance and relationships with catalogued stem rust resistance

Downy mildew resistance originating from Allium roylei Stearn provides a complete resistance to onions and is based on one, dominant gene. Since A. roylei can successfully be hybridized with onion (A. cepa L.), a breeding scheme aimed at the introgression of this gene was initiated ca. 20 years ago. Several setbacks in this programme were encountered, firstly the identified molecular marker linked to the downy mildew resistance locus became increasingly difficult to use and finally lost its discriminating power and secondly the final step, making homozygous introgression lines (ILs), turned out to be more difficult then was hoped. GISH analysis showed that the chromosomal region harbouring the resistance locus was the only remaining piece of A. roylei in the nuclear background of onion and it also confirmed that this region was located on the distal end of chromosome 3. It was hypothesized that some factor present in the remaining A. roylei region was lethal when homozygously present in an onion genetic background. The identification of an individual with a smaller and more distally located introgression fragment and homozygous ILs in its progeny validated this hypothesis. With the help of these nearly isogenic lines four AFLP® markers closely linked to the resistance gene were identified, which can be used for marker-aided selection. The introduction of downy mildew resistance caused by Peronospora destructor into onion is a significant step forward in the development of environmentally-friendly onion cultivars. 相似文献
