草花育苗大棚定时灌溉系统研制

研究了采用DS12C887提供实时时钟、单片机AT89S51进行控制的草花育苗大棚灌溉系统.该控制系统采用模块化的设计,把时钟单元设计成了一个单独的模块,这样可以向任意多的定时单元提供实时时钟信号,所以控制系统有很好的扩展性,可以根据大棚的数目来扩展控制器的数目.通过现场调试,其运行满足实际要求.     

Protective effect of folate and vitamin B12 on Alzheimer-like pathological changes in brain of aged rats with hyperhomocysteinemia

AIM:To explore the effect of hyperhomocysteinemia on the Alzheimer-like pathological changes in the brain of aged rat. METHODS:The rats were treated with homocysteine(Hcy) by intravenous injection through vena caudalis with or without a simultaneous supplementation of folate and Vitamin B₁₂(FB) in the drinking water for 28 weeks. The distribution and aggregation of tau protein were detected by Bielschowsky silver staining. Western blotting was used to determine the protein levels of tau phosphorylation, glycogen synthase kinase 3β(GSK-3β) and protein phosphatase 2A(PP2A). RESULTS:Treatment with Hcy induced hyperhomocysteinemia in aged rats and resulted in the formation of neurofibrillary tangles in the brain. Supplementation of FB effectively reduced the tangles. Hyperhomocysteinemia also induced Alzheimer-like tau hyperphosphorylation at multiple sites(Ser^199/202and Ser³⁹⁶). Hyperhomocysteinemia activated GSK-3β and inhibited the activity of PP2A. Supplementation of FB alleviated the changes of tau and the activities of GSK-3β and PP2A. CONCLUSION:Supplementation of FB ameliorates the hyperhomocysteinemia-induced Alzheimer-like pathological changes of tau protein possibly through regulating the activity of GSK-3β and PP2A in aged rats.     

Role of ghrelin in cell protection by up-regulating HSP70 through ERK1/2 signaling pathway in PC12 cells

AIM:To study the role of ghrelin in cell protection by up-regulating heat shock protein 70 (HSP70) and inhibiting apoptosis induced by oxidative stress through extracellular regulated protein kinases 1/2 (ERK1/2) signaling pathway in the PC12 cells. METHODS:Sodium nitoprusside (SNP) was used to induce oxidative stress injury in the PC12 cells. The cultured PC12 cells were divided into SNP-injured group (incubated with SNP at 0.5 mmol/L for 6, 12, 18 and 24 h), ghrelin pretreatment group (ghrelin at 100 nmol/L was given 30 min before adding SNP); HSP70 inhibitor group (quercetin at 10 μmol/L was added 60 min before ghrelin treatment), ERK inhibitor group (ERK 1/2 inhibitor PD98059 was added 60 min before ghrelin treatment) and control group (added same amount of culture medium only). The apoptotic rate was detected by flow cytometry. The protein expression was determined by Western blot and immunocytochemistry. RESULTS:Compared with control group, the apoptotic rate of PC12 cells in SNP-injured group was significantly increased (P<0.05). Compared with SNP-injured group, ghrelin (100 nmol/L) pretreatment significantly inhibited SNP-induced apoptosis of PC12 cells (P<0.05), and significantly up-regulated the protein expression of HSP70 (P<0.05). Time-effect analysis showed that ghrelin had the most significant effect at 18 h after SNP injury. Quercetin, an inhibitor of HSP 70, significantly reduced the anti-apoptotic effect of ghrelin (P<0.05). Ghrelin pretreatment promoted the phosphorylation of ERK1/2. ERK1/2 inhibitor PD98059 significantly inhibited the effects of ghrelin on up-regulation of HSP70 expression (P<0.05). CONCLUSION:Ghrelin upregulates the expression of HSP70 and inhibits the apoptosis in the PC12 cells induced by oxidative stress by promoting the phosphorylation of ERK1/2.     

高粱bmr-6和bmr-12基因对产量和品质性状的效应比较

比较了在Atlas、Early Hegari-Sart、Kansas Collier、Rox Orange和Wheatland 5种高粱遗传背景下,bmr-6和bmr-12基因对产量性状及品质性状的影响。结果表明,bmr-6和bmr-12基因使株高显著降低,同时增加了植株的茎粗;而对分蘖数和叶片数的影响较小。bmr-6和bmr-12对产量性状是负面影响,但在不同的遗传背景下不一致。对于品质性状,bmr-6一般是使中性洗涤纤维和酸性洗涤纤维降低,而bmr-12则对这两者影响不大。bmr-6和bmr-12都使酸性洗涤木质素和酸不溶灰分降低。综合考虑,bmr-12的育种利用价值比bmr-6大。     

基于线粒体基因组COI和12S rRNA序列的魁蚶中国群体分类学地位研究

东北亚地区不同地理种群魁蚶(Scapharca brouhtonii)的分类学地位仍存在较大争议。为探讨魁蚶中国群体的分类地位,测定了魁蚶中国群体线粒体基因组COI和12S rRNA序列,并从GenBank中下载了蚶科26种贝类的同源序列进行分析。以贻贝科的贻贝(Mytilus edulis)为外群,用邻近法和最大简约法构建了蚶科贝类的分子系统进化树,分析其分子系统进化关系和分类等级。结果显示,魁蚶中国群体的COI和12S rRNA基因的碱基组成、密码子使用率及变异位点等与魁蚶日本群体和粗饰蚶属的Anadara sativa相似,且遗传距离很小,进化树中聚为一支,亲缘关系很近,初步确定魁蚶中国群体的分类地位和魁蚶日本群体一致。本研究结果初步阐明了魁蚶中国群体的分子系统进化地位,有利于掌握魁蚶的遗传背景和资源状况,以应用于自然资源的保护和推动养殖业的发展。     

水稻瘤矮病毒P12蛋白抗血清制备及其应用

为了进一步研究水稻瘤矮病毒(Rice gall dwarf virus,RGDV)P12的功能,将RGDV-GX P12编码区亚克隆至原核表达载体,并导入大肠杆菌诱导表达;重组的P12融合蛋白经Ni-NTA His·Bind Resins纯化后用于免疫小鼠制备抗血清,并进行Western blotting分析。结果显示,约41 kD大小的RGDV P12融合蛋白可在大肠杆菌中高效表达;利用该重组蛋白所制备的抗血清可特异地与融合表达和非融合的P12蛋白发生强烈的免疫学反应。利用该特异性抗血清在病毒粒子样品中检测不到P12蛋白,而在病株总蛋白样品中可检测到1条与预期大小一致的明显条带,表明RGDV P12是一种非结构蛋白。     

Construction of a recombinant adenovirus carrying IL- 12 -IRES-CK gene and its expression in hepatocyte

AIM: To construct a bicistronic recombinant adenovirus carrying creatine kinase (CK) and human interleukin 12 (hIL-12) gene, and then detect the expression of both genes in vitro and metabolic product of CK in vivo. METHODS: Two PCR products, CK and hIL- 12 genes linked by IRES, were inserted into adenoviral vector. Adenovirus particles carrying CK-IRES-IL- 12 gene was generated through homologous recombination, packaging and propagation. Rabbit hepatocytes were isolated and transfected with adenovirus particles. CK and IL-12 gene expression was confirmed by Western blotting and ELISA, respectively.RESULTS: The expression of CK and human IL-12 in hepatocyte were confirmed in vitro. Metabolic product of CK, phosphocreatine (PCr), could be detected in liver by non-invasive phosphorus-31 magnetic resonance spectroscopy (³¹P MRS) in vivo.CONCLUSION: In vivo detection of ectopic expression of CK gene in liver can be achieved by non-invasive ³¹P MRS through adenovirus transduction. The bicistronic recombinant adenovirus Ad5-hIL-12-IRES-CKb carrying CK and hIL- 12 provides a useful tool for the further study of using a reporter gene expression (CK) dynamically, non-invasively monitor a therapeutic gene expression (IL-12) in liver.     

桉树立地生产力与养分生产力参数的确定

桉树试验林样本分析结果表明，桉树树杆中氮，磷，钾养分浓度的倒数与养分吸收量早明显的负线性相关。基于这一相关性，对实测的数据作回归分析。估测的立地生产力极限和养分生产力参数值，以及相应的立地养分效应曲线具有较高的精确度。本文为研究林木生长和营养的动态函数关系提供了新的思路和方法。     

高产抗逆大豆新品种石豆12号的选育

石豆12号是由石家庄市农林科学研究院以石豆1号为母本,汾豆63为父本经有性杂交选育而成。2015-2016年参加河北省夏播大豆区域试验,平均产量3 284.0 kg·hm~(-2),较对照冀豆12增产6.22%。2016年参加河北省夏播大豆生产试验,平均产量3 229.7 kg·hm~(-2),较对照冀豆12增产4.91%。2017年通过河北省农作物品种审定委员会审定,准予推广。石豆12号的主要特点是高产、抗病、抗倒伏,适宜在河北省中南部夏播种植。     

棉花GhMADS12基因的克隆及其表达载体的构建

MADS-box基因是真核生物中一类重要的转录调控因子,调节着植物根、茎、叶到花的发育和果实的成熟。本研究以中棉所36花发育期均一化全长cDNA文库为基础,分离出一个棉花的MADS-box基因全长cDNA命名为GhMADS12。同时利用pBI121双元载体成功构建了GhMADS12基因的植物过表达载体,并利用基因枪轰击法瞬时表达载体,验证了载体的可靠性,为下一步转基因验证基因的功能奠定了基础。
