The evaluation of endophyte toxin residues in sheep fat

Abstract

AIM: To monitor changes in concentrations of lolitrem B and epoxy-janthitrems in the fat of sheep grazing perennial ryegrass infected with wild-type- and AR37-endophyte, respectively, during the time of year when ryegrass staggers would be expected to be observed.

METHODS: Ten 5-month-old lambs with no previous exposure to endophytes were grazed on either wild-type (containing lolitrem B, n = 5) or AR37 (containing epoxy-janthitrems, n = 5) endophyte-infected perennial ryegrass (Lolium perenne L.) pastures between October 2008 and June 2009. Animals were regularly assessed for ryegrass staggers using the Keogh scale (0 = no signs, 5 = severe tremors). When a score of > 3.5 was observed animals were removed from the treatment pastures for 1 month. Fat biopsy samples were taken from each animal at approximately monthly intervals and analysed for endophyte metabolites using high performance liquid chromatography (HPLC) methods developed during this study. Regular herbage samples were also taken and concentrations of endophyte metabolites measured.

RESULTS: Efficient and reproducible methods to analyse both lolitrem B and epoxy-janthitrems in fat were developed. Concentrations of lolitrem B and epoxy-janthitrems in herbage and in sheep fat increased from late November to peak in mid-February. Ryegrass staggers was observed in both groups of sheep at this time. Following 1 month of grazing non-infected pasture mean concentrations in fat of lolitrem B decreased by 43% from 61.8 to 35.3 ppb, and of epoxy-janthitrems by 38% from 1032.0 to 639.5 ppb. Maximum concentrations in herbage of epoxy-janthitrems (35.7 ppm) were higher than of lolitrem B (3.4 ppm), but signs of staggers were less severe in sheep grazing pasture containing the former compared with the latter (median Keogh scores in late February were 2 and 3, respectively), consistent with epoxy-janthitrems being low potency toxins.

CONCLUSION: This study demonstrated that concentrations of epoxy-janthitrems and lolitrem B in sheep fat increased quickly during the initial phase of the study when concentrations in pasture increased, and decreased when animals were removed from pastures containing these compounds. These data will be used in the risk assessment of the endophyte metabolites.     

云粳37特征特性与半机械直播栽培技术

介绍水稻品种云粳37的特征特性,并总结了其高产栽培技术,为该品种轻简高效栽培提供技术支撑。云粳37为粳型常规水稻品种,在贵州省遵义市种植,全生育期为153 d,株高120 cm左右,穗长18.3 cm,有效穗数256万穗/hm^2,每穗总粒数143.6粒,结实率88.7%,千粒重24.94 g,示范种植单产在6750~7500 kg/hm^2;具有耐寒、感光性强、抗病、米质好、香味重、食味好、高产、生育期早等特点。     

潮霉素抗性基因筛选标记质粒的构建及LL37的表达

为了实现以潮霉素磷酸转移酶基因(hpt)为选择标记的质粒在毕赤酵母中高效表达抗菌肽LL37,在pGAPZαA质粒的基础上,通过EcoRⅤ、BamHⅠ双酶切将Zeocin抗性基因替换成潮霉素磷酸转移酶基因,获得的表达质粒命名为pGAPHαA。根据LL37氨基酸序列,通过毕赤酵母密码子偏嗜性改造,合成的LL37基因片段克隆到pGAPHαA质粒中,获得重组分泌型表达质粒pGAPHαA-LL37,线性化后电击转化毕赤酵母SMD1168,经潮霉素B筛选阳性转化子。72 h表达上清LL37蛋白含量约为167 μg /mL,表达产物耐热性好,对大肠杆菌DH5α、猪链球菌最小抑菌浓度分别为2.94、7.06 μg/mL。     

高抗稻瘟病三系杂交稻恢复系泸恢37的选育及应用

泸恢37是四川省农业科学院水稻高粱研究所利用蜀恢527/多恢57的杂交后代F7优良株系与四川省农科院植保所提供的抗稻瘟病种质资源04R-1051杂交育成的籼型恢复系。泸恢37含有Pikm和Pik-1基因,具有高抗稻瘟病、恢复力和配合力强、制种产量高的特点。泸恢37于2012年8月通过四川省农作物品种审定委员会组织的专家技术鉴定。利用该恢复系配制的组合产量高、抗性强、适应性广,其中,川香优37于2013年通过四川省农作物品种审定委员会审定。     

丰产高糖抗倒甘蔗新品种桂糖37选育研究

桂糖37（原名：桂糖03-2357,亲系：湛蔗92-126×CP72-2086）发芽出苗率好,分蘖多,发株率高,早生快发,前、中期生长快,后期生长势优。中至中大茎、早熟、高糖、高产、宿根性强、实心、抗风抗倒,耐粗放种植。产蔗量为94.88t/hm2,较新台糖22（88.29 t/hm2）增产7.47%;蔗糖分14.32%,与新台糖22（14.44%）蔗糖分相近;含糖量为13.62t/hm2,较新台糖22（12.71 t/hm2）增产7.16%;纤维素含量较新台糖22高约1.5个百分点。     

Therapeutic approaches targeting the prion receptor LRP/LR

Transmissible spongiform encephalopathies known as prion diseases are a group of fatal neurodegenerative disorders that affect both humans and animals. The generally accepted principle of the disease is that the conversion of the cellular prion protein (PrP(c)) into the disease associated isoform PrP(Sc) leads to spongiform degeneration of the brain and amyloid plaque formation. Until now no therapy leading to potential alleviation or even cure of the disease exists. It is therefore important to develop therapeutic approaches for the treatment of TSEs since these infections are inevitably fatal and, especially in the case of vCJD, they affect youngsters. Besides current conventional therapeutic strategies, this review summarizes new therapeutic tools targeting the prion receptor LRP/LR.     

中国对虾血淋巴细胞的原代培养及其与量子点标记的VP37p的作用

血淋巴细胞是对虾白斑综合征病毒(WSSV)靶细胞之一,WSSV结构蛋白与血淋巴细胞的作用在病毒感染中起重要作用。论文主要研究中国对虾血淋巴细胞的原代培养及在原代细胞培养条件下观察重组表达的对虾白斑综合征病毒VP37片段(VP37p)与对虾血淋巴细胞的作用。试验表明,中国对虾血淋巴细胞体外培养可存活12d,体外培养的对虾血淋巴细胞与量子点标记的VP37p具有相互作用。     

结核分支杆菌核酸适配体研究进展

结核分支杆菌（MTB）是引起多种宿主感染结核病的重要病原菌,由于结核分支杆菌耐药菌株的产生,导致结核病在世界范围内呈高发趋势。由于 MTB 检出率较低,开发一种简单快速的诊断方法迫在眉睫。指数富集配体系统进化技术（SELEX）的原理是从随机寡核苷酸文库中筛选到能特异性结合靶物质的寡核苷酸配体（称为适配体）。论文对近几年国内外通过 SELEX 技术对结核分支杆菌筛选出的适配体做一综述,适配体的靶物质分别为分泌蛋白 MPT64,CFP10-ESAT6蛋白,多磷酸激酶2（PPK2）以及结核分支杆菌（H37Rv 株）整个菌体。体外筛选的结核分支杆菌适配体,能对结核病做出早期诊断,并能作为 MTB疫苗的免疫佐剂,在临床诊断及药物研究等方面有很大应用前景。     

小麦VPM1后代Yr17-Lr37-Sr38、Pch1基因分子标记检测

含有偏凸山羊草2NS染色体的普通小麦VPM1因含有3个紧密连锁的抗锈基因簇Yr17-Lr37-Sr38及抗眼斑病Pch1基因被育种者广泛应用。本实验利用Yr17-Lr37-Sr38基因簇的N基因组特异VEN-TRUP/LN2标记及Pch1基因的XustSSR2001-7DL标记对济麦22×VPM1杂交F2代群体的170个单株进行分子标记检测,结果发现含有Yr17-Lr37-Sr38基因簇的有122株,含有Pch1基因的有33株,同时含有Yr17-Lr37-Sr38及Pch1基因的21株。
