AIM: To study the effects of noninvasive delayed limb ischemia preconditioning (NDLIP) on animal cardiac function, myocardial morphology and myocardial apoptosis after myocardial infarction (MI). METHODS: Healthy SD male rats[n=45, weighing (250±10) g] were randomly divided into 3 groups:MI group:the animal model of MI was established by surgical ligation of left anterior descending artery (LAD) after 2 weeks; NDLIP group:after the success of the MI animal model, NDLIP was carried out every other day until the 4th, 6th and 8th weeks; sham group:as the negative control group, the animals were taken heart LAD threading but no ligation. All rats were fed conventionally. At the end of the 4th, 6th and 8th weeks, all rats were made ventricular intubation, and then the hemodynamic parameters were recorded. The blood samples were withdrawn from the abdominal aorta and the serum was separated via centrifugation. The serum contents of Bcl-2 and Bax were measured by ELISA. Left ventricular anterior wall was homogenized. The mitochondrial respiratory chain complexes Ⅰ, Ⅱ, Ⅲ and Ⅳ in the myocardial tissues were detected by ELISA. RESULTS: At the end of the 4th, 6th and 8th weeks, compared with MI group, left ventricular systolic pressure in NDLIP group was significantly increased, while left ventricular end-diastolic pressure in NDLIP group was significantly decreased (both P<0.05). Mitochondrial respiratory chain complexesⅠ, Ⅱ, Ⅲ and Ⅳ in NDLIP group were significantly increased (P<0.05). The serum level of Bcl-2 in NDLIP group was significantly increased and Bax level was reduced remarkably (both P<0.01). CONCLUSION: NDLIP improves the hemodynamic indexes, promotes the mitochondrial respiratory function and inhibits cell apoptosis, thus improving the prognosis of MI.  相似文献

Summary Several independent tests have shown that climate and weather during sexual reproduction influence the adaptive properties of the Picea abies progenies. This phenomenon is expressed in seed orchards established by moving parent trees, propagated as grafts, from north to south, from high to low elevation, or from outdoor to indoor greenhouse conditions. The progenies exhibit delayed flushing in the spring, later growth cessation of leader shoots in the summer, delayed bud-set, higher frequency of lammas shoots and delayed development of frost hardiness in the autumn compared to progenies reproduced in the colder native environment. The altered performance is persistent. We have found no effect on progenies of photoperiod and temperature treatments given to the males during meiosis and pollen production. However, when crosses were made in early spring (March), inside a heated greenhouse (short day, high temperature), the progenies are less frost hardy during cold acclimation than progenies from identical crosses performed in late spring (May; long day high temperature) in the greenhouse. The most hardy offspring were from crosses performed under outdoor conditions in May (long day, low temperature). These results indicate that some stages in reproduction, such as female meiosis, pollen tube growth, syngamy and early embryo development, are sensitive to temperature and/or photoperiod which then alter the phenotypic performance of the offspring. The most likely explanation is the existence of a regulatory mechanism affecting the expression of genes controlling adaptive traits. If this is true, it must have implications for the genetic interpretation of provenance differences in Norway spruce.  相似文献

AIM: To investigate the effects of maternal limb ischemic preconditioning (LIP) on the mitochondrial structures and functions of the hippocampal neurons induced by reoxygenation in the intrauterine distress fetal rats. METHODS: Pregnant rats (n=40) were randomly divided into 4 groups: sham (S) group, LIP group, fetal distress (FD) group and LIP+FD group. Intrauterine ischemia model was established through the experimental design. The ultrastructure of the mitochondria in CA1 area of the hippocampus was observed. The mitochondrial membrane potential and reactive oxygen species (ROS) were measured. The content of ATP and MDA in the hippocampus tissue was detected. The activity of Mn-SOD was observed. RESULTS: Compared with sham group, the ultrastructure of mitochondria in CA1 area of the hippocampus was damaged in FD group and LIP+FD group. The mitochondrial membrane potential, the content of ATP and the activity of Mn-SOD were decreased. However, the content of ROS and MDA was increased. Compared with FD group, the ultrastructure of mitochondria in CA1 area of the hippocampus was intact in LIP+FD group. Furthermore, the reduced mitochondrial membrane potential and ATP content were inhibited. The activity of Mn-SOD was increased, but the content of ROS and MDA was decreased in LIP+FD group. CONCLUSION: Limb ischemia preconditioning inhibits the damage the mitochondria of fetal hippocampal neurons induced by reoxygenation in the intrauterine distress fetal rats.  相似文献

AIM: In order to study the relationship between the ERK and p38 MAPK activation and the protection of 11, 12-epoxyeicosatrienoic acid (11, 12-EET) and ischemia preconditioning (IP), the effects of 11, 12-EET and ischemic preconditioning on phosphorylated ERK and p38 MAPK during ischemia and reperfusion in rat myocardium were examined. METHODS: The rat heart was subjected to ischemia for 5 min by ligating the left anterior descending coronary artery followed by reperfusion for 5 min (two times) to undergo ischemia preconditioning. The rats were divided into 5 groups: (1) control; (2) sham group; (3) ischemia/reperfusion (I/R) group, in which the rat heart suffered from 60 min ischemia followed by 30 min reperfusion; (4) IP plus I/R group; (5) EET plus I/R group, in which 6.28×10-8 mol/L 11, 12-EET was injected intravenously 20 min before I/R. The heart function was examined, and phosphorylated ERK and p38 MAPK were detected by Western blot. RESULTS: At 30 min reperfusion, +dp/dtmax, -dp/dtmax and LVDP decreased significantly in I/R group compared with sham group, IP plus I/R group and EET plus I/R group; Phosphorylated ERK1/2 level was higher in I/R group than sham group, but was lower in I/R group than IP plus I/R group and EET plus I/R group; Phosphorylated p38 MAPK level was lower in control, sham, IP plus I/R and EET plus I/R group than I/R group. CONCLUSION: 11,12-EET protects rat heart against ischemia/reperfusion injury, the mechanism may be related to activation of ERK1/2 and inhibition of p38 MAPK.  相似文献

AIM: To study the protective effect of ischemia preconditioning (IPC) on ischemia/reperfusion (IR)-damaged myocardium in young and old rats. METHODS: Male Wistar rats aged at 3 months (young) and 20 months (old) were used to establish myocardial IPC model and IR model with the method of Langendorff heart perfusion. The rats were divided into young ischemia/reperfusion (YIR) group, young ischemic preconditioning (YPC) group, old ischemia/reperfusion (OIR) group and old ischemic preconditioning (OPC) group. Transmission electron microscopy was used to observe the ultrastructural changes of myocardial tissue and myocardial mitochondria. The myocardial infarction area was determined by TTC staining. The lactate dehydrogenase (LDH) content in coronary effluent fluid and the levels of superoxide dismutase (SOD) and malondialdehyde (MDA) in myocardial tissues were detected by the method of colorimetry. The levels of nitrated and carbonylated proteins in myocardial tissue were measured by ELISA. The myocardial cell apoptosis was analyzed by TUNEL assay. The mitochondrial respiratory function and mitochondrial permeability transition pore opening induced by calcium load were evaluated by oxygen electrode method. RESULTS: Compared with YIR group, the myocardial infarction area in YPC group was obviously smaller, SOD activity in myocardial tissues increased, LDH activity in coronary effluent fluid and the content of MDA decreased, and the levels of nitrated and carbonylated proteins in the cardiac tissues reduced. In YPC group, the mitochondrial membrane structure appeared intact, cristae of the mitochondria showed close arrangement, and the matrix was compressed under the electron microscope. Myocardial mitochondrial respiratory control rate, state Ⅲ oxygen consumption and the P/O ratio in YIR group all significantly increased, proton leak decreased, mitochondrial swelling induced by calcium distinctly reduced, and myocardial apoptosis rate declined. No significant difference of the above indexes between OIR group and OPC group was observed. Compared with YPC group, myocardial ultrastructural damage increased clearly, cardiac oxidative stress increased, mitochondrial respiratory function declined, and cell apoptosis and necrosis increased in OPC group. CONCLUSION: Ischemic preconditioning has protective effect against myocardial IR injury in young rat hearts, while old rat hearts were less sensitive to ischemic preconditioning, leading to bluntness of cardioprotection with IPC in aging hearts. This may be related to mitochondrial injury and severe cellular apoptosis caused by increase of cardiac oxidative stress levels in the aging ischemic preconditioning heart.  相似文献

AIM: To identify the proteins interacted with conventional protein kinase Cγ (cPKCγ) which are involved in hypoxic preconditioning (HPC), and to investigate the role of cPKCγ-interacted heat-shock protein 60 (HSP60) in the development of HPC and ischemia (I).METHODS: Healthy male BALB/c mice were randomly divided into normoxia(Norm) and HPC groups. Based on whether middle cerebral artery occlusion (MCAO) was performed, the mice were divided into Norm+sham group, Norm+I group, HPC+sham group and HPC+I group ( all n=6). Immunoprecipitation, two-dimensional electrophoresis and mass spectrometry were applied to identify the proteins interacted with cPKCγ. The changes of HSP60 expression in the brain of the mice after HPC and MCAO were analyzed by Western blotting.RESULTS: The interaction of cPKCγ and HSP60 was confirmed by co-immunoprecipitation result. Compared with Norm groups, the expression level of cPKCγ-interacted HSP60 obviously increased in particulate fraction of cerebral cortex in HPC mice. In the MCAO ischemic animals, the level of HSP60 expression was significantly higher in ischemic core and penumbra in Norm+I group and HPC+I group than that in Norm+sham group. HSP60 expression in ischemic core was lower in HPC+I group than that in Norm+I group.CONCLUSION: cPKCγ-HSP60 signal pathway might be involved in the development of cerebral hypoxic preconditioning in MCAO mice.  相似文献

本研究以未进行低氧预适应的魁蚶(Scapharca broughtonii)为对照组(C组),分析了2次低氧预适应(H2组)和4次低氧预适应(H4组)的魁蚶在溶解氧(DO)约为2.0 mg/L低氧胁迫48 h内的摄食、呼吸代谢和酶活力的变化规律。结果显示,3组魁蚶的摄食率(IR)在胁迫初期急剧下降,后期均随时间的延长逐渐恢复,至48 h时,H组恢复程度显著高于C组(P<0.05=;C组、H2组和H4组魁蚶的耗氧率(OR)随时间变化呈逐渐升高的趋势,48 h比0 h分别提高了1.15、1.08、0.73倍;3组排氨率(NR)表现出不同的变化趋势,至48 h时,C组、H2组和H4组分别为0 h的1.67、1.30、0.97倍;C组的氧氮比(O/N)相对平稳,H组的变化范围相对较大。3组的细胞色素C氧化酶(COX)随着低氧胁迫时间的延长呈逐渐降低的趋势,乳酸脱氢酶(LDH)活力和还原型谷胱甘肽酶(GSH)含量整体呈上升趋势,与对照组相比,预低氧组的酶活力在低氧胁迫期间变化相对平稳,应激反应小。研究表明,魁蚶经低氧预适应后再次受到低氧胁迫时,IR升高,OR降低,酶活性相对稳定,低氧预适应能提高魁蚶的耐低氧能力。本研究丰富了魁蚶低氧耐受相关研究的基础数据,为进一步探讨魁蚶低氧耐受机制和创制耐低氧新种质提供了参考资料。  相似文献

AIM: To examined the effects of hypoxic preconditioning(HPC) on oxygen-glucose deprivation(OGD)-induced PC12 cells, and to investigate its possible mechanisms of autophagy.METHODS: Cultured PC12 cells were randomly divided into control group, HPC group, 3-methyladenine(3-MA) group, HPC+OGD group, 3-MA+HPC+OGD group and OGD group. CCK-8 assay was used to detect the cell viability. The caspase-3 activity was also tested. TUNEL staining and flow cytometry were used to detect the cell apoptosis. The protein levels of apoptosis-related protein caspase-3 and autophagy-marked protein LC3-2 and beclin-1 were determined by Western blot.RESULTS: Compared with control group, the viability of PC12 cells was significantly reduced, and the activity of caspase-3 was significantly increased in OGD group. Compared with 3-MA+ HPC+OGD group and OGD group, the viability of PC12 cells was significantly increased, and the activity of caspase-3 was significantly reduced in HPC+OGD group(P<0.05). The PC12 cell injury was apparent after OGD with a great increase in the apoptotic rate(P<0.05). Compared with OGD group, the apoptotic rate significantly decreased in HPC+OGD group(P<0.05). Compared with control group, the protein level of cleaved caspase-3 was significantly increased in OGD group(P<0.05). Compared with OGD group, the protein level of cleaved caspase-3 was significantly decreased, and the levels of LC3-2 and beclin-1 were significantly increased in HPC+OGD group(P<0.05).CONCLUSION: OGD decreases cell survival and induces apoptosis.Activation of cell autophagy may be the mechanism by which hypoxic preconditioning protects the PC12 cells from OGD induced injury.  相似文献
