The influence of co‐infection on concentration and accumulation of genetically different isolates of Potato virus Y (PVY) in potato and tobacco plants and the efficiency of transmission by Myzus persicae of PVY isolates from doubly versus singly infected plants were evaluated. The vector ability to simultaneously transmit two virus isolates was examined. Eight PVY isolates represented three strain groups: PVYO (pathotype and serotype O), PVYNW (pathotype N and serotype O), and PVYNTN (pathotype and serotype N). Different diagnostic methods, including DAS‐ELISA, multiplex RT‐PCR, aphid transmission tests and bioassays, were applied to detect the presence of PVY isolates in source and assay plants. Significant reductions in concentrations of certain PVY isolates during co‐infection with other isolates were found both in potato and tobacco plants. The observed effects were both isolate‐ and host‐dependent in form. The highest rates of virus transmission by single aphids were recorded with PVYNTN isolates, and the lowest ones with PVYO isolates. Individual aphids of M. persicae were able to simultaneously transmit two PVY isolates. The frequency of transmission was generally low, but it reached as high as 20% for one of the isolate combinations. The findings presented in the work provide proof for antagonistic within‐plant interactions between isolates of PVY, with some implications of these interactions for virus transmission by aphid vectors. Consequently, this research contributes to a better understanding of the epidemiology of the disease caused by PVY.  相似文献

The impact of ectomycorrhizal fungi or rhizosphere bacteria on tree seedling growth and nutrient uptake is well known. However, few studies have combined those microorganisms in one experiment to clarify their relative contribution and interactions in nutrient acquisition. Here, we monitored the respective contributions of pine roots, two ubiquitous forest ectomycorrhizal fungi Scleroderma citrinum and Laccaria bicolor, and two S. citrinum-mycorrhizosphere bacterial strains of Burkholderia glathei and Collimonas sp., on mineral weathering, nutrient uptake, and plant growth. Pinus sylvestris plants were grown on quartz–biotite substrate and inoculated or not with combinations of mycorrhizal fungi and/or bacterial strains. Magnesium and potassium fluxes were measured and nutrient budgets were calculated. Both ectomycorrhizal fungi significantly increased Mg plant uptake. No significant effects of the two bacterial strains were detected on the K and Mg budgets, but co-inoculating the mycorrhizal fungus S. citrinum and the efficient mineral-weathering B. glathei bacterial strain significantly improved the Mg budget. Similarly, co-inoculating S. citrinum with the Collimonas sp. bacterial strain significantly improved the pine biomass compared to non-inoculated pine plants. In order to better understand this process, we monitored the survival of the inoculated bacterial strains in the quartz–biotite substrate, the pine rhizosphere, and the mycorrhizal niche. The results showed that the two bacterial strains harboured different colonization behaviours both of which depended on the presence of the ectomycorrhizal partner. The populations of the Burkholderia strain were maintained in all these environments with a significantly higher density in the mycorrhizal niche, especially of S. citrinum. In contrast the population of the Collimonas strain reached the detection level except in the treatment inoculated with S. citrinum. These results highlight the need for taking into account the ecology of the microorganisms, and more specifically the fungal–bacterial interactions, when studying mineral weathering and plant nutrition.  相似文献

Lotus japonicus was used to study the distribution and interconnections of 15 elements in plant tissues, including essential and non-essential elements: boron (B), sodium (Na), magnesium (Mg), potassium (K), calcium (Ca), manganese (Mn), iron (Fe), nickel (Ni), copper (Cu), zinc (Zn), arsenic (As), strontium (Sr), molybdenum (Mo), cadmium (Cd) and cesium (Cs). Large amounts of B and Ca accumulated in plant leaves, while Fe, Na, Ni, As and Cd tended to mainly occur in the roots, and Mo was the only element to accumulate in the stems. The elemental compositions within plants were severely disturbed by treatment with toxic elements. Competition between element pairs in the same group (e.g. K and Cs; Ca and Sr) was not found. Iron, Cu and Zn accumulation were induced by Cd and Ni addition. When natural variants grew in a nutrition solution with subtoxic levels of As, Cd, Cs, Ni, Mo and Sr, intriguing relationships between the elements (such as Fe, As and K; Mg and Ni; Mn and Ca) were revealed using principal-component analysis. This study on the plant ionome offers detailed information of element interactions and indicates that chemically different elements might be closely linked in uptake or translocation systems.  相似文献

Tropical insect species show year-round breeding activity due to favourable climatic conditions. However, most species also display seasonal reproductive peaks, but little is known about underlying causes of temporal density changes. We investigated population dynamics of the pest predator Rhynchium haemorrhoidale (F.) (Hymenoptera: Eumenidae) and its natural enemies in relation to season, climate and varying shade tree composition in cacao agroforestry systems in Central Sulawesi (Indonesia). Nesting of R. haemorrhoidale showed clear seasonality with highest densities in the wet season and lowest in the dry season, which was not related to changes in temperature. Wasp densities increased with land-use intensity, presumably because less-shaded areas offer more favourable climatic conditions and higher densities of the major prey, the cacao pest Agathodes caliginosalis (Lepidoptera: Pyralidae). Pupal body size was higher in June than in November indicating increasing intraspecific competition due to high wasp densities and food shortage at the beginning of the wet season. Body size between habitat types was similar, thus season appeared to be more important than habitat in terms of food supply. High wasp densities in the wet season were associated with high diversity of the altogether seven parasitoid species. In conclusion, we found a pronounced seasonality of the wasp R. haemorrhoidale and a preference for little shaded agroforestry, which may be due to enhanced temperature and density of its prey. Our data indicate a high potential for biological control of a major cacao pest by management of nesting sites of R. haemorrhoidale.  相似文献

Anadromous Arctic charr, Salvelinus alpinus (L.), was introduced to a sub‐Arctic river–lake system near the village of Kujjuuaq, Nunavik, and the stable isotope values and diets of key resident fish species were used to assess changes in feeding patterns. Stable isotope values for most species did not differ significantly between the pre‐ and post‐introduction periods, with observed shifts being within the bounds of expected natural variation. Lake chub, Couesius plumbeus (Agassiz), were the single species to show a difference between study periods, with a small but significant increase in δ15N. No significant post‐introduction changes were seen in lake trout, Salvelinus namaycush (Walbaum), omnivory or in any of the assessed quantitative food web metrics. Gut contents of major fish species similarly showed significant temporal overlap between the pre‐ and post‐introduction periods, and there was no significant change in species' weight–length relationships. The minor ecological impact was interpreted in relation to the availability of open niches exploitable by ecological generalists such as Arctic charr. The explanation accords with the known habitat and feeding flexibility of Arctic charr and the ecological immaturity of sub‐Arctic lakes known to have driven adaptive variation among Arctic charr. Findings suggest that anadromous Arctic charr may be introduced at moderate densities to other sub‐Arctic watersheds without major negative food web consequences for other resident fish species.  相似文献

Sclerotinia soft rot, caused by Sclerotinia sclerotiorum, is a severe disease of cultivated carrots (Daucus carota ssp. sativus) in storage. It is not known whether Sclerotinia soft rot also affects wild carrots (D. carota ssp. carota), which hybridise and exchange genes, among them resistance genes, with the cultivated carrot. We investigated the susceptibility of wild carrots to S. sclerotiorum isolates from cultivated carrot under controlled and outdoor conditions. Inoculated roots from both wild and cultivated plants produced sclerotia and soft rot in a growth chamber test. Two isolates differed significantly in the ability to produce lesions and sclerotia on roots of both wild carrots and cv. Bolero. Flowering stems of wild carrots produced dry, pale lesions after inoculation with the pathogen, and above-ground plant weight was significantly reduced 4 weeks after inoculation in a greenhouse test. Wild and cultivar rosette plants died earlier and fewer plants survived when inoculated with the pathogen under outdoor test conditions. Cultivar plants died earlier than wild plants, but survived as frequently. Plants inoculated in the crown died earlier and at a lower frequency than plants inoculated on leaves. Wild carrots may thus serve as a host of S. sclerotiorum and thus eventually benefit from any uptake of resistance genes, among them transgenes, via introgression from cultivated carrots.  相似文献

In Cameroon, seed cotton yields have not increased over the last 20 years because of the shortening of the rainy season and the worsening socioeconomic context. Farmers consequently often delay planting their crops. The main objective of this study was to investigate whether local indeterminate long-season cottons, grown at the recommended density, were more consistent with the farmers’ current constraints than determinate short-season cultivars from Latin America that could be sown more densely. We carried out a 3-year three-location survey in northern Cameroon, which included two planting dates (recommended and delayed) and two planting densities (recommended and high). We show that these three factors acted independently. Late planting had a highly negative impact on most traits at both plant and plot scale by delaying flowering, reducing seed cotton yield and fibre quality. Dense sowing mainly had an impact on individual plant traits by reducing boll retention and elongating main-stem internodes. Local cultivars have already evolved favourably (enhanced earliness, yield performance, harvest index, ginning out-turn, and fibre maturity) and could be improved further by crossing with highly determinate cottons. However, such a strategy requires further investigation to ensure that a more determinate growth pattern would not have a negative impact on the adaptive response of the traditional cotton plant to other adverse conditions.  相似文献
