Based on an analysis of their reactivity with porcine peripheral blood lymphocytes (PBL), only three of the 57 mAbs assigned to the T cell/activation marker group were grouped into cluster T9 along with the two wCD8 workshop standard mAbs 76-2-11 (CD8a) and 11/295/33 (CD8b). Their placement was verified through the use of two-color cytofluorometry which established that all three mAbs (STH101, #090; UCP1H12-2, #139; and PG164A, #051) bind exclusively to CD8+ cells. Moreover, like the CD8 standard mAbs, these three mAbs reacted with two proteins with a MW of 33 and 35 kDa from lymphocyte lysates and were, thus, given the wCD8 designation. Because the mAb STH101 inhibited the binding of mAb 76-2-11 but not of 11/295/33, it was given the wCD8a designation. The reactivity of the other two new mAbs in the T9 cluster with the various subsets of CD8+ lymphocytes were distinct from that of the other members in this cluster including the standards. Although the characteristic porcine CD8 staining pattern consisting of CD8low and CD8high cells was obtained with the mAb UCP1H12-2, a wider gap between the fluorescence intensity of the CD8low and CD8high lymphocytes was observed. In contrast, the mAb PG164A, not only exclusively reacted with CD4/CD8high lymphocytes, but it also failed to recognize CD4/CD8 double positive lymphocytes. It was concluded that this mAb is specific for a previously unrecognized CD8 epitope, and was, thus, given the wCD8c designation. A very similar reactivity pattern to that of PG164A was observed for two other mAbs (STH106, #094; and SwNL554.1, #009). Although these two mAbs were not originally positioned in the T cell subgroup because of their reactivity and their ability to inhibit the binding of PG164A, they were given the wCD8c designation. Overall, five new wCD8 mAbs were identified. Although the molecular basis for the differences in PBL recognition by these mAbs is not yet understood, they will be important in defining the role of CD8+ lymphocyte subsets in health and disease.  相似文献

Abstract

To increase the accuracy of remotely sensed data for agricultural forecasting, pixel values must be corrected for atmospheric effects and converted to spectral reflectance. The objective of this research was to compare two atmospheric correction methods of Landsat imagery under a range of atmospheric conditions. Ground‐based dark‐object subtraction (GDOS) is an image‐based calibration method that used in situ ground data that the dark‐object subtraction (DOS) method did not use, whereas atmospheric calibration (AC) is a model‐based calibration method that required a standard atmospheric profile refined with the use of in situ atmospheric data. GDOS and AC methods improved the reflectance values and had relationships with measured bands, which were approximately 1 to 1 in all bands. However, the GDOS generally had lower root‐mean‐square errors (RMSE) than AC. Data from this study suggest that at the present time the GDOS method may be more accurate than the AC method.  相似文献

This article presents a validation of a method, proposed in Podlaski (2003), for estimating the degree of tree damage. This method belongs to a group of small-area methods. To estimate the degree of tree damage in individual P3 sub-blocks of the SINUS system of information on the natural environment, a classification based on the degree of defoliation was used. Spatial distribution of the degree of tree damage was estimated through unrestricted simple random sampling. The maximum total estimation error for proportions of fir and beech with a damage degree of zero, and also with second and third degrees jointly, reached a maximum of 35.6% for fir, and 33.8% for beech. P3 sub-blocks characterised by the highest proportion of the healthiest trees (at least 15% being undamaged trees) occurred in patches in the case of fir, and in larger, compact areas in the case of beech. Because spatial diversification in the health of fir, and to a smaller degree in the case of beech, occurs at small scales, small-area methods for estimating tree condition are necessary. An analogous scheme, combining small-area with large-area methods (utilising sub-blocks of the network of large-area systems in a small-area inventory, and establishing a common database), could be used on a wider scale; for example, by supplementing the program of estimating tree health carried out by the United Nations Economic Commission for Europe (Lorenz et al. 2002) with local inventories.  相似文献

To investigate the biosynthesis and stereochemistry of syringylglycerol-8-O-4′-(sinapyl alcohol) ether (SGSE), a syringyl 8-O-4′ neolignan, feeding experiments and enzyme assays using Eucommia ulmoides were carried out. Diastereoselective formation of erythro-SGSE was found. When [8-14C]sinapyl alcohol was administered to excised shoots of E. ulmoides, 14C was incorporated into free SGSE and SGSE glucosides. In stems, incorporation into (+)-erythro-[14C]SGSE (0.037%) with 9.1% enantiomeric excess (% e.e.) was found; incorporation into the threo isomer was not detectable. Erythro-[14C]SGSE glucosides (0.047%) dominated over threo forms (0.007%) with 74.0% diastereomeric excess (% d.e.); both diastereomers were levorotatory with 32.0% e.e. and 18.3% e.e., respectively. In leaves, higher incorporation into (−)-erythro-[14C]SGSE (0.500%, 15.9% e.e.) than into the threo isomer (0.206%, 7.4% e.e.) was observed (41.6% d.e.). (−)-Erythro-[14C]SGSE glucosides (1.692%, 25.0% e.e.) were produced at higher rates than threo isomers (0.177%, 16.4% e.e.) with 81.0% d.e. In incubations of a mixture of [8-14C]sinapyl and [8-14C]coniferyl alcohols with an insoluble enzyme preparation from stems of E. ulmoides, erythro-SGSE was preferentially produced. The highest % d.e. Taking advantage of the introduction of the black-tailed deer to the Queen Charlotte Islands (British Columbia, Canada), we used dendrochronological analyses to understand the consequences of deer browsing on Sitka spruce growth. We compared shape, radial growth, height growth and age of young spruce in three sites. We identified two types of trees growing side by side: (1) stunted and heavily browsed spruce, smaller than the browsing limit and (2) escaped spruce that were taller than the browsing limit but still browsed in their lower part. The compact and heavily ramified shape in stunted spruce was the result of repeated and intense browsing. In escaped spruce this was also the case below the browsing limit (1.16 m±0.07 m), in sharp contrast with the normal shape that escaped spruce resumed above the browsing limit. We show that the release of browsing pressure, once the tree reaches the browsing limit, is characterised by an abrupt increase in radial growth. Before release, trees show a growth stagnation characterized by narrow rings (0.5 mm per year) and small annual height increments (<5 cm per year). After release, trees show a growth stabilisation characterised by wider rings (3 mm per year) and larger annual height increments (20 cm per year). We use this pattern to estimate frequency and age at release and their possible variation over time. Age differences between stunted and escaped spruce are highly significant and indicate that, despite of browsing, most if not all trees will ultimately reach the browsing limit and escape. Heavy deer pressure (30 deer per km2) delays spruce sapling recruitment by about 8 years. This delay varies in relation to site quality and seems to have increased over time, suggesting an increase in browsing pressure.  相似文献

Sugi (Cryptomeria japonica D. Don) wood was treated with supercritical water (374°C, 22.1MPa), and fractionated into the water-soluble portion, the methanol-soluble portion, and the methanol-insoluble residue. The methanol-soluble portion mainly consisted of the lignin-derived products. To characterize the compounds in the methanol-soluble portion, gel permeation chromatographic (GPC) and gas chromatographic-mass spectrometric (GC-MS) analyses were performed. The GPC analysis indicated that the methanol-soluble portion contained lignin-derived monomeric and dimeric products. GC-MS analysis detected 31 products which were expected to be monomeric compounds, and 18 of these were identified to be guaiacol, methylguaiacol, ethylguaiacol, vinylguaiacol, eugenol, propylguaiacol, vanillin, cis-isoeugenol, homovanillin, trans-isoeugenol, acetoguaiacone, propioguaiacone, guaiacylacetone, 2-methoxy-4-(1-hydroxypropyl)phenol, homovanillic acid, 2-methoxy-4-(prop-1-en-3-one)phenol, coniferyl aldehyde, and ferulic acid. In addition, 22 dimeric products were detected, and 4 of these were believed to be compounds with biphenyl type (5-5), diphenylethane type (-1), stilbene type (-1), and phenylcoumaran type (-5) structures. These results clearly indicated that the methanol-soluble portion included various monomeric and dimeric compounds produced as a result of the cleavage of ether linkages and propyl chains of lignin.  相似文献
