小麦苗枯病菌(Clavibacter fangii,Cf)是引起小麦细菌性苗枯病的病原,本研究用16S~23S rDNA间的内源转录间隔区(internally transcribed spacer,ITS)序列通用引物L1(5'-AGTCGTAACAAGGTAGCCGT-3')和L2(5'-GTGCCAAGGCATCCACC-3')扩增Cf和其它相关细菌的基因组DNA;并对其PCR产物进行回收、克隆和测序,将所获序列和其它已报道的细菌ITS序列进行多重比较后设计出Cf的特异性引物I1(5'-TGCCAAGTCACACTGAGACGA-3')和I2(5'-CAATGATCTACCACCCTCCGA-3')。此引物可以从Cf中扩增出351bp的特异性片段,而其余参试的21个细菌PCR反应结果均为阴性。该方法可以应用于小麦苗枯病菌的快速、可靠检测。此外,本研究对多种植物病原棒形杆菌的ITS序列进行比较研究,发现其具有一定的分类意义。 相似文献

The Pocono mesic till barrens (PMTB) are a unique assemblage of fire-maintained shrub communities that support numerous rare species. Historically these barrens covered a large area in the vicinity of Long Pond, Pennsylvania, USA. However, due largely to regional fire suppression instituted in the early 1960s, over 70% of the area covered by barrens succeeded to fire-intolerant forest that does not support the rare species. We investigated the influence of forest proximity on barrens succession across three geomorphic types during periods of high fire frequency and fire suppression, testing the hypothesis that forest processes such as seed rain, shading, and detrital enrichment of soils enhances barrens succession through a contagion effect. Evidence of a forest contagion effect should be shown by increased rates of barrens succession with increasing proximity to the nearest forest edge. In order to detect a forest contagion effect, barrens persistence and barrens succession were modeled in proximity zones of 0-50 m, 50-100 m, 100-200 m, and greater than 200 m from the nearest forest edge. We used existing GIS data layers for fire, geomorphology, and vegetation distribution in 1938, 1963, and 1992. The layers were modified and overlain using ArcView software to determine persistence and succession rates for each unique combination of layers in each proximity zone from 1938 to 1963 (pre-fire suppression) and 1963 to 1992 (post-fire suppression). ANCOVA results indicate that proximity to the nearest forest edge significantly affected barrens persistence rates in both time periods, but succession rates were significantly affected in 1938 to 1963 only. Twenty-eight percent of the 1938 barrens succeeded to forest by 1963; 56% of the 1963 barrens became forest by 1992. Results support previous findings that barrens persistence is enhanced by increased fire frequency, and that barrens persist longer where they overlie flat glacial till than on other geomorphology types. 相似文献

ABSTRACT1. The objective of this study was to develop a machine vision method for analysing exterior parameters of chicken eggs to automate the stage of primary sorting.2. The developed algorithm based on predetermined thresholds calculated egg quality indicators, including geometric dimensions, shape index and the mottling grade. The algorithm was implemented with an experimental setup that combined the image-based and the candling methods. A total of 400 egg samples were analysed.3. Comparison of results of the algorithm with those obtained using the traditional manual method showed that mean value of radii values difference was 0.095 ± 0.058 mm for the sharp and 0.080 ± 0.047 mm for the blunt end of the egg, with standard deviations of 0.58 mm and 0.49 mm, respectively.4. The correlation coefficient between the shape index values determined by the two methods was 0.93; the standard deviation of absolute differences between corresponding values was 1.05%.5. The results of mottling grade estimation were compared using F-measure and confusion matrix.6. [Objective] The aim of this study was to obtain the flanking sequences of T-DNA in the transgenic cotton containing a GbVe1 over-expression cassette. [Method] The T-DNA insertion copy number in the transgenic GbVe1 cotton was analyzed by southern blot. Flanking sequences of the transgenic lines with putative single T-DNA insertion copy were obtained using high-efficiency Thermal asymmetric interlaced polymerase chain reaction (hiTAIL-PCR). The T-DNA insertion sites were further confirmed by PCR with specific primers. [Result] RB-flanking sequences (119-1 018 bp) and LB-flanking sequences (243-516 bp) were obtained from three transgenic lines with low copy number of T-DNA insertion. The AT content was more than 63% in these flanking sequences. A same single insertion site in the intron of Gohir.D01G157600.1 was found in the two transgenic lines 7/100826-152 and 12/100826-393, while two separated insertion sites, one also in the intron of Gohir.-D01G157600.1 and the other in the intergenic region of A12 chromosome, were found in the transgenic line 1/w-ch14. A deletion of 21 bp was found in the insertion site in the intron of Gohir.D01G157600.1. The T-DNA insertion in the intron of Gohir.D01G157600.1 was further confirmed by the specific PCR. [Conclusion] The flanking sequences of T-DNA in the transgenic GbVe1 cotton were obtained and the specific transformation event in the intron of Gohir.D01G157600.1 was further confirmed by PCR. 相似文献
