为探讨不同抗精神分裂症药物对吐鲁番斗鸡攻击行为影响的分子机理,试验将36只斗鸡随机分为12组,并选取利培酮、阿立哌唑、盐酸曲唑酮、盐酸舍曲林4种抗精神分裂症药物,且每种药物设有低剂量组(L)、正常剂量组(N)及高剂量组(H),每天人工口服给药。分别在第1、2、5、8、11、14天时,翅下静脉采血,并采用实时荧光定量PCR法对口服不同剂量药物的吐鲁番斗鸡各阶段血液中5-羟色胺转运体(5-hydroxy-tryptamine transporter,5-HTT)、色氨酸羟化酶1(tryptophan hydroxylase 1,TPH1)的mRNA表达量进行检测。结果显示,口服4种药物后,利培酮组和阿立哌唑组斗鸡血液中5-HTT、TPH1基因mRNA的表达变化具有相似性,5-HTT基因整体呈下降趋势,TPH1基因均表现为波动性上升,且利培酮组TPH1基因mRNA的表达量及上升幅度高于阿立哌唑组;盐酸曲唑酮组和盐酸舍曲林组5-HTT基因mRNA的表达均从第5天开始呈直线上升,并于第14天达到峰值,且盐酸舍曲林组TPH1基因的表达呈微上调,盐酸曲唑酮组则无明显变化。利培酮组和阿立哌唑组斗鸡攻击行为减弱与其血液中5-HTT基因mRNA的低表达有关,本试验结果为进一步研究药物影响吐鲁番斗鸡攻击行为的分子机理提供了重要参考依据。 相似文献

为选择适合在高海拔,寒冷地区推广的多年生混播牧草品种,采用近年培育出的甘农1号杂花苜蓿与碱茅,在肃南县进行了混播试验,结果表明,高寒地区碱茅与甘农1号杂花苜蓿混播有较强的抗寒性和越冬能力,碱茅与苜蓿以7:4比例混播,可获得最佳产草量和牧草品质。 相似文献

根据已报道的堆型艾美耳球虫(Eimeria acervulina)子孢子表面抗原基因cSZ1的cDNA序列设计特异性引物,以孢子化12h的E.acervulina广东株卵囊的总RNA为模板,用RT-PCR方法成功克隆了其子孢子表面抗原基因cSZ1(cSZ1(Gd))的cDNA。所克隆cSZ1(Gd)的cDNA全长940bp,其中第3~512位是其阅读框架,共编码170个氨基酸,两端为非编码区。与已报道的cSZ1基因的cDNA序列相比,cSZ1(Gd)有4个位置的核苷酸发生了变异,即原序列中第294、443、569、586位的G、G、G、A在cSZ1(Gd)分别为A、A、A、G,二者核苷酸序列的同源性为99.57%(936/940),其中阅读框架的核苷酸同源性为99.61%(508/510)。比较根据核苷酸序列推导的氨基酸序列,第294位的变异导致了所编码氨基酸由缬氨酸(V)变为异亮氨酸(Ⅰ),而第443位的变异则为无义突变,氨基酸序列的同源性为99.41%(169/170)。 相似文献

Highly pathogenic influenza A virus subtype H5N1 causes significant poultry mortality in the six countries where it is endemic and can also infect humans. Egypt has reported the third highest number of poultry outbreaks (n = 1084) globally. The objective of this cross-sectional study was to identify putative risk factors for H5N1 infections in backyard poultry in 16 villages in Damietta, El Gharbia, Fayoum, and Menofia governorates from 2010–2012. Cloacal and tracheal swabs and serum samples from domestic (n = 1242) and wild birds (n = 807) were tested for H5N1 via RT-PCR and hemagglutination inhibition, respectively. We measured poultry rearing practices with questionnaires (n = 306 households) and contact rates among domestic and wild bird species with scan sampling. Domestic birds (chickens, ducks, and geese, n = 51) in three governorates tested positive for H5N1 by PCR or serology. A regression model identified a significant correlation between H5N1 in poultry and the practice of disposing of dead poultry and poultry feces in the garbage (F = 15.7, p < 0.0001). Diarrhoea caused by enterotoxigenic Escherichia coli (ETEC) expressing F4 (F4ab, F4ac and F4ad) fimbriae is a significant cause of mortality and morbidity in newborn and weaned pigs. The locus controlling susceptibility towards ETEC F4ab/ac has been mapped to SSC13q41, in which TFRC (transferrin receptor) was localized and considered as a positional candidate gene for ETEC F4ab/ac receptor. In this study, we determined susceptibility/resistance to ETEC F4ab/ac in a total of 755 F2 animals from a White Duroc x Erhualian intercross using a microscopic enterocyte adhesion assay. We identified two TFRC polymorphisms (SNPs 591 A>G and 632 A>G) in a single exon after comparative sequencing analysis of 2371-bp amplicons containing the complete coding region of TFRC using RNA of eight full-sib F2 animals with susceptible and resistant phenotypes. The intron sequences flanking the two exon polymorphisms were obtained, revealing an intron polymorphism (SNP 291 C>T). We genotyped the 19 founder animals of the White Duroc x Erhualian intercross for the identified polymorphisms, showing that only the 291 C>T polymorphism is a highly informative marker. We further genotyped all 59 F1 and 755 F2 animals for the 291 C>T polymorphism, and the association of this polymorphism with susceptibility/resistance to ETEC F4ab/ac in these F2 animals was evaluated by the transmission disequilibrium test. The result showed that the 291 C>T polymorphism is not a causal mutation, however, has a significant linkage disequilibrium with the ETEC F4ab/ac, especially F4ac receptor locus. 相似文献

The majority of studies on the immunobiology of Trichinella species have centred on the larval muscular phase (L1) with a view to identifying immunodominant antigens located on the surface of the cuticle and in the larval secretions; the nucleus of the parasite-host interaction. These antigens have been classified as eight groups (TSL-1-TSL-8), of which those belonging to the group TSL-1 have been most intensely studied. The principal constituents are glycoproteins, glycan carriers that contain a unusual sugar, the tyvelose (3,6-dideoxy-d-arabinohexose). Studies aimed at improving serodiagnostic techniques to detect trichinellosis indicate that these antigens are ideal candidates. They are capable of inducing a strong humoral response involving the generation of specific antibodies against beta-tyvelose, a sugar that seems to be exclusive to the Trichuroidea. Furthermore, these glycoproteins appear to fulfil an important function in the development and maintenance of the parasite in the muscular niche, and they appear to be fundamental for the invasion of the intestinal epithelium. It has also been demonstrated that specific monoclonal antibodies against tyvelose can mediate a degree of immunoprotection in the rat through the phenomenon known as rapid expulsion. 相似文献

本文采用6级Andersen生物空气采样器和常用真菌计数培养基,在鸡舍入口处收集气溶胶真菌,经培养和纯化鉴定出78种真菌,其中包括黄曲霉、烟曲霉、黑曲霉及镰刀菌等在内的50余种真菌。研究表明鸡舍入口处空气真菌污染程度与鸡舍内没有显著差异。早晨气溶胶真菌浓度明显低于中午(P=0.07)和晚间(P=0.05)的浓度,且夏秋季节浓度相对较高,峰值在7月份。鸡舍内气溶胶真菌的年平均浓度为2.3×103 CFU/m3。黄曲霉、烟曲霉、黑曲霉在鸡舍空气中的浓度相对较高,分别超过真菌气溶胶组成平均浓度77.7 CFU/m3、63CFU/m35、7.1 CFU/m3,而镰刀菌是常见的空气真菌,其总平均浓度为74.6 CFU/m3,比平均值高出45.3 CFU/m3。平均CMD值分别为3.1μm;GSD分别为2.1。每分钟可沉积到支气管和直接侵入肺泡的活性真菌量分别是居室的3.4倍和3.3倍。本研究探讨了真菌气溶胶不同时间和季节的变化规律,并从其组成、浓度及粒子大小定量评估采样点鸡舍真菌气溶胶的危害,为有效控制禽类真菌病的流行提供科学依据和预警资料。 相似文献

The aim of this study was to evaluate single and double centrifugation tube methods for concentrating equine platelets. Whole blood samples were collected from clinically normal horses and processed by use of single and double centrifugation tube methods to obtain four platelet concentrates (PCs): PC-A, PC-B, PC-C, and PC-D, which were analyzed using a flow cytometry hematology system for hemogram and additional platelet parameters (mean platelet volume, platelet distribution width, mean platelet component concentration, mean platelet component distribution width). Concentrations of transforming growth factor beta 1 (TGF-beta(1)) were determined in all the samples. Platelet concentrations for PC-A, PC-B, PC-C, and PC-D were 45%, 44%, 71%, and 21% higher, respectively, compared to the same values for citrated whole blood samples. TGF-beta(1) concentrations for PC-A, PC-B, PC-C, and PC-D were 38%, 44%, 44%, and 37% higher, respectively, compared to citrated whole blood sample values. In conclusion, the single and double centrifugation tube methods are reliable methods for concentrating equine platelets and for obtaining potentially therapeutic TGF-beta(1) levels. 相似文献
