OPAY02型2条多态性条带经克隆、测序和引物设计后,转换成SCAR标记,并对86个新扬州鸡随机交配后代基因组DNA进行了PCR扩增.2条带DNA序列与红色原鸡基因组序列比对结果表明,大分子量条带与位于红色原鸡第3号染色体上序列有98%的同源性,共检测到8个SNPS,其中195位的碱基T→G,316位的A→T,538位的G→A,731位的T→A,1 147位的G→A,1 329位的T→C,1 927位的C→T,2 081位的C→T,小分子量条带与红色原鸡没有同源序列,推测新扬州鸡野祖除红色原鸡外,还有其它来源.SCAR标记分析表明,经条件优化随机扩增的OPAY02型标记稳定、可靠,可用于遗传分析.2条带所在座位群体基因型平衡性测验结果表明,所测新扬州鸡群体处于平衡状态,选择可以打破平衡,有利于动物育种.  相似文献

An experiment was conducted with Saanen goats fed fresh grass ad libitum to compare 51Cr-mordanted fibre and even-chain alkanes sprayed onto either grass leaves or stems to estimate faecal output, total mean retention time (TMRT) and parameters obtained from faecal marker concentration curves, such as k1 (slow rate of passage), k2 (fast rate of passage) and transit time (TT). 51Cr-mordanted grass showed the lowest fractional rates of passage (k1 and k2) and hence the largest value of TMRT. There were not significant (P > 0.05) differences between even-chain alkanes sprayed onto leaves or stems for k2, TT and TMRT, but k1 estimates were higher (P < 0.05) for stems than for sprayed leaves. Despite the marker used, TMRT values were negatively correlated with the level of dry matter intake (r = − 0.81, − 0.80 and − 0.80 for 51Cr-mordanted fibre and even-chain alkanes adsorbed onto leaves or stems, respectively). Average faecal outputs estimated from faecal concentrations of 51Cr-mordanted fibre and even-chain alkanes were not different from the actual outputs but there were differences between markers in the accuracy of estimation. The highest mean square prediction error (MSPE) and the poorest correlation between observed and estimated faecal output values corresponded to even-chain alkanes adsorbed onto stems. Values estimated using 51Cr-mordanted fibre and even-chain alkanes adsorbed onto leaves were significantly correlated with faecal outputs (r = 0.94 and r = 0.92, respectively), with MSPE being greater for the latter marker.  相似文献

测定分析人工饲养及在非麻醉(清醒)状态下108只健康成年恒河猴的血常规、血液生化及血小板和凝血功能正常值,并分析各项检测数据的性别差异。血常规测定值17项,其中,雌猴较雄猴极显著高(P<0·01)的项目为红细胞分布宽度(RDW)。血液生化测定值17项,其中,雌猴比雄猴显著低(P<0·05)的项目有丙氨酸转氨酶(ALT),显著高(P<0·05)的项目有碱性磷酸酶(ALP)。血小板和凝血功能测定值9项,其中,雌猴比雄猴明显高(P<0·05)的项目为活化部分凝血活酶时间(APTT),极显著高(P<0.01)的项目为凝血酶时间(TT)。建立了人工饲养条件下健康成年恒河猴的血常规、血液生化、血小板和凝血功能正常参考值指标,丰富和完善了恒河猴动物实验和疾病诊断中血液分析的基础知识。  相似文献

Verticillium wilt, caused by Verticillium albo-atrum or V. dahliae, is an important disease of many worldwide crop species. In Europe, V. albo-atrum isolates infecting hop express different levels of virulence, inducing mild or lethal disease syndromes, and it is therefore an attractive model for studying the virulence of this pathogen. In this work, eleven amplified fragment length polymorphism (AFLP) primer combinations were used to analyze genetic variability among 55 V. albo-atrum hop isolates from four European hop growing regions, as well as isolates from other hosts and V. dahliae isolates. Cluster analysis divided V. albo-atrum and V. dahliae isolates into two well-separated groups. Within the V. dahliae cluster, isolates were separated without host specific grouping, although no host adapted isolates were included. In V. albo-atrum, the alfalfa isolates were distinct from isolates of other hosts, where a high association with virulence was observed in hop and tomato isolates. All lethal hop isolates were genetically different from mild hop isolates. The lethal hop isolates from England and Slovenia expressed the same virulence phenotype, although they showed a different AFLP pattern. The mild hop isolates formed two subgroups, to which isolates clustered irrespective of geographical location. These data suggest multiple origins of V. albo-atrum hop isolates, and the possible appearance of new virulent isolates in the future in other hop growing regions.  相似文献

The study on behavioral hormoligosis in oviposition preference in Bemisia tabaci (Genn.) on cotton was conducted, at Entomological Research Farm, Punjab Agricultural University (PAU), Punjab, India, during 2001 crop season and repeated in the same season. Multiple-choice test was followed for conducting the experiment. Quinalphos (250, 375, and 500), carbaryl (625, 938, and 1250), acephate (750, 1125, and 1500), endosulfan (438, 656, and 875), and fenvalerate (25, 38, and 50 g ai/ha) were repeatedly sprayed on potted plants of American cotton (var. LH-1556). The impact of these insecticides was evaluated in term of oviposition preference by B. tabaci to treated plants. Also, it investigated changes in biochemical components of treated cotton leaves and the correlation with oviposition preference. The results revealed that, fenvalerate treated plants were more preferred by whitefly for oviposition. Maximum number of eggs was observed on fenvalerate treated plants, 38, 50, and 25 g/ha (39.3, 37.3, and 36.1 eggs/leaf, respectively) followed by acephate 1500 g/ha (26.9 eggs/ leaf) compared with untreated control (14.1 eggs/leaf). Almost similar trend of results was observed in the repeated experiment. The results obtained from biochemical studies revealed that all the insecticidal treatments caused reduction in total sugars compared with untreated control except fenvalerate and low dose of quinalphos. All insecticides caused increase in total free amino acids and brought significant changes in total phenols and pH value of treated plants. These results have confirmed the behavioral hormoligosis in oviposition preference that induced by fenvalerate and acephate in B. tabaci, which may be one of the causes behind its resurgence on plants repeatedly treated with these insecticides.  相似文献

Inhibitory effects of amicarthiazol, in vitro, against the growth of seven plant bacterial pathogens and its biochemical actions on Xanthomonas citri were investigated. The growth of Erwinia carotovora, Ralstonia solanacearum, and Pseudomonas syringe pv. syringae was not sensitive to amicarthiazol whilst Xanthomonads were all strongly inhibited in NB liquid medium; EC50 of X. citri was 2.63 μg ml−1 and the effectiveness of amicarthiazol depended on the timing of application. The inhibitions on the secretion of gross EPS and gross EPR as well as the activities of amylase, cellulase, and pectase of X. citri were increased with the concentrations of amicarthiazol, whereas the conductivities of the extracellular product suspensions and the protease activity were somewhat accelerated by the concentration less than 10 or 20 μg ml−1 and inhibited only at higher concentration. EPS which was purified from X. citri showed obvious antagonistic effects on the inhibition of amicarthiazol, but not on the inhibition of Cu(OH)2. The endogenous respirations and the activity of succinic dehydrogenase of X. citri were also inhibited and the inhibitions were increased with the toxic concentrations. Furthermore, the potential induced physiological adaptability of X. citri to amicarthiazol and the comparison with Cu(OH)2 and carboxin were also discussed.  相似文献

In this study, the cross‐amplification of a commercial multiplex set of 11 cattle (Bos taurus) microsatellites was tested on a panel of 35 European bison (Bison bonasus) individuals. After polymerase chain reaction optimization, all loci cross‐amplified successfully in investigated bisons. Number of alleles and observed and expected heterozygosity per locus are in the range of 2–4, 0.086–0.629 and 0.288–0.621 respectively. The availability of a heterologous set of multiplexed microsatellite markers derived from cattle opens an avenue for collecting profound genetic data for efficient conservation management strategies of the European bison.  相似文献

This research was conducted to assess polymorphism among local genotypes of common fig available in Jordan (one of countries of origin). Leaf samples were collected in spring for DNA isolation from 20 different local genotypes (5 cultivars and 15 landraces). Two more wild types and one foreign cultivar were included. The genotypes were assessed using the randomly amplified polymorphic DNA (RAPD) technique. Six of the 19 screened primers showed reproducible polymeric profiles. Out of 62 amplified bands, 48 were polymorphic (77%). They generated 1104 data entries (591 for present and 513 for absent bands). After determining Jaccard similarity index, some genotypes showed high genetic similarity (90% between F20 and F22), while other were less similar (3–18% between F11 and all other genotypes). Moreover, the primers were evaluated for their discriminating power, where primer RAPD06 showed the weakest power (0.431), while highest values of 0.989 and 0.996 were achieved for primers RAPD02 and RAPD13, respectively.  相似文献
