Six isolates of Mycoplasma bovoculi obtained from cattle herds with bovine keratoconjunctivitis were analyzed by gel electrophoresis and immunoblotting techniques. All six strains showed similarity in their protein profiles although no two patterns were identical. Antigenic differences between strains were detected in immunoblots reacted with post-exposure calf serum. A common 94 kDa protein band designated p94 was detected in all six strains reacted with monoclonal antibody MA25.5 developed to one of the strains. The p94 was also recognized in these strains by the calf serum. Trypsin treatment of intact mycoplasma cells resulted in the removal of p94 from immunoblots reacted with MA or hyperimmune rabbit serum. Other trypsin-resistant antigens shared between strains or being strain-specific in nature were identified when trypsin-treated mycoplasma cells were reacted with hyperimmune rabbit serum. The p94 antigen was shown to be of mycoplasmal origin by radio-immunoprecipitation using the MA or hyperimmune rabbit serum. These studies identify the presence of a surface antigen (p94) on M. bovoculi membrane in all strains examined that is trypsin sensitive by the use of monoclonal antibody, calf serum and hyperimmune rabbit serum.

Six souches de Mycoplasma bovoculi, d'origine bovine (animaux affectés de keratoconjunctivite) ont été analysées par des techniques d'éléctrophorèse et par ‘immunoblotting’. Les six souches ont montré des similitudes dans leurs profils proteiniques. Cependant, aucun profil n'a été identique. Des differences antigéniques entre les souches ont étés detectées suite au traitement des immunoblots avec un sérum bovin infecté par M. bovoculi. L'anticorps monoclonal MA25.5 dirigé contre une des souches a réagi avec une bande proteinique (94 kDa) commune aux six souches. Cette bande a été designee p94. Cette bande p94 a été également identifiée chez tous les souches en utilisant le sérum bovin. Le traitement des mycoplasmes intactes par la trypsine a éliminé la bande p94 des immunoblots: cela a été démontr é par l'usage d'un anticorps monoclonal, ou par un sérum hyperimmun de lapin. D'autres antigènes résistants à la trypsine, communs ou spécifiques aux souches ont été identifiés quand les mycoplasmes traités par le trypsine ont été mélangés au sérum hyperimmun de lapin. La radioimmunoprécipitation par l'anticorps monoclonal ou le sérum hyperimmun de lapin a demontré que l'antigène p94 est d'origine mycoplasmique. Ces études identifient la présence d'un antigène de surface (le p94) localisé sur la membranc de M. bovoculi chez toutes les souches examinées. Cet antigène est sensible à la trypsine. The aims of this follow-up study were: (a) to evaluate the role of ECT technology as a risk factor for several diseases; and (b) to determine if the effects of these diseases on cows' reproductive performance and as risk factors for culling are influenced by exposure to ECTs. Diseases considered were retained placenta, metritis, ketosis, cystic ovaries, silent heat, milk fever, clinical mastitis, and foot and leg problems. We used historical and contemporary controls (with control herds selected to match the experimental herds for size and location). Data consisted of 10264 Swedish Red and White (SRB) and 5461 Swedish Friesian (SLB) lactation records in 150 herds of which 33 used cow-trainers. Logistic regression was used to estimate the effects of parity and exposure to electric cow-trainers on the risks of diseases and the effects of diseases and exposure to electric cow-trainers on risk of culling. The least-squares procedure was used to estimate the effects of diseases on reproductive performance.
The dominant effects associated with use of electric cow-trainers were an increased risk for silent heat, clinical mastitis, ketosis and culling relative to cows in herds not using cow-trainers. Diseases had negative effects on reproductive performance and the effects were larger for cows in herds using cow-trainers. In herds using electric cow-trainers, the largest increase in the interval from first service to conception (58 days) was caused by the occurrences of silent heat, cystic ovaries and the combination of two or more diseases. Retained placenta, metritis, cystic ovaries, clinical mastitis and a combination of two or more diseases increased the risk of culling about two times relative to healthy primiparous cows with the increase being greater for cows in herds using cow-trainers. Silent heat did not increase risk of culling in control groups, but was the largest risk factor for culling in the exposed group. We concluded that exposure to electric cow-trainers increased the incidence risk of silent heat, clinical mastitis, and ketosis and changed silent heat from a neutral disease with respect to culling to a major risk factor. Finally, exposure to cow-trainers increased the general negative effect of diseases on the cows' reproductive performance and risk for culling. 相似文献
The accuracy and precision of an end-tidal infrared CO2 monitor and customized capnography tubing system designed to measure gastrointestinal intraluminal CO2 partial pressure (PiCO2) were tested in vitro. Samples were taken from the customized capnography tubing placed in either 5% or 10% CO2 gas at discrete intervals ranging from one minute to twelve minutes. For a given time interval, the tubing PCO2 measurement was a constant fraction of the actual PCO2 (all standard errors < 0.02). For increasing time intervals, the ratio of the tubing PCO2 to actual PCO2 increased logarithmically. In the 5% and 10% CO2, the regression coefficients were 0.89 and 0.85 for 8 French tubing and 0.99 and 0.91 for 6 French tubing. Beacuase of its accuracy and precision, this system may provide automated gastrointestinal CO2 partial pressure monitoring at short intervals (e.g. 5 minutes), facilitating testing of the role of gastrointestinal PiCO2 information in treatment algorithms. Interspecific allo-tetraploidy was induced by a combination of hybridization of Megalobrama amblycephala ♀× Megalobrama terminalis ♂ and thermal shock. A thermal shock at 40 °C for 2 min applied to eggs 33 or 26 min (τ0: 1.40) after fertilization resulted in a significantly ( P <0.01) higher yield of allo-tetraploids than hybrid crosses without heat shock. The yield of allo-tetraploids in embryos from mid-gastrula to hatching stages was 15.7–24.4% in different thermal shock treatment groups. The allo-tetraploidy rate in 330-day-old juveniles was 9.3% and 6.7% in different year treatment groups. After 2 years of rearing, allo-tetraploidy males attained sexual maturity with milk-like milt at the age of 2+, while allo-tetraploidy females at the age of 2+, and even at 3+, did not show any signs of sexual maturation exhibiting obviously delayed maturity. Only a few allo-tetraploidy females (three individuals) showed maturation characters at the age of 4+ and these females were induced to spawn. 相似文献
In this work, the forward extraction of defatted wheat germ protein (DWGP) by reverse micelles was studied. The reverse micellar systems were formed by sulphosuccinic acid bis (2-ethylhexyl) ester sodium salt (AOT), isooctane and KCl solution. The effects of AOT concentration, pH, KCl concentration, extraction time, the amounts of defatted wheat germ flour (DWGF), W0 (the molar ratio of water to surfactant, i.e. W0 = [H2O]/[AOT]) and temperature on the forward extraction efficiency of DWGP were tested. On the basis of single-factor experiments, the optimum extraction was achieved by response surface methodology (RSM). The experimental results lead to the conclusion that the highest forward extraction efficiency of DWGP was reached at the AOT concentration 0.06 g/mL, pH 8, KCl concentration 0.1 mol/L, time 30 min, the amounts of DWGF 0.500 g, W0 25 and temperature 36 °C. Under these conditions, the forward extraction efficiency of DWGP achieved 37%. 相似文献
AIM: To investigate the mutative law of the serum S100-B protein in patients undergone coronary artery bypass grafting with or without cardiopulmonary bypass (CPB-CABG or non-CPB CABG), and to compare the degree of cerebral injury in patients during CABG between the two different surgical manipulations.METHODS: 30 patients were divided into two groups: CPB-CABG group (group A) and non-CPB CABG group (group B). Every group included 15 patients. Blood from jugular vein was phlebotomized in every patients at the following 8 time points: before operation (OP), after anaesthesia, end of grafting, end of OP, 2 h after OP, 6 h after OP, 12 h after OP and 24 h after OP. The serum concentrations of S100-B protein in the samples were measured.RESULTS: The level of serum S100-B protein increased markedly at the beginning of CABG, and the peak value of group A (2.32 μg/L±0.26 μg/L) was treble higher than that in the patients in group B (0.71 μg/L±0.14 μg/L). The levels of S100-B protein tend to normal level in both groups 24 h after OP. The level of S100-B protein had notable relation with the time of CABG in group A and the manipulative time on the heart in group B.CONCLUSION: The degree of cerebral damage can be valued by investigating the mutative law of the serum S100-B protein in patients undergone CPB-CABG and non-CPB CABG. The non-CPB CABG reduces the degree of cerebral damage and avoids the functional disorder of central nerve. 相似文献
