大麦芽阿魏酸酯酶的分离纯化及其部分酶学性质的测定 总被引:2,自引:1,他引:2
本试验采用硫酸铵分级沉淀、透析、DEAE-52离子交换层析对大麦芽中的阿魏酸酯酶(FAE)进行了分离纯化,并测定了其部分酶学性质。结果显示,分离纯化后获得了阿魏酸酯酶纯品,纯化倍数达34倍;经SDS-PAGE电泳显示为单一条带,并测定其相对分子质量约为29.3 ku;最适反应温度为50℃;最适pH为5.5,但阿魏酸酯酶在60℃以下,pH 4.5~7.5之间有较好的稳定性;Zn2+、Cu2+、Fe3+对酶活有很强的抑制作用,Na+和EDTA对酶有一定的激活作用。 相似文献

从采取的新鲜麋鹿血液中提取麋鹿基因组DNA,通过PCR的方法从麋鹿基因组中扩增麋鹿PrP蛋白核心片段的DNA,并分别在引物的5′和3′端引入EcoRⅠ和HindⅢ两个酶切位点,下游引物将原序列TATTAC突变为TAAT从变成两个终止密码子。通过两个酶切位点将麋鹿PrP蛋白核心片段DNA连接到pET-32a(+),转化DH5α,通过酶切鉴定和PCR鉴定,筛选出阳性克隆送菌液测序,从测序结果分析阳性克隆是否符合要求。测序正确后将核心片段基因连接到pET-32a(+),阳性质粒命名为MPrP-pET-32a(+),阳性质粒转化E.coli BL21(DE3),进行诱导表达、蛋白质纯化鉴定。 相似文献

Sixty-four Wistar rats were divided into 8 equal groups and kept for 36 days in individual boxes. Three of the groups were given full synthetic diets containing various animal fats: beef tallow, pork fat or fish oil. A control group was fed the diets without the fat. The other four groups were fed the same diets with lecithin supplementation. At the end of experiment, sections taken from the liver were stained with haematoxylin–eosin and Sudan III to indicate fat infiltration. Liver enzyme levels, total bilirubin, albumin and two products of lipd metabolism were measured in serum from all the animals. The addition of lecithin to the diets did not influence the level of enzymes in the serum except in rats fed the diet containing beef tallow. A relationship between the type of diet, lecithin supplementation and the total cholesterol concentration in serum was observed. The concentration of HDL-cholesterol was only influenced by lecithin supplementation and that of triglycerides by the type of fat in the diet. The addition of lecithin to the diet was associated with proliferation of Kupffer cells, and an increased number of binuclear cells. Fatty degeneration of hepatocytes was less pronounced in all groups following lecithin supplementation. Lecithin supplementation of the diet did not elicit any pathological lesions and may be considered as a hepatocyte protector. 采用蛋鸡离体外翻肠段培养法,将小肠分为十二指肠、空肠前段、中段、后段和回肠五个部位肠段,洗净外翻。2种二肽培养液(Gly-Gly肽和Gly-L-Leu肽溶液)作为两组对照培养液,每种二肽培养液中添加肽酶抑制剂(Bestatin和Amastatin),作为试验培养液,39.5℃培养25min,每5min取样一次,测定培养液中的二肽含量。结果表明,培养5min后,含Gly-Gly肽的对照和试验培养液中均未能检测到完整Gly-Gly肽。而含Gly-L-Leu肽的对照培养液Gly-L-Leu肽的含量降低到16.75%,试验培养液则下降到43.40%。而试验培养液中Gly-L-Leu肽的含量10min后无显著降低(P>0.05)。各培养时段试验培养液Gly-L-Leu含量均显著高于对照培养液(P<0.05)。由此认为,离体肠囊在培养过程中能够迅速释放肠肽酶及一些生物活性物质,快速水解2种二肽。肽酶抑制剂(Bestatin和Amastatin)能抑制Gly-L-Leu的水解,但不能抑制Gly-Gly的水解。 相似文献

旨在分析小麦粉中添加木聚糖酶(xylanase, Xyl)对藏羊羔羊胃肠道发育的影响。选择同质性良好的2~3月龄高原型藏羊公羔60只,随机分为两组,每组30只,各组内设置5个重复。对照组(Control group)饲喂不含木聚糖酶的日粮,试验组(Test group)饲喂含0.2%木聚糖酶的日粮,试验分为预饲期10 d和正试期90 d。饲养试验结束时,两组各随机选取5只羔羊进行屠宰,采集消化道组织置于多聚甲醛固定,并收集血液、瘤胃和空肠内容物。结果显示:1)与对照组相比,试验组瘤胃的角质层厚度、颗粒层厚度,瓣胃的乳头长度、乳头宽度、角质层厚度、颗粒层厚度,皱胃的黏膜厚度以及十二指肠的肌层厚度,空肠的绒毛高度、黏膜厚度,回肠的肌层厚度显著提高(P<0.05);2)对照组瘤胃和血清的LPS含量显著高于试验组(P<0.05);3)相较于对照组,试验组瘤胃的糜蛋白酶、胰蛋白酶、脂肪酶活性以及空肠的糜蛋白酶、α-淀粉酶活性显著提高(P<0.05);4)试验组瘤胃的总抗氧化能力水平、谷胱甘肽过氧化物酶及过氧化氢酶活性以及空肠的总抗氧化能力水平、谷胱甘肽过氧化物酶、超氧化物歧化酶... 相似文献

This study was to investigate the effects of in ovo feeding (IOF) L‐arginine (Arg) solution on the development of digestive organs, the duodenal mucosa of broiler embryos and hatchlings, and the growth performance of chicks during the first week post‐hatch. A total of 720 fertilized eggs with similar weight were randomly allocated to three groups, consisting of eight replicates of 30 eggs each. Three treatments were arranged as non‐injected control, diluent‐injected (0.75% NaCl solution) group and Arg solution‐injected group containing 1% Arg, dissolved in diluent. At 17.5 days of incubation, 0.6 ml of IOF solution was injected into amniotic fluid of each egg of injected groups. Results showed IOF of Arg solution increased (p < .05) the chick embryo weight at 19 days of incubation; the body weight gain of post‐hatch broilers during 1–7 days; the weights of liver, pancreas, proventriculus and gizzard; the concentrations of duodenal ghrelin, vasoactive intestinal peptide and glucagon‐like peptide 2; and the duodenum mucosal enzyme activities of alkaline phosphatase, maltase, sucrase and inducible nitric oxide synthase of 7‐day‐old post‐hatch broilers compared with other groups. The IOF of Arg solution also increased (p < .05) the villus height (VH) and the ratio of VH to crypt depth (CD) and decreased (p < .05) the CD in duodenum of broiler embryos and post‐hatch hatchlings, except for the CD at 19 days of incubation. In conclusion, IOF of 1% Arg solution into the amnion at 17.5 days of incubation could improve the development of digestive organs, the duodenal morphology, the releasing of gastrointestinal hormones and mucosal enzyme activities of broiler embryos and hatchlings and finally the growth performance of chicks during the first week post‐hatch. Therefore, IOF of appropriate Arg solution could be an effective technology for regulating early nutrition supply and subsequent growth development in poultry industry. 相似文献
