Cell wall disassembly in ripening climacteric fruit is a highly complex process where ethylene plays a crucial role. Ethylene inhibitors can be used to explore the changes in the cell wall matrix and cross-linked polysaccharides in ethylene-regulated processes. The results of applying the ethylene receptor blocking inhibitor 1-methylcyclopropene (1-MCP) and the ethylene-releasing compound ethephon (2-chloroethylphosphonic acid) indicate that softening of ‘Maradol’ papaya fruit is dependent on ethylene. When fruit were induced to ripen extensively by exposure to a high dose of ethephon, 1-MCP inhibited the subsequent softening dramatically, but when inhibition of the ethylene response was caused by application of 1-MCP, subsequent fruit treatment with ethephon promoted extensive loss of galactose from the water-soluble polysaccharides, but this was not accompanied by fruit softening. The cell wall changes accompanying normal fruit softening were pectin solubilization and polyuronide depolymerization and these processes occurred simultaneously. Polygalacturonase likely is responsible for the ripening-associated changes in ‘Maradol’ papaya fruit texture and pectin polymer integrity. An increase in extractable fruit polygalacturonase follows the increased presence of pectin-derived oligosaccharides. 相似文献

Methods must be developed to improve both the processability and physical properties of zein-based articles. Typical plasticizing agents that improve processability have a negative impact on physical properties at elevated humidities. Continued efforts must be employed to discover improved plasticizers. Carboxylic acids having more than one carboxylic acid moiety have been evaluated in zein melt formulations produced in a torque rheometer. These reagents were effective plasticizers for zein, lowering zein viscosity, and delaying the onset of rapid viscosity increase. These reagents altered viscosity differently than the traditional plasticizers such as triethylene glycol. These additives also lowered the tensile strength (TS) of zein formulations at 50% RH. Surprisingly, TS at 70% RH was higher than that at 50% RH at elevated levels of these additives. NMR analysis of Soxhlet extracted samples showed that these multivalent carboxylic acids do not cross-link the zein under the reaction conditions employed. 相似文献

Button mushrooms (Agaricus bisporus) were dipped for 2 min in different concentrations (1%, 2% and 3%) of alginate, then placed in jars and ventilated continuously with 100% O2 for up to 16 d at 4 °C. Mushroom respiration rate, weight loss, texture, color, percent open caps, chemical properties and activities of polyphenol oxidase (PPO), and peroxidase (POD) were measured. The results indicate that treatment with alginate coating (2%) + 100% O2 maintained a high level of firmness, and delayed browning and cap opening. In addition, alginate coating (2%) + 100% O2 also delayed changes in the soluble solids concentration, total sugars and ascorbic acid, and inhibited the activity of PPO and POD throughout storage. Our study suggests that the use of alginate coating under a high oxygen modified atmosphere has the potential to maintain button mushroom quality and extend its postharvest life to 16 d. 相似文献

The effects of postharvest application of aminoethoxyvinylglycine (AVG) and 1-methylcyclopropene (1-MCP) on ethylene production and fruit quality, and thus on transportation and shelf-life, were evaluated in melting-flesh peaches. AVG (150 mg L−1) significantly reduced ethylene production, and the effect was enhanced in combination with 1-MCP (1 μL L−1). However, fruit treated with AVG alone softened to untreated control levels 2 d after harvest (DAH). Treatment with 1-MCP significantly reduced the rate of softening until 2 DAH, but the fruit rapidly softened thereafter, and reached untreated control levels by 4 DAH. A combination of AVG and 1-MCP significantly reduced fruit tissue softening throughout ripening. The effect of each chemical on flesh firmness indicated that 1-MCP affected fruit response in the early stages of ripening up to 4 DAH, and AVG significantly reduced softening in the latter stages from 4 to 9 DAH. Peaches treated with AVG and 1-MCP retained their ground color during ripening, but the effect of each chemical on color is unclear. A highly selective imprinted polymer was synthesized by a surface molecular imprinting technique in combination with a sol-gel process. The imprinted polymer was evaluated by FT-IR and static and kinetic adsorption experiments. The results showed that the imprinted sorbent exhibited good recognition and selective ability, offered a faster kinetics for the adsorption and desorption of Sudan I than the non-imprinted sorbent, a saturated binding capacity (Qmax) that reached 33.47 mg g-1. The prepared sorbent was applied for the determination of trace Sudan I through on-line solid-phase coupled with high-performance liquid chromatography (SPE-HPLC). With a loading flow rate of 1.5 mL min-1 for sampling 50 mL, an enrichment factor of 1266 was achieved. The detection limit (S/N = 3) was 1.2 ng L-1, and the peak area precision (RSD) for five replicate detections of 0.01 microg L-1 Sudan I was 3.66%. The Sudan I in the chilli powder from the local market was determined at three levels (0.25, 0.5, and 1.0 ng g-1) with recoveries ranging from 80.31 to 94.02%. 相似文献

【目的】建立碱提甘蔗皮多糖的优化工艺,进一步探究其结构特征,并评价其对α-葡萄糖苷酶的抑制作用。【方法】采用单因素试验和响应面分析法(RSM)对碱提甘蔗皮多糖(SPAP)提取工艺进行优化,采用苯酚硫酸法测定多糖含量。SPAP经Sevag法除蛋白、AB-8除色素后进行结构表征,主要包括高效液相色谱法(HPLC)检测多糖分子量分布,气相-质谱色谱仪(GC-MS)进行单糖分析及傅里叶红外光谱仪(FT-IR)对官能团进行分析。采用4-硝基苯基-D-吡喃葡糖苷(PNPG)法测定多糖提取物对α-葡萄糖苷酶抑制作用。【结果】SPAP的最佳提取条件为提取温度37℃、氢氧化钠(NaOH)浓度5%、料液比1∶46(g·mL-1)、提取次数4次,在此条件下SPAP得率达到10.84%。经除蛋白、色素后,SPAP含量达到86.54%,主要由阿拉伯糖、木糖、葡萄糖和半乳糖4种单糖组成,分子量为3.03×103 kD,可能为吡喃型杂多糖,呈现α或β构型。此外,SPAP表现出良好的α-葡萄糖苷酶抑制作用,其抑制率达到78.31%。【结论】对碱提甘蔗皮多糖进行工艺优化,可有效利用原料、提高产出和效率;由HPLC、FT-IR、GC-MS等方面初步阐述了SPAP的结构特性,同时SPAP对α-葡萄糖苷酶表现出良好的抑制作用,具有一定的降糖潜力,研究结果为进一步研究SPAP的构效关系提供了理论依据。 相似文献

为了解乳品来源蜡样芽胞杆菌携带的毒力因子、分子特征及耐药性,对2016—2019年分离自乳品的122株蜡样芽胞杆菌,采用纸片扩散法(K-B纸片法)测定了分离株对氯霉素、庆大霉素等9种抗生素的耐药性,使用聚合酶链式反应法检测了菌株携带的毒力因子,同时采用脉冲场凝胶电泳(pulsed field gel electrophoresis,PFGE)检测分离株的分子特征。结果表明,122株蜡样芽胞杆菌均对氯霉素、庆大霉素、阿米卡星敏感;对红霉素、克林霉素和环丙沙星的中介率分别为5.74%、4.92%和0.82%, 9.84%的菌株对复方新诺明具有耐药性,8.20%的菌株对四环素具有耐药性,4.10%的菌株对利福平具有耐药性。122株蜡样芽孢杆菌都检出了蜡样芽孢杆菌的非溶血型肠毒素基因nheA、nheB及肠毒素FM基因entFM;nheC、bceT、cytK、hblA、hblC、hblD、ces和EM1基因的检出率分别为99.18%、37.70%、31.15%、29.51%、24.59%、8.20%、、6.56%和6.56%。122株蜡样芽胞杆菌被分成15簇,未见明显的优势带型。由此可见,2016—2019年收集的蜡样芽胞杆菌乳品分离株携带多种毒力基因,约10%的菌株对复方新诺明、四环素、利福平等具有耐药性,PFGE呈现多种带型,为蜡样芽孢杆菌引起的食源性疾病的预警、预防和治疗提供参考。 相似文献
