Pituitaries of various teleosts belonging to 25 orders were immunostained with antisera raised against synthetic fragment peptides corresponding to conservative regions of gonadotropin subunits (mummichog FSH 50-60 and mummichog LH 91-106). Both immunoreactive FSH cells and immunoreactive LH cells were successfully identified in the fishes of almost every order of the superorder Acanthopterygii and several species of the superorders Paracanthopterygii and Polymixiomorpha, such as mullet, alfonsino, flyingfish, mackerel, flounder, cod, beardfish, etc. These antisera are therefore considered as universal antisera for immunocytochemical application to acanthopterygian fishes. Extensive diversity in the abundance of the FSH cells and the LH cells among species was noted even in fishes with similar gonadal stages, indicating the possibility that the respective roles of FSH and LH may vary considerably among species in advanced teleosts.Evident but generally weak immunoreactivities to anti-mummichog LH 91-106 were observed in the fishes of the superorder Cyclosquamata; and slight or weak immunoreactivities to the antiserum were observed in the fishes of several more primitive taxa (superorder Stenopterygii, Protacanthopterygii, Ostariophysi, subdivision Clupeomorpha, and subdivision Elopomorpha). No immunoreactivity to anti-mummichog FSH 50-60 was observed in these fishes. These results are consistent with the phylogenetic status of the fishes and the degree of conservativeness in the amino acid sequences of the antigen regions.  相似文献

Bovine spongiform encephalopathy (BSE) is a fatal neurodegenerative disorder of cattle characterized by accumulation of the disease-associated prion protein (PrP(Sc)) in the central nervous system (CNS). The immunohistochemical patterns and distribution of PrP(Sc) were investigated in the CNS, brains, and spinal cords of 7 naturally occurring BSE cases confirmed by the fallen stock surveillance program in Japan. No animals showed characteristic clinical signs of the disease. Coronal slices of 14 different brain areas in each case were immunohistochemically analyzed using an anti-prion protein antibody. Immunolabeled PrP(Sc) deposition was widely observed throughout each brain and spinal cord. Intense PrP(Sc) deposition was greater in the thalamus, brainstem, and spinal cord of the gray matter than in the neocortices. The topographical distribution pattern and severity of PrP(Sc) accumulation were mapped and plotted as immunohistochemical profiles of the different brain areas along the caudal-rostral axis of the brain. The distribution pattern and severity of the immunolabeled PrP(Sc) in the CNS were almost the same among the 7 cases analyzed, suggesting that the naturally occurring cases in this study were at the preclinical stage of the disease. Immunohistochemical mapping of the PrP(Sc) deposits will be used to clarify the different stages of BSE in cattle.  相似文献

This study aims to understand Th2 immune responses and alternative macrophage activation against nematode parasites in aged mice. Eighteen-month (18 M) and three-month (3 M) old C3H/HeN mice were inoculated with Heligmosomoides polygyrus (Hp) larvae. Real-time PCR analysis indicated that interleukin (IL)-4 and IL-13 gene expression was elevated in both groups after infection, but the expression level was significantly low in 18 M mice. Macrophage phenotype was monitored by measuring arginase-1 gene expression and immunofluorescence staining in small intestine, showing a decrease in the number of alternatively activated macrophages (AAMacs) around worm cysts in 18 M mice. These results suggest that the Th2 immune response in aged mice against a nematode parasite was not sufficiently induced to promote AAMacs.  相似文献

Characteristics of CPE produced by porcine enteroviruses (PEV) were examined by Immunoperoxidase (IP) staining method. Viral antigens were detected earlier than appearance of CPE. Distinctive characteristics of the three CPE types were clearly showed by the method. In cross reactions by IP staining, the titers of the staining were high (1:6,400 to 1:25,600), even though neutralizing titers of PEV with CPE II or III were low (1:200 to 1:400). The very close relationship was detected between PEV with the same CPE type, but the very low relationships were detected between PEV with the different CPE type. The relationships in CPE I group were various. The results by IP staining were more relative to CPE type than the serotype. Thus, IP staining is one method to classify PEV clearly.  相似文献

Analysis of chromosome-sized DNA and genome typing ofTaylorella equigenitalis NCTC11184, Kentucky 188, and five strains ofT. equigenitalis isolated in Japan were carried out. The three restriction enzymes used,ApaI,NaeI andNotI, cleaved the genomic DNAs of five Japanese strains ofT. equigenitalis into relatively limited numbers of restriction fragments, which were well resolved on crossed-field gel electrophoresis (CFGE). The respective profiles after CFGE of the restriction fragments from all five strains were essentially identical to each other after digestion byApaI,NaeI orNotI. Hence it appears that these strains have a common genome type with respect to these three restriction enzymes. The antiviral effects of recombinant bovine interferon-tau (rboIFN-tau) on bovine leukemia virus (BLV) were examined in vitro and in vivo. In the in vitro experiments, BLV titers decreased in FLK-BLV cells and in peripheral blood mononuclear cells of BLV-infected cattle treated with rboIFN-tau at a concentration higher than 10(2) U/ml. In order to examine the in vivo effects of rboIFN-tau, 10 BLV-infected cattle were subcutaneously injected with rboIFN-tau. In the first experiment, 6 cows were administrated with 10(5) U/kg body weight of rboIFN-tau 3 times per week for 4 weeks, while in the second experiment 4 cows were administrated with 10(6) U/kg body weight of rboIFN-tau 3 times per week for 3 weeks. No adverse effects were observed after the administration of rboIFN-tau. In experiment No. 1, the mean BLV titers in cattle decreased in the post-rboIFN-tau administration period compared to the pre-rboIFN-tau administration period. In experiment No. 2, the mean BLV titers in cattle decreased in the rboIFN-tau administration period. These results suggest that rboIFN-tau decreases BLV titers in vitro and in vivo and that rboIFN-tau possibly reduces the degree of BLV titer in cattle without severe side effects.  相似文献

Neuroaxonal dystrophy (NAD) was examined in two Papillon dogs and a mix breed dog between Papillon and Chihuahua. In addition, cerebellar cortical abiotrophy (CCA) in a Papillon dog, which had similar clinical and magnetic resonance imaging (MRI) features to those of NAD, was also investigated. The common clinical symptoms of all dogs affected with NAD and CCA, were pelvic limb ataxia and cerebellar ataxia including intention tremor, head tremor, and hypermetria in the early onset. These clinical signs were progressed rapidly, and two dogs with NAD were euthanized by owner's request and the other two died by aspiration pneumonia. MRI examinations and gross observations at necropsy revealed moderate to severe cerebellar atrophy in all cases of NAD and CCA. The most typical histological change of NAD was severe axonal degeneration with marked spheroid-formation in the dorsal horn of the spinal cords, the nuclei gracilis, cuneatus, olivalis and its circumference in the medulla oblongata. The spheroids were characterized as large eosinophilic or granular globes within the enlarged myelin sheaths, sometimes accompanied by moderate accumulation of microglias and/or macrophages. In contrast, such spheroid formation was minimal in the brain of CCA. In the cerebellum, mild to moderate loss of the Purkinje and granular cells were recognized in three dogs with NAD, whereas these changes were more prominent in a dog with CCA. Although the clinical signs and MRI findings relatively resembled between NAD and CCA, the histopathological features considered to be quite differ, suggesting distinct pathogenesis and etiology. Since both NAD and CCA are proposed as the autosomal recessive hereditary disorders, careful considerations might be needed for the breeding of Papillon and Chihuahua dogs.  相似文献

To evaluate the effects of a new synbiotic consisting of Lactobacillus casei subsp. casei (Lcc) and dextran (Dex) on milk production, a total of 58 Holstein dairy cows, which became pregnant and gave birth to calves at regular intervals and lactated steadily and continuously, were selected. The study had a completely randomized design, and the animals were divided into two groups. Group A was fed with a basic diet only, and Group B was fed with a basic diet supplemented with the synbiotic consisting of freeze-dried Lcc and mixed feed containing Dex for one year from August 2004. After supplementation with the synbiotic, milk yields and components of Group B were compared with those of Group A in the August, December of 2004, April and August of 2005. Milk yields of Group B were greater than those of Group A. There were significant differences (p<0.01 or 0.05) between these groups for all values. Furthermore, total amounts of fat, protein and solid non-fat in Group B significantly increased in comparison with those of Group A. In addition, the somatic cell counts of Group A significantly increased in August of 2004 and 2005 in comparison with those of Group B. Thus, the new synbiotic consisting of Lcc and Dex can increase the milk production of Holstein dairy cows throughout the year.  相似文献
