Natural clay minerals can play an important role in crude remediation of wastewater polluted with the heavy metals (HMs) Cu,Zn and Ni.The presence and timing of addition of natural dissolved organic matter (DOM) have a significant effect on the HM removal by clay mineral sorbents.However,the influence of the presence of DOM on the remediation of the used clay mineral sorbents once saturated with HMs is largely unknown.To resolve this,clay mineral-rich soil column of varying composition,loaded (i) with Cu,Zn and Ni only,(ii) first with DOM followed by Cu,Zn and Ni,or (iii) with DOM,Cu,Zn and Ni simultaneously,was used in a set of desorption experiments.The soil columns were leached with 0.001 mol L-1 CaCl2 dissolved in water as control eluent and 0.001 mol L-1 CaCl2 dissolved in DOM as treatment eluent.During the preceding loading phase of the sorbent,the timing of DOM addition (sequential or concurrent with HMs) was found to have a significant influence on the subsequent removal of the HMs.In particular when the column was loaded with DOM and HMs simultaneously,largely irreversible co-precipitation took place.Our results indicate that the regeneration potential of clay mineral sorbents in wastewater treatment will be significantly reduced when the treated water is rich in DOM.In contrast,in manured agricultural fields (where HMs enter together with DOM),HM mobility will be lower than expected from interaction dynamics of HMs and clay minerals.  相似文献

A full-length cDNA of a glutathione S-transferase (GST) was cloned from a cDNA library of the local Egyptian cattle tick Boophilus annulatus. The 672 bp cloned fragment was sequenced and showed an open reading frame encoding a protein of 223 amino acids. Comparison of the deduced amino acid sequence with GSTs from other species revealed that the sequence is closely related to the mammalian mu-class GST. The cloned gene was expressed in E. coli under T7 promotor of pET-30b vector, and purified under native conditions. The purified enzyme appeared as a single band on 12% SDS-PAGE and has a molecular weight of 30.8 kDa including the histidine tag of the vector. The purified enzyme was assayed upon the chromogenic substrate 1-chloro-2,4-dinitrobenzene (CDNB) and the recombinant enzyme showed high level of activity even in the presence of the beta-galactosidase region on its 5' end and showed maximum activity at pH 7.5. The Km values for CDNB and GSH were 0.57 and 0.79 mM, respectively. The over expressed rBaGST showed high activity toward CDNB (121 units/mg protein) and less toward DCNB (29.3 units/mg protein). rBaGST exhibited peroxidatic activity on cumene hydroperoxide sharing this property with GSTs belonging to the GST alpha class. I50 values for cibacron blue and bromosulfophthalein were 0.22 and 8.45 microM, respectively, sharing this property with the mammalian GSTmu class. Immunoblotting revealed the presence of the GST molecule in B. annulatus protein extracts; whole tick, larvae, gut, salivary gland and ovary. Homologues to the GSTmu were also detected in other tick species as Hyalomma dromedarii and Rhipicephalus sp. while in Ornithodoros moubata, GSTmu homologue could not be detected.  相似文献

Three hundred Oreochromis niloticus were divided into five groups; the control fish (CNT) were fed a basal diet, beta glucan (βG) group was fed 0.5 g/kg?1 βG, atrazine (ATZ) group was exposed to 1/5 96‐hr LC50 (1.39 mg/L) ATZ, the (βG/ATZ) group was fed βG while being exposed to ATZ, and the (βG then ATZ) group was supplemented with βG for fifteen days before exposed to ATZ. ATZ exposure caused a decline in growth that was ameliorated by βG. ATZ reduced the levels of total and different types of leucocytes. Additionally, ATZ exposure caused reductions in total proteins, globulins, α 1‐globulin, α 2‐globulin, ?‐globulin, immunoglobulin M, lysozymes, superoxide dismutase, nitric oxide and catalase but increases in hepatic transaminases and malondialdehyde without any variations in albumin and β‐globulin. Exposure to ATZ also resulted in a rise in the mRNA level of IL‐8. In contrast, expression of IgM, SOD and CAT were decreased in the tilapias exposed to ATZ. Exposure to ATZ increases the susceptibility response to Aeromonas sobria challenge, as indicated by an increase in cumulative mortality post‐challenge. Supplementation with βG fifteen days before (βG then ATZ group), counteracted the adverse effects of ATZ on the immune, biochemical and antioxidants values, though only slight alleviation was observed with simultaneous treatment (βG/ATZ group). Our results established that ATZ has adverse impacts on immune responses, antioxidant equilibrium and its related genes. While, supplementation with βG before exposure to ATZ may be valuable for counteracting the possible damage caused by ATZ water pollution than its simultaneous treatment with ATZ.  相似文献

Over 225,000 independent Agrobacterium transferred DNA (T-DNA) insertion events in the genome of the reference plant Arabidopsis thaliana have been created that represent near saturation of the gene space. The precise locations were determined for more than 88,000 T-DNA insertions, which resulted in the identification of mutations in more than 21,700 of the approximately 29,454 predicted Arabidopsis genes. Genome-wide analysis of the distribution of integration events revealed the existence of a large integration site bias at both the chromosome and gene levels. Insertion mutations were identified in genes that are regulated in response to the plant hormone ethylene.  相似文献

Angiotensin-converting enzyme (ACE) inhibitory capacity was evaluated in hydrolysates from sea cucumber Isostichopus badionotus. The pepsin-pancreatin and Alcalase®-Flavorzyme® sequential systems were used. Hydrolysates were characterized in terms of degree of hydrolysis (DH), electrophoretic profile, and inhibitory activity. The Alcalase®-Flavorzyme® system produced hydrolysates with the highest DH, with that produced at 90 min exhibiting the highest ACE inhibitory action (86%). This hydrolysate was chosen for purification by ultrafiltration with five molecular weight cut-offs (< 1, 1–3, 3–5, 5–10, and > 10). The > 10-kDa fraction exhibited the highest ACE inhibitory action (80.7%) and was fractionated by gel fractionation chromatography. Of the 11 resulting fractions, IX had the highest ACE inhibitory action (73.4%) and lowest IC50 value (2-μg prot/mL). Hydrolysis of sea cucumber I. badionotus with the Alcalase®-Flavorzyme® system produced hydrolysates with ACE inhibitory action. These contained a peptide with an IC50 value comparable to those of synthetic ACE inhibitors.  相似文献

Photocatalytic reduction of CO2 in seawater into chemical fuel, methanol (CH3OH), was achieved over Cu/C-co-doped TiO2 nanoparticles under UV and natural sunlight. Photocatalysts with different Cu loadings (0, 0.5, 1, 3, 5, and 7 wt%) were synthesized by the sol–gel method and were characterized by XRD, SEM, UV–Vis, FTIR, and XPS. Co-doping with C and Cu into TiO2 remarkably promoted the photocatalytic production of CH3OH. This improvement was attributed to lowering of bandgap energy, specific catalytic effect of Cu for CH3OH formation, and the minimization of photo-generated carrier recombination. Co-doped TiO2 with 3.0 wt% Cu was found to be the most active catalyst, giving a maximum methanol yield rate of 577 μmol g-cat?1 h?1 under illumination of UV light, which is 5.3-fold higher than the production rate over C-TiO2 and 7.4 times the amount produced using Degussa P25 TiO2. Under natural sunlight, the maximum rate of the photocatalytic production of CH3OH using 3.0 wt% Cu/C-TiO2 was found to be 188 μmol g-cat?1 h?1, which is 2.24 times higher than that of C-TiO2, whereas, no CH3OH was observed for P25.  相似文献
