Stimulation of Toll-like receptors (TLRs) triggers activation of a common MyD88-dependent signaling pathway as well as a MyD88-independent pathway that is unique to TLR3 and TLR4 signaling pathways leading to interferon (IFN)-beta production. Here we disrupted the gene encoding a Toll/IL-1 receptor (TIR) domain-containing adaptor, TRIF. TRIF-deficient mice were defective in both TLR3- and TLR4-mediated expression of IFN-beta and activation of IRF-3. Furthermore, inflammatory cytokine production in response to the TLR4 ligand, but not to other TLR ligands, was severely impaired in TRIF-deficient macrophages. Mice deficient in both MyD88 and TRIF showed complete loss of nuclear factor kappa B activation in response to TLR4 stimulation. These findings demonstrate that TRIF is essential for TLR3- and TLR4-mediated signaling pathways facilitating mammalian antiviral host defense. 相似文献

An analog of angiotensin II, [Sar(1), Ile(8)]-angiotensin II, has a potent and long-lasting competitive antagonistic effect against angiotensin II when tested for its myotropic action on the isolated rabbit aorta and for its effect on blood pressure in anesthetized cats and dogs. Compared to [Ile(8)]-angiotensin II, the new analog has equal antagonistic potency on the isolated system but a much greater potency in vivo. It is assumed that sarcosine in position 1 protects the peptide against enzymatic degradation and enhances its half-life. This study demonstrates that the modification in both positions 1 and 8 are important for the in vivo antagonistic potencies of angiotensin analogs. 相似文献

Problems caused by water shortage in a paddy-field district with a pipeline network system are different from those in a district with an open channel system. Abnormally low rainfall in Japan caused a very serious water shortage in 1994. A survey was carried out in the Hokuriku region, about 300 km in the north of Tokyo, and a typical paddy cultivation area, to determine problems in irrigation practices caused by water shortage and to find countermeasures for the problems. The following results were obtained. A tank model was proposed to estimate water requirements at the field level. The results showed that the amount of rainfall during the irrigation period in 1994 was only 27.1% of an average year and the rate of water sufficiency at the field level was 70.6%. Then, a simulation method was proposed to estimate hydraulic phenomena in a pipeline network system. The result showed that the pipeline network system distributed water to each hydrant unequally during water shortage. Based on simulations, the methods to equalize water supply to each hydrant and to set up reuse system of water were proposed. 相似文献

Lime-N (calcium cyanamide, CaCN2) acts as both fertilizer and pesticide. Lime-N may reduce nitrous oxide (N2O) emission from soil, although its effectiveness and the relative mechanisms are not well understood. The aim of the study was to quantify the effect of lime-N on N2O emission from the acidic soil of tea fields. The study design consisted of two treatments: conventional fertilizer (CF) (application of conventional organo-chemical fertilizer) and lime-N (LN) (application of approximately 53 % of the applied N as lime-N and the remaining as conventional organo-chemical fertilizer). Both treatments had the same amount of N, P2O5, and K2O applied to soil between plant canopies; fertilizer was incorporated into soil. We measured N2O emissions and environmental and microbial parameters of soil between plant canopies and under the canopy of tea plants, including the concentrations of dicyandiamide and cyanamide derived from lime-N. Nitrous oxide emission from soil between plant canopies was lower in the LN treatment than in the CF treatment, and soil ammonium oxidation activity and soil denitrification rate decreased after lime-N application. We applied the acetylene inhibition technique and analyzed isotopomer ratios of N2O; the results of both techniques suggested that denitrification was the major process of N2O production in the soil between plant canopies, despite relatively low water-filled pore space. Cumulative N2O emission over the 366 days of the experiment was 36.0 % lower in the LN treatment than in the CF treatment (P?<?0.05). Matricaria chamomilla L., known as "chamomile", has been used as an herbal tea or supplementary food all over the world. We investigated the effects of chamomile hot water extract and its major components on the prevention of hyperglycemia and the protection or improvement of diabetic complications in diabetes mellitus. Hot water extract, esculetin (3) and quercetin (7) have been found to show moderate inhibition of sucrase with IC50 values of 0.9 mg/mL and 72 and 71 microM, respectively. In a sucrose-loading test, the administration of esculetin (50 mg/kg body weight) fully suppressed hyperglycemia after 15 and 30 min, but the extract (500 mg/kg body weight) and quercetin (50 mg/kg body weight) were less effective. On the other hand, a long-term feed test (21 days) using a streptozotocin-induced rat diabetes model revealed that the same doses of extract and quercetin showed significant suppression of blood glucose levels. It was also found that these samples increased the liver glycogen levels. Moreover, chamomile extract showed potent inhibition against aldose reductase (ALR2), with an IC50 value of 16.9 microg/mL, and its components, umbelliferone (1), esculetin (3), luteolin (6), and quercetin (7), could significantly inhibit the accumulation of sorbitol in human erythrocytes. These results clearly suggested that daily consumption of chamomile tea with meals could contribute to the prevention of the progress of hyperglycemia and diabetic complications. 相似文献

Overfishing can affect life history traits, resulting in population collapse and oftentimes a decrease in length-at-age and maturation size in fish populations. However, little is known about the recovery mechanisms and time scales of these traits in exploited wild populations. In the study reported here, we documented long-term shifts in growth and mature size in Miyabe charr Salvelinus malma miyabei associated with a history of recreational fishing in Lake Shikaribetsu for approximately 80 years. Downsizing in the charr was observed when the charr population collapsed due to intensive recreational fishing. However, subsequent moratoriums and the introduction of fishing regulations, especially the implementation of a catch-and-release policy, during the following 10–30 years facilitated the recovery of population size, length-at-age and mature fish size. This study provides important insights into the biological changes and required recovery time scales of a heavily harvested population and supports management and conservation strategies. 相似文献

The enhancing effect of cross-linked ricin (CL-ricin) on the cell-mediated and humoral immune response of mice to non-viable Mycoplasma pulmonis was studied. The cell-mediated immune response was evaluated by means of the delayed-type footpad swelling, and the humoral immune response by means of the indirect hemagglutination test. Mice pre-treated subcutaneously with non-viable M. pulmonis and CL-ricin showed significantly increased delayed-type footpad swelling when they were injected in the footpad with the same antigen 7 days later. Delayed-type footpad swelling was not detected in mice pre-treated only with non-viable M. pulmonis or CL-ricin followed by footpad injection with non-viable M. pulmonis. Injection of non-viable M. pulmonis in the footpad on Days 3, 7, 14, 21, 28, 35 and 42 after pre-treatment with non-viable M. pulmonis and CL-ricin resulted in significant footpad swelling. Delayed-type footpad swelling was transferred by intravenous injection of spleen cells from mice which had been pre-treated 7 days previously with non-viable M. pulmonis and CL-ricin into non-treated recipient mice. Intravenous injection of anti-mouse thymus cell serum into mice previously pre-treated with non-viable M. pulmonis and CL-ricin reduced the delayed-type footpad swelling significantly. Mice pre-treated subcutaneously with non-viable M. pulmonis and CL-ricin showed a marked increase in serum antibody titers compared with those that received non-viable M. pulmonis alone. 相似文献
