The temporal and spatial distribution of Tomato spotted wilt virus (TSWV) was studied in a 2,085 m2 tomato field with a total of 4,080 plants during 10 weeks from 5 July to 6 September in 2004. First infected plants were detected 4 weeks after the source plants were placed in the field and their number continued to increase by week 7. A total of 147 plants (3.6%) were recorded to be infected according to symptom occurrence and serological test by the end of the experiment. Thrips monitoring was carried out from May 25 to October 25 in the same year. Two thrips species, Frankliniella intonsa (Tryborn) and Thrips tabaci (Lindemann), were determined in the experimental area, with the latter one being the more prevalent (84.9%) species. There was a significant correlation (r?=?0.988, P?<?0.01) between the number of thrips species and the number of infected plants in the field. Majority of infected plants were not located near (0–10 m) an internal virus source within the experimental area, whereas 45.6% of the infected plants were located at a distance of 21–30 m. The number of infected plants at a distance of 21–25 m was significantly higher than at all other distances (P?<?0.05). There was no gradual increase in the distances of TSWV-infected plants within 0–30 m as time progressed. The virus seems to be transmitted by vectors through primary infections and scattered within 30-m distance, but limited secondary infections seem likely to occur after this distance in the case of a small internal virus source. 相似文献

Intracellular pH-regulatory mechanisms are acquired by growing mouse oocytes with meiotic competence, and these mechanisms become fully active when the oocytes develop to the germinal vesicle (GV) stage as shown in CF1 and Balb/c strains mice. On the other hand, there is some evidence showing that intracellular pH-regulatory mechanisms are inhibited at the stages of Metaphase I (MI) and II (MII) oocytes in the CF1 strain mouse and hamster. Since it has been shown that the intracellular pH regulatory mechanism can be functionally different among mouse strains (e.g., CF1, Balb/c), the aim of this study was to investigate the activity of HCO3-/Cl- exchanger (anion exchanger, AE), which protects cells against alkalosis during the meiotic maturation process, in the GV oocyte up to the pronuclear (PN) zygote derived from the Balb/c strain mouse. Intracellular pH (pHi) was recorded using a microspectrofluorometric technique during meiotic maturation stages. KSOM-based solutions were used as culture and recording solutions. AE activity was determined using a Cl- removal assay and was reported as the change in pHi per minute. AE activity was high in GV stage oocytes but was significantly inhibited at the MI and MII stages. AE activity was higher in the PN zygote stage. This activity was significantly inhibited in all oocyte and zygote stages by 4,4'-Diisocyanatostilbene-2,2'-disulfonic acid disodium salt. After alkalosis induction, the pHi of MI and MII stage oocytes did not completely recover; however, almost complete recovery occurred in the GV stage oocytes and PN zygotes. This study was carried out to evaluate milt quality in male Caspian brown trout (Salmo trutta caspius) over the course of the winter spawning season. Milt samples were collected biweekly during December and January. Chemical composition of seminal fluid, sperm production (milt volume, sperm density, spermatocrit,) and sperm motility characteristics (percentage and duration of motility) were measured. Milt volume, sperm density, osmolality, seminal minerals (Ca2+, Mg2+, K+, Na+, Cl?), and total protein gradually decreased over the spawning season. Glucose and triglyceride content of milt did not show significant changes over the spawning season. Milt pH and the percentage and duration of motility were comparatively stable, declining only at the end of the season. Significant positive correlations were found between sperm density and seminal minerals, total protein and spermatocrit; percentage of motile spermatozoa and seminal minerals, total protein; and duration of motility and K+, Cl?, total protein, and pH. Results show that season has a significant influence on milt quality in male Caspian brown trout, with the best milt being available at the beginning of spawning season. 相似文献

ABSTRACT Caspian roach (Rutilus rutilus caspicus) were exposed to 0, 30, 40, 45, 50, 55, 60, 70, 100, 150 200, 250, 300, and 400 ppm formalin to determine LC50. Respectively, 0.5 to 96 h LC50 values were 252 to 49 ppm. The lowest observed effect concentrations (LOEC) and no observed effect concentrations (NOEC) varied between 40 to 150 and 30 to 100 ppm, respectively, across the tested time spectrum. Regression suggested that the concentrations of 88.4, 80.3, 51.2, and 40.1 ppm are maximum concentrations for 0.5, 1, 12, and 24 h bath treatment, respectively, which are not in the range suggested in the literature. 相似文献

Antioxidants have been widely used as additives to provide protection against oxidative degradation of foods by free radicals. The effect of thyme essence (Zataria multiflora Boiss) on the rancidity development in cobia (Rachycentron canadum) fillets during frozen storage was studied. Cobia fillets were treated with thyme essence (250 and 500 ppm) and then stored at??18°C for up to 6 months. Rancidity development was measured by several biochemical indices including free fatty acids (FFA), peroxide value (PV), thiobarbituric acid (TBA), and complemented by sensory analysis (flesh odor, consistency, and appearance). Also, pH and expressible moisture were measured during 6-month storage. Proximate composition was also determined in the first day. TBA, PV, and FFA levels increased in all treatments due to lipid oxidation. Thyme essence showed an antioxidative effect in cobia fillets during frozen storage as indicated by TBA, PV, and FFA levels. Results showed that FFA, primary and secondary oxidation products, expressible moisture (EM), and pH of thyme essence treated samples were significantly lower than those of the control samples (p < 0.05). Thyme essence retarded oxidative changes in frozen cobia fillets, and the best oxidation inhibition was obtained using thyme essence at 500 ppm. 相似文献

Multilayer microencapsulation of fish oil was evaluated by using fish gelatin (treated with or without microbial transglutaminase [MTGase]), chitosan, and maltodextrin for its ability to control lipid oxidation. This study showed that a mixture containing gelatin and maltodextrin obtained the highest percentage of core release. The oxidative stability of the formulated mixtures and the bulk oils was investigated during a period of 60 days. The peroxide value (PV) was assessed as a parameter for primary oxidation products, and p-anisidine (p-AV) and thiobarbituric acid reactive sub-stances (TBARS) was used to analyze secondary oxidation compounds. Observation of oxidation products showed that combinations of gelatin and maltodextrin made by adding MTGase and a mixture of gelatin and chitosan were able to increase the oxidative stability, and increases in PV and p-AV and TBARS were found for all oils. 相似文献

- Many river rehabilitation projects have been criticized for failing to meet their goals or for being insufficiently monitored. There is, therefore, an urgent need to develop robust approaches for assessing treatment efficacy and thus guide the increasing investment in rehabilitation.
- Instream biotopes (formerly called ‘functional habitats’ or ‘mesohabitats’ by different authors) and their macroinvertebrate assemblages were used to assess the effectiveness of entire-channel hydromorphological rehabilitation of a 1.8-km stretch of a lowland stream through the town of Market Harborough, Leicestershire, UK.
- The project successfully enhanced the physical diversity, measured in the rehabilitated reach as the coefficients of variability for channel water depth and width, wet surface area, number of instream biotopes, and biotope diversity.
- The project also enhanced the biodiversity conservation value, measured as macroinvertebrate total density, total biomass, richness, diversity, Ephemeroptera, Plecoptera, and Trichoptera (EPT) richness, EPT diversity, EPT count%, and EPT biomass%, all of which significantly increased following rehabilitation. Chironomidae count% and biomass% significantly decreased following rehabilitation. Rehabilitation was also successful in significantly increasing macroinvertebrate shredder, scraper, and filter-feeder group density.
- Changes in the macroinvertebrate community metrics in the rehabilitated reach were related significantly to changes in the percentages of cover of instream biotopes and increases in biotope diversity in the rehabilitated reach.
- Macroinvertebrate structural and functional metrics can provide quantitative data for assessing reach-level rehabilitation outcomes, if samples are collected in a pre-defined sampling protocol stratified at the instream biotope level.
- The practical implications of this work are that the design of rehabilitation projects, if based upon the recreation of biotope heterogeneity, will succeed in improving biological value and restoring the area to near naturalness if a suitable upstream source of macroinvertebrates for natural recolonization is available. The study shows that the concept of biotopes has an important role to play in river conservation management.
