Transient leucopenia, thrombocytopenia and anaemia associated with severe acute phenobarbital intoxication in a dog

A nine-year-old Labrador retriever dog was admitted to the emergency unit of the Hebrew University Veterinary Teaching Hospital with acute-onset tremors and coma. It had recently ingested a large quantity of phenobarbital and had a high serum phenobarbital concentration. On this basis, a diagnosis of acute phenobarbital intoxication was made. Significant leucopenia, thrombocytopenia and mild anaemia developed on the third day after admission. The leucopenia resolved on day 6 and the thrombocytopenia on day 13. The red blood cell count remained low for the next month. The dog was discharged on day 13 at which time it was ambulatory but weak. It was completely recovered clinically eight days later. In summary, high levels of serum phenobarbital as a result of acute intoxication induced pancytopenia, which improved when the serum phenobarbital level was normalised.     

Effect of Steeping Conditions on Wet-Milling Characteristics of Hard Red Winter Wheat

A batch-wise small-scale wet-processing laboratory for whole wheat kernel has been designed and constructed to produce wheat starch and gluten from wheat grains. Hard red winter wheat kernels were steeped in three steeping media: SO₂ solution, lactic acid, and hydrochloric acid. Acid concentrations of 0.1, 0.3, and 0.5%, were used for SO₂ solutions and hydrochloric acid, and 0.1, 0.6, and 3.0% for lactic acid. After 16, 20, and 24 hr of steeping, the wheat was wet-milled. Yields and protein contents of wet-milling fractions were compared. Both high concentration of steeping media and long steeping time increased the starch yield and decreased the protein contents of the starch. However, the steeping time and acid concentration could be reduced from 24 to 20 hr and from 0.5 to 0.3%, respectively, without any statistically significant difference in starch yields or protein contents of the starch. Consistency and color of the starch were affected by both steeping time and acid concentrations of steeping media.     

Short term soil priming effects and the mineralisation of biochar following its incorporation to soils of different pH

The aim of this work was to determine the magnitude of the priming effect, i.e. short-term changes in the rate (negative or positive) of mineralisation of native soil organic carbon (C), following addition of biochars. The biochars were made from Miscanthus giganteus, a C4 plant, naturally enriched with ¹³C. The biochars were produced at 350 °C (biochar350) and 700 °C (biochar700) and applied with and without ryegrass as a substrate to a clay-loam soil at pH 3.7 and 7.6. A secondary aim was to determine the effect of ryegrass addition on the mineralisation of the two biochars.After 87 days, biochar350 addition caused priming effects equivalent to 250 and 319 μg CO₂-C g⁻¹ soil, in the low and high pH soil, respectively. The largest priming effects occurred at the start of the incubations. The size of the priming effect was decreased at higher biochar pyrolysis temperatures, which may be a way of controlling priming effects following biochar incorporation to soil, if desired. The priming effect was probably induced by the water soluble components of the biochar. At 87 days of incubation, 0.14% and 0.18% of biochar700 and 0.61% and 0.84% of biochar350 were mineralized in the low and high pH soil, respectively. Ryegrass addition gave an increased biochar350 mineralisation of 33% and 40%, and increased biochar700 at 137% and 70%, in the low and high pH soils, respectively. Certainly, on the basis of our results, if biochar is used to sequester carbon a priming effect may occur, increasing CO₂-C evolved from soil and decreasing soil organic C. However, this will be more than compensated for by the increased soil C caused by biochar incorporation. A similar conclusion holds for accelerated mineralisation of biochar due to incorporation of fresh labile substrates. We consider that our results are the first to unequivocally demonstrate the initiation, progress and termination of a true positive priming effect by biochar on native soil organic C.     

Molecular cloning and expression of kidney‐type glutaminase from common carp (Cyprinus carpio) and its up‐regulation by glutamine in primary culture enterocyte

Glutaminase (GLS) is the key enzyme of glutamine (Gln) metabolism and utilization. In this study, a cDNA encoding GLS protein was identified from common carp Cyprinus carpio intestine. The open reading frame of GLS cDNA encodes a polypeptide of 595 amino acids, which shows a high similarity with its zebrafish Danio rerio counterpart. Bioinformatic analysis showed the protein belongs to kidney‐type GLS. The putative protein has glutaminase domain and ankyrin repeats domain, which are highly conserved among vertebrate orthologues. Real‐time quantitative PCR analysis revealed that the abundance of GLS mRNA was the highest in the white muscle, followed by the brain, eyeball and pituitary. Glutaminase was ubiquitously expressed in all intestinal segments of common carp. The activity of GLS did not distribute uniformly along the entire length of the intestine. In primary culture enterocyte, and the expression of GLS mRNA is up‐regulated quickly and effectively by Gln.     

Reduction in the yield and viability of carrot seed in relation to the occurrence of the plant bug Lygus campestris L.

Summary Research was carried out into the cause of a disorder occurring in seed plants of carrots characterized by backward fruits and resulting in a reduced seed yield.It appeared that both fertilization and initial development of the endosperm and embryo were normal, but that later on a sudden check in the growth of the fruit occurred, accompanied by disintegration of the contents of the ovule.Bugs of the species Lygus campestris L. were found to occur in considerable numbers on the seed plants of carrots. In experiments a reduced seed production with the same macroscopic and microscopic symptoms could be obtained when bugs were added to caged carrot umbels.It could be established that the presence of bugs caused not only a reduction in the seed production due to abortion of endosperm and embryo, but also a reduction in the germination percentage of the full-grown seeds due to embryolessness.When the bugs were brought on the umbels before or during anthesis the reduction in seed viability and expecially the reduction in seed production proved to be more serious than when the bugs were added after anthesis.     

A unified approach for estimating population size in capture-recapture studies with arbitrary removals

A unified approach is suggested to estimate the population size for a closed population in discrete time. Individuals can be removed after capture at any time during the experiment. The usual recapture and removal experimentsare shown to be particular cases of the general formulation. The capture probability is assumed to have a logistic function that depends on individual covariates and can be time dependent. The unified approach involves a two-step procedure. A conditional likelihood function is used to estimate the covariates coefficients and a Horvitz-Thompson type estimator to estimate the population size. The asymptotic and small-sample properties of the resulting estimators are in vestigated. A real example is given.
