OBJECTIVE: To determine the effect of endotoxin (lipopolysaccharide [LPS]) on vasoactive mediator production by cultured equine digital vein endothelial cells (EDVECs). SAMPLE POPULATION: EDVECs obtained from forelimb digital veins of 7 healthy adult horses. PROCEDURES: EDVECs were incubated with or without LPS (1 microg/mL) for 0, 2, 4, 6, 22, and 24 hours. The EDVECs were incubated for 18 hours with LPS (10 pg/mL to 1 microg/mL) with or without ibuprofen, cycloheximide, or L-nitroarginine methyl ester. Medium concentrations of prostacyclin, cyclic guanosine monophosphate, endothelin-1, and thromboxane A(2) were determined. Changes in inducible nitric oxide synthase and cyclooxygenase-2 expression were determined. RESULTS: LPS stimulated mean 4.2- and 14.1-fold increases in EDVEC prostacyclin and cyclic guanosine monophosphate production, respectively, after 22 hours. These effects were LPS concentration-dependent (LPS concentrations that induced a response halfway between the maximum response and baseline of 1.50 and 1.22 ng/mL, respectively). The LPS-induced cyclic guanosine monophosphate production was significantly inhibited (to basal concentrations) by L-nitroarginine methyl ester, and prostacyclin production was inhibited by cycloheximide and ibuprofen. Production of thromboxane A(2) by EDVECs was not detected. Endothelin-1 accumulated in the medium, but LPS did not enhance its production. Inducible nitric oxide synthase expression in EDVECs was not detected with the available antibodies, whereas LPS stimulated cyclooxygenase-2 expression in a time- and concentration-dependent manner. CONCLUSIONS AND CLINICAL RELEVANCE: LPS stimulated vasoactive mediator production by equine endothelial cells, which may play a role in LPS-induced digital hypoperfusion. 相似文献

This study was conducted to determine the concordance of results for a pair of structural
isomers, 2-nitropropane (2-NP) and 1-nitropropane (1-NP), using the rat medium-term liver
carcinogenesis bioassay (Ito test) and previously published long-term carcinogenicity
tests. Male F344 rats were given a single intraperitoneal injection of DEN (200 mg/kg
b.w.) to initiate hepatocarcinogenesis. After 2 weeks, they received per os 0, 0.8, 4 or
20 mg/kg/day of 2-NP or 1-NP six times a week and were subjected to two-thirds partial
hepatectomy at week 3. Non-initiated groups receiving 0 or 20 mg/kg/day were also
included. The animals were sacrificed for quantitative analysis of GST-P-positive foci at
week 8. With the highest dose of 2-NP, significantly increased numbers and areas of
GST-P-positive foci were demonstrated as compared with the respective control but were not
noted with 1-NP. In the non-DEN-initiated groups, many small GST-P-positive foci of less
than 0.2 mm in diameter were also induced in the rats treated with 2-NP at 20 mg/kg/day
but were lacking with 1-NP. These results strongly support that 2-NP is a complete

OBJECTIVE: To determine clinical characteristics and mode of inheritance of seizures in a family of Standard Poodles. DESIGN: Case series. ANIMALS: 90 Standard Poodles descended from the same maternal bloodline (30 with probable idiopathic epilepsy [PIE] and 60 without any history of seizures). PROCEDURES: Researchers contacted owners to determine whether dogs had ever had any seizures and, if so, the nature of any such seizures and any potential underlying causes. Dogs were considered to have PIE if they were between 6 months and 7.5 years old at the time of seizure onset and had no evidence of any underlying cause. To determine the mode of inheritance, segregation analyses were designed to allow the family to be analyzed as a whole, as opposed to as nuclear families. Competing models of inheritance were compared statistically for their ability to explain the data. RESULTS: Of the dogs with PIE, 28 (93%) had focal onset seizures with or without secondary generalization. Median age of onset was 3.7 years; 6 dogs were > 5 years old at the onset of seizures. Segregation analyses strongly suggested that PIE was inherited as a simple recessive autosomal trait with complete or almost complete penetrance. CONCLUSIONS AND CLINICAL RELEVANCE: Results suggested that in this family of Standard Poodles, PIE was inherited as a simple recessive autosomal trait with complete or almost complete penetrance. Cytochrome P450 (CYP) is important for metabolism of not only xenobiotics such as drugs, but also endogenous compounds including arachidonic acids. CYP4A11, CYP4F3v2, CYP4F11, and CYP4F45 have been identified in cynomolgus macaque, an animal species widely used for investigation of drug metabolism due to its evolutionary closeness to human. However, their metabolic functions have not been investigated. In this study, proteins were heterologously expressed in Escherichia coli and characterized by metabolic assays using arachidonic acids as substrates that are metabolized by CYP4 isoforms in human. The results showed that all four CYPs metabolized arachidonic acids. Therefore, cynomolgus macaque CYP4A11, CYP4F3v2, CYP4F11, and CYP4F45 are functional enzymes. 相似文献

为研究淋雨和添加剂对苜蓿(Medicago sativa)青贮品质的影响,试验设新鲜和淋雨苜蓿青贮2个因素,添加剂分别为对照、甲酸、绿汁发酵液、改良绿汁发酵液、玉米粉、绿汁发酵液+玉米粉、改良绿汁发酵液+玉米粉7个水平,共14个处理。青贮90 d后,测定各处理的pH、氨态氮、有机酸、营养成分和硝基化合物含量。结果表明:淋雨和添加剂均极显著降低苜蓿青贮饲料pH、氨态氮、乳酸和乙酸含量(P<0.01)。改良绿汁发酵液+玉米粉组的乳酸含量显著高于对照和其他处理组(P<0.05)。淋雨和添加剂显著影响苜蓿青贮饲料的干物质、酸性洗涤纤维、中性洗涤纤维含量和缓冲能值(P<0.05),添加剂对粗灰分、粗蛋白、粗脂肪和水溶性碳水化合物含量有显著影响(P<0.05)。淋雨、添加剂及其交互作用极显著影响苜蓿青贮饲料的硝酸盐和亚硝酸盐含量(P<0.01)。添加剂可改善淋雨苜蓿青贮的发酵品质,其中改良绿汁发酵液+玉米粉组合效果最明显。 相似文献
