Agrobacterium tumefaciens‐mediated transformation (ATMT) is being increasingly recognized as an effective insertional mutagenesis tool in studies of filamentous fungi for gene discovery and functional analysis. We developed and optimized ATMT for 2 Colletotrichum species, Colletotrichum gloeosporioides and Colletotrichum acutatum, the causative agents of Colletotrichum leaf disease in rubber trees in Southern China. A. tumefaciens strain AGL‐1 carrying an ILV1 gene and a green fluorescent protein gene were used to transform the conidia of these 2 Colletotrichum species. The transformation efficiency was correlated with the co‐cultivation duration and bacterial cell concentrations, which reached 300–400 transformants per 1 × 106 conidia after optimization. Southern blot analysis indicated that about 60.0% of the C. gloeosporioides transformants and 46.2% of the C. acutatum transformants had a single copy of T‐DNA in their genomes. Fungal genomic DNA segments flanking the T‐DNA were identified in the transformants through thermal asymmetrical interlaced polymerase chain reaction followed by sequencing. The flanking sequences from 4 C. acutatum and 7 C. gloeosporioides transformants showed moderate or weak homology to the NCBI database entries. Some sequences matching those reported virulence‐related genes. The results suggest that the T‐DNA inserted mutants banks constructed are useful for the discovery of new or important genes and to elucidate their function in the 2 Colletotrichum species. 相似文献

Summary Morphological and chemical examinations of the extractives of the tissues and shake contents of eight Dacrydium and one Podocarpus species have been made. The whitish deposits in heart-shakes are without exception mainly podocarpic acid (PCA). This compound occurs also in the tissue of samples but only when shakes are found nearby. Accordingly PCA is regarded as an anomalous extractive, synthesized in response to those conditions resulting in shake formation. In samples with shakes PCA is present in very small amounts at the sapwood-heartwood boundary and in one sample there were traces even in the innermost sapwood. It was notable that shakes containing deposits can arise in those Dacrydium species with coloured heartwood and with a known tendency for wetwood formation. On the other hand Dacrydium franklinii does not develop coloured heartwood or wetwood or shakes, and the lack of these features may have taxonomic significance.The third author expresses his gratitude to the Division of Building Research, CSIRO, Highett, Vic., for a visiting Fellowship during 1974. 相似文献

Riparian forests are classified as endangered ecosystems in general,particularly in sahelian countries like Burkina Faso because of human-induced alterations and civil engineering works.The modification of this important habitat is continuing,with little attention being paid to the ecological or human consequences of these changes.The objective of this study is to describe the variation of woody species diversity and dynamic in riparian forests on different type of watercourse banks along phytogeographical gradient in Burkina Faso.All woody species were systematically measured in 90 sample plots with sides of 50 m × 20 m.Density,dominance,frequency and species and family importance values were computed to characterize the species composition.Different diver-sity indices were calculated to examine the heterogeneity of riparian forests.A total of 196 species representing 139 genera and 51 families were recorded in the overall riparian forests.The species richness of individuals with dbh ≥ 5cm increased significantly from the North to the South along the phytogeographical gradient and varied significantly between the different types of riparian forests.Similarity in tree species composition between riparian forests was low,which indicates high beta diversity and reflects differences in habitat conditions and topography.The structural characteristics varied significantly along the phyto-geographical gradient and between the different types of riparian forests.The diameter class distribution of trees in all riparian forests showed a reverse "J" shaped curve except riparian forest of stream indicating vegetation dominated by juvenile individuals.Considering the ecological importance of riparian forest,there is a need to delineate and classify them along watercourses throughout the country. 相似文献

Sharks fisheries have declined globally due to over‐ and unregulated fishing. As with many collapsed and unmonitored coastal fisheries, information is difficult to obtain, yet it is important to understand the historical changes determining population trends and evaluate the current status of sharks in order to conserve these vulnerable species. Here, we document for the first time the history and general condition of the shark fisheries of Southern China, specifically Hong Kong, and Guangdong, Fujian and Hainan Provinces. This study shows, through the use of historical literature and anecdotal accounts, including fisher interviews, that all known shark fisheries in the region collapsed between the 1970s and the 1990s. Of the 109 species present historically in the South China Sea, only 18 species were recorded in current market surveys, of which all were landed as bycatch and 65% were below the size of sexual maturity. Markets are dominated by smaller species, including the spadenose shark (Scoliodon laticaudus) and the whitespotted bambooshark (Chiloscyllium plagiosum). Marketed large shark species are almost all below the size of sexual maturation, evidence of growth overfishing and a factor in recruitment overfishing. Some species, like the whale (Rhincodon typus) and basking sharks (Cetorhinus maximus), are clearly vulnerable to local extinction without intervention. Given the inherent vulnerability of sharks and the overfished states of many sharks, there is clearly an urgent need to formulate impacting conservation and management plans for these rapidly declining species in a region that has the highest demand for shark products globally. 相似文献

The degradation behavior of an emerging contaminant is a key factor in its environmental risk assessment. Existing risk assessment methods based on EC degradation data commonly neglect the time-varying volatility of the degradation, the possible correlations in degradation between different ECs, and the estimation errors. To fill the gaps, this paper proposes an EC risk assessment framework based on the Wiener process. We first focus on degradation data from competitive experiments, which are adopted to evaluate a useful risk indicator, i.e., the bimolecular rate constant of a degradation reaction. A two-dimensional Wiener process model is developed to capture the degradation behaviors of the target EC and a reference contaminant in the experiment. Point and interval estimations of desired quantities, including the rate constant and the degradation half-life, are developed. We further extend the model to the multivariate case, which is applicable to waste water treatment where multiple ECs degrade in a mixed solution. A risk indicator for the mixed solution is proposed, based on which a minimal treatment time can be determined. Both point and interval estimation procedures of the risk indicator and the minimal treatment time are proposed. Two EC degradation datasets are used to demonstrate the proposed methodologies. Abstract. Allozyme variation at 32 protein genetic loci was studied in four populations of Macrobrachium nipponense (de Haan) from two contrasting habitats, wholly freshwater lacustrine sites and brackish water riverine areas from different parts of Asia (Hong Kong, Canton, Wuhan, Japan). The average values of observed heterozygosity were 7·45% for lacustrine and 12·30% for riverine sites, the percentage of polymorphic loci (0·99) was 32·5%. The freshwater/lacustrine populations had lower estimates of genetic variability than the river/brackish water populations. The average genetic similarity and genetic distance (Nei's) were 0·92 and 0·084 respectively. The results were compared with those of the only other study on this species. Xyloglucan endotransglycosylase is an identified relaxation factor with functions of easing and extending of plant cell walls.Its activities are directly related to plant growth and elongation of organisms.Anthocephalus chinensis is a tall and fast growing evergreen tree species.We cloned the full cDNA sequence of AcEXT genes which is abundantly expressed in the cambium of A.chinensis.The sequence analysis of nucleotides and amino acids revealed the presence of a 1396 bp full cDNA sequence,including a 960 bp complete open reading frame(ORF) encoding a 320 amino acid protein.The deduced amino acid sequence of AcXET was homologous to the other known XET proteins and contained the conserved EIDFE catalytic site which was specific to all the XETs.Our data should serve as a foundation for further insight into AcXET gene molecular mechanisms during wood formation and cell wall engineering of woody plants. 相似文献

La cladosporiose demeure l'une des principales maladies des cultures de tomate en serre car la lutte chimique reste peu satisfaisante. Elle est parfois naturellement ralentie par l'installation spontanée d'un hyperparasite de Fulvia fulva (syn. Cladosporium fulvum), Hansfordia pulvinata, antagoniste actif de l'agent pathogène. L'élaboration d'une méthode de lutte biologique a été envisagée. Une production industrielle de ce biopesticide est en cours de mise au point. En serres de production, 80% ou plus des taches de cladosporiose sont recouvertes par l'hyperparasite. Cependant, l'efficacité du controle est limitée par l'incapacité de H. pulvinataà entrer en contact avec F. fulva lors de sa phase biotrophe ainsi que par la faible autodissémination de l'hyperparasite. Un apport de phoséthyl-Al (1.6 g l-1) a amélioré l'efficacité de la méthode. Ce fongicide, compatible avec H. pulvinata, inhibe la germination des spores de F. fulva. Dans cette lutte intégrée, l'hyperparasite abaisse la pression d'inoculum du parasite et le fongicide limite les nouvelles contaminations. Au vu des premières expérimentations en serre, cette méthode pourrait etre préconisée en début d'attaque de cladosporiose et devrait réduire la fréquence des traitements, favorisant en cela une large lutte intégrée et l'emploi des auxiliaires. 相似文献
