Isoelectric focusing of infectious particles of four strains of porcine pseudorabies viruses (Indiana, Iowa, Shope and an avirulent live-virus vaccine strains) are described. The pseudorabies virus strains exhibited great mobility in the electric field typical of viruses of the herpes group. Strains were considered electrophoretically homogeneous based on their respective isoelectric points. Maximal virus infectivity was concentrated in reproducible, stationary zones representing 73 to 86% of the total virus infectivity initially applied throughout the gels. All pseudorabies virus strains during processing and after isoelectric focusing retained their ability as whole complete particles to typically infect porcine kidney cell cultures. Virus from gel fractions produces foci in cell cultures that specifically reacted with pseudorabies virus fluorescent antibody conjugate. Prevention of foci could be demonstrated by neutralizing with pseudorabies virus monospecific antiserum. Maximal infectivity titers were demonstrated to be directly related to isoelectric points. Strain differences, in relation to virulence in swine, apparently is not related to isoelectric points.  相似文献

Abstract. This paper reports results from a four year study to investigate the suitability of porous ceramic cups to measure solute leaching on shallow chalk soils. Measurements were carried out in one field following surface applications of nitrate and bromide tracers and in two fields after only bromide was applied. Soil water samples were collected from porous cups at 30,60 and 90cm depth after every 25 mm of drainage, and soil samples from 0–30, 30–60 and 60–90 cm were collected monthly eachwinter. Soil matric suctions andvolumetric moisture content were measured in one winter. Leaching losses, measured with ceramic cups were compared with those measured by soil analysis. Porous cups installed in chalk at 60 and 90 cm depth were only able to collect samples regularly when soil matric suctions were less than 15 kPa. Water held at such low suctions is likely to move quickly through relatively large fissures in the chalk. The slow rate of equilibration between solute concentrations in water moving in macrofissures and those in water moving through micropores of the chalk matrix, means that porous cups may not provide good estimates of leaching losses if they are installed in chalk rock.  相似文献

An experiment was conducted with 4 groups of hens: a control group of laying hens, a group subjected to food deprivation for 7 d, a group subjected to food deprivation for 7 d, then re-fed for 10 d and a group subjected to food deprivation for 7 d then re-fed for 10 d with two chicks per hen introduced during the last 7 d of refeeding. Food deprivation provoked the involution of the oviduct and ovarian regression, as well as a decrease in the plasma concentrations of luteinising hormone (LH) and sex steroids (progesterone, testosterone and oestradiol). After 3 d of re-feeding, there was a significant increase in the plasma concentrations of LH and steroid hormones. This phenomenon was even more marked after 10 d of re-feeding; most of the hens of the third group which were not given chicks, were at the point of lay. The presence of chicks resulted in the expression of maternal behaviour and suppressed a rapid return to laying. This was especially marked in hens showing typical maternal behaviour traits, for which the morphological and endocrine measurements indicated a decrease in the activity of the hypothalamo-hypophyseal-gonadal axis.  相似文献

Reproductive data were collected on 4,596 cow exposures in a rotational crossbreeding study that spanned four generations (1970 through 1988). Angus (A), Brahman (B), Charolais (C) and Hereford (H) breeds were included in three two-breed (A-B, C-B and H-B), three three-breed (A-B-C, A-B-H and C-H-B) and one four-breed (A-B-C-H) rotational mating system. Contemporary straightbreds were produced each generation. Reproductive traits of interest were calving rate, calf survival, weaning rate, calf birth date and calving assistance. Overall means and standard errors for calving rate, calf survival, weaning rate, calf birth date and calving assistance were 82.1 +/- .6%, 92.8 +/- .4%, 76.2 +/- .6%, 50.4 +/- .3 d and 4.2 +/- .3%, respectively. Rotational mating systems had similar calving and calf survival rates that were greater (P less than .01) than those of straightbreds. Three- and four-breed rotation systems had similar weaning rates (81.5 +/- 1.1 and 80.8 +/- 2.0%) that were higher than those for two-breed rotations (77.1 +/- 1.2%). Two- and three-breed rotation systems had fewer assisted births (3.5 +/- .6 and 2.2 +/- .6%) than straightbreds (5.5 +/- .6%) and the four-breed rotation (5.4 +/- 1.0%). Three-breed rotation calves were born earlier in the calving season (46.8 +/- .6 d; P less than .05) than straightbred, two-breed or four-breed rotation calves (51.8 +/- .6, 52.1 +/- .6 and 51.1 +/- 1.0 d). Hereford-Brahman two-breed rotation cows had higher (P less than .05) calving and weaning rates than A-B or C-B two-breed rotation cows (87 +/- .4 vs 81.9 +/- 1.9 and 81.5 +/- 1.8%; 81.7 +/- 2.0 vs 76.4 +/- 2.1 and 74.1 +/- 2.0%). All three-breed rotation combinations had similar calving, calf survival and weaning rates, assistance at calving and calf birth dates. Rotational combinations tended to be more superior (P less than .05) to weighted purebred means for calving, calf survival and weaning rates than for calf birth date and calving assistance.  相似文献

Fusarium moniliforme culture material containing fumonisin B1 at 300 mg/kg was incorporated into a broiler starter ration and fed ad libitum to 1-day-old broiler chicks for 2 weeks in two experiments. Clinical features of the disease produced included diarrhea, a 19% reduction in body weight, a 30% increase in relative liver weight, and a worsening of feed conversion by 20 points at 2 weeks of age. Histologically, chicks fed fumonisin had multifocal hepatic necrosis, biliary hyperplasia, muscle necrosis, intestinal goblet-cell hyperplasia, and rickets. Simultaneous feeding of 0.5% aluminosilicate had no effect on the clinical disease or lesions. The clinical disease and lesions induced mimicked those of a viral enteritis.  相似文献

A dog being treated for demodicosis with ivermectin was presented for intermittent vomiting. The vomiting progressed to hematemesis and an underlying coagulopathy was diagnosed. The etiology of the coagulopathy was determined to be ingested brodifacoum. Ultrasound evaluation of the abdomen revealed thickened gastric wall that was suspected to be intramural hemorrhage. Most likely, the intramural hemorrhage and resulting thickening of the stomach wall led to the clinical signs and metabolic alkalosis. This case represents an typical presentation of hemorrhage secondary to rodenticide intoxication. (J Vet Emerg Crit Care 2001; 11(1):27–31, 2001).  相似文献

To determine the transmissibility of chronic wasting disease (CWD) to cattle and to provide information about clinical course, lesions, and suitability of currently used diagnostic procedures for detection of CWD in cattle, 13 calves were inoculated intracerebrally with brain suspension from mule deer naturally affected with CWD. Between 24 and 27 months postinoculation, 3 animals became recumbent and were euthanized. Gross necropsies revealed emaciation in 2 animals and a large pulmonary abscess in the third. Brains were examined for protease-resistant prion protein (PrP(res)) by immunohistochemistry and Western blotting and for scrapie-associated fibrils (SAFs) by negative-stain electron microscopy. Microscopic lesions in the brain were subtle in 2 animals and absent in the third case. However, all 3 animals were positive for PrP(res) by immunohistochemistry and Western blot, and SAFs were detected in 2 of the animals. An uninoculated control animal euthanized during the same period did not have PrP(res) in its brain. These are preliminary observations from a currently in-progress experiment. Three years after the CWD challenge, the 10 remaining inoculated cattle are alive and apparently healthy. These preliminary findings demonstrate that diagnostic techniques currently used for bovine spongiform encephalopathy (BSE) surveillance would also detect CWD in cattle should it occur naturally.  相似文献
