Seeing with a new light: synchrotron radiation

The special properties of synchrotron light are leading to a rapid increase in its utilization for both research and technology. At wavelength in the ultraviolet region of the broad spectrum in these beams a number of atomic, molecular, and solid-state spectroscopies are being pursued; soft x-rays are being used for spectroscopy, lithography, microscopy, and topography; at still shorter wavelengths, advantage is taken of scattering properties to probe the structure of matter. Characteristics of synchrotron radiation and of the sources which produce it are described, and some typical investigations and applications are presented to suggest the versatility of these sources.     

The unusual nature of recent snowpack declines in the North American cordillera

In western North America, snowpack has declined in recent decades, and further losses are projected through the 21st century. Here, we evaluate the uniqueness of recent declines using snowpack reconstructions from 66 tree-ring chronologies in key runoff-generating areas of the Colorado, Columbia, and Missouri River drainages. Over the past millennium, late 20th century snowpack reductions are almost unprecedented in magnitude across the northern Rocky Mountains and in their north-south synchrony across the cordillera. Both the snowpack declines and their synchrony result from unparalleled springtime warming that is due to positive reinforcement of the anthropogenic warming by decadal variability. The increasing role of warming on large-scale snowpack variability and trends foreshadows fundamental impacts on streamflow and water supplies across the western United States.     

Coronary Disease in Spawning Steelhead Trout Salmo gairdnerii

Coronary degeneration was absent in young trout taken in fresh water and rare in immature fish at sea, but the incidence and severity were sharply greater in migrating fish and almost uniformly present in spawning fish. Several fish taken after they had reentered salt water after spawning had no lesions; lesions in fish taken during their second spawning migration were not cumulative. These facts suggest that the process is reversible.     

Public perceptions of natural disturbance in Canada’s national parks: The case of the mountain pine beetle (Dendroctonus ponderosae Hopkins)

Since the 1990s, the mountain pine beetle (MPB) population has exploded in western Canada. In national parks, MPB has the potential to impact visual quality and safety of visitors, and to spread beyond park boundaries to the industrial forest landbase. Control measures have been initiated in some parks to lessen these impacts. A study was undertaken to examine public attitudes, knowledge, issue salience, and management preferences for MPB in Banff and Kootenay national parks. Data were collected by mail survey in 2003 from 1385 residents living in or near the parks. MPB was an important issue for the majority of respondents and they had low knowledge of MPB, expressed negative attitudes towards it, and supported measures to control it. Preferred control measures included those directed at the current infestation. Proactive approaches in uninfested forests were generally not supported. Issue salience and knowledge were the best predictors of attitudes toward the MPB. Attitudes were the best predictors of support for no intervention in beetle infestations in national parks. Management implications include the lack of knowledge and support for natural disturbance and ecological integrity policies in national parks.     

Inherited hyperchylomicro-naemia in the cat: Lipoprotein lipase function and gene structure

Hyperchylomicronaemia was identified in a four-week-old Siamese kitten with lethargy, in-appetence, hindlimb ataxia and profound anaemia. The kitten was euthanased and at necropsy a thrombus was found occluding the caudal aorta. Two littermates later presented with lethargy, inappetence and hypertriglyceri-daemia which resolved after being weaned on to a low fat diet. A similar condition was subsequently diagnosed in a kitten born to the same sire but a different queen. The expression of hyperchylomicronaemia in two related litters was suggestive of an inherited, familial defect in the function of lipoprotein lipase (LPL). The activity of this enzyme was reduced in all three parents, the two recovered cases and two related, but apparently unaffected kittens, compared with a control group of unrelated cats belonging to the breeder. This reduction in activity was not attributable to defective activation of LPL by its serum cofactor apolipoprotein C-II or the presence in plasma of a factor that inhibited LPL. The gene that codes for LPL was examined by restriction fragment length polymorphism analysis using a human LPL cDNA probe. The results showed that the cat has a similar, but not identical, LPL gene to man. However, there were no differences in the restriction fragment patterns obtained from affected, unaffected and control animals.     

Effect of general anesthesia and minor surgical trauma on urine and serum measurements in horses

OBJECTIVE: To characterize the effect of general anesthesia and minor surgery on renal function in horses. ANIMALS: 9 mares with a mean (+/- SE) age and body weight of 9+/-2 years and 492+/-17 kg, respectively. PROCEDURE: The day before anesthesia, urine was collected (catheterization) for 3 hours to quantitate baseline values, and serum biochemical analysis was performed. The following day, xylazine (1.1 mg/kg, IV) was administered, and general anesthesia was induced 5 minutes later with diazepam (0.04 mg/kg, IV) and ketamine (2.2 mg/kg, IV). During 2 hours of anesthesia with isoflurane, Paco2 was maintained between 48 and 52 mm Hg, and mean arterial blood pressure was between 70 and 80 mm Hg. Blood and urine were collected at 30, 60, and 120 minutes during and at 1 hour after anesthesia. RESULTS: Baseline urine flow was 0.92+/-0.17 ml/kg/h and significantly increased at 30 and 60 minutes after xylazine administration (2.14+/-0.59 and 2.86+/-0.97 ml/kg/h respectively) but returned to baseline values by the end of anesthesia. Serum glucose concentration increased from 12+/-4 to 167+/-8 mg/dl at 30 minutes. Glucosuria was not observed. CONCLUSIONS AND CLINICAL RELEVANCE: Transient hyperglycemia and an increase in rine production accompanies a commonly used anesthetic technique for horses. The increase in urine flow is not trivial and should be considered in anesthetic management decisions. With the exception of serum glucose concentration and urine production, the effect of general anesthesia on indices of renal function in clinically normal horses is likely of little consequence in most horses admitted for elective surgical procedures.     

Dyserythropoeisis, sideroblasts/siderocytes and hemoglobin crystallization in a dog

Dyserythropoiesis characterized by enhanced intramedullary destruction, pathologic sideroblasts and siderocytes, and hemoglobin crystallization was detected in a female Cocker Spaniel presented for poor exercise tolerance. Examination of peripheral blood revealed intraerythrocytic crystals, granulation of erythrocytes, nucleated erythroid cells, reticulocytosis and marked variation in erythrocyte morphology in the absence of anemia. Bone marrow examination revealed sideroblasts, a low M:E ratio and evidence of enhanced intramedullary destruction of erythroid cells. Electron microscopy of peripheral blood and bone marrow confirmed pathologic mitochondrial iron accumulation in erythroid cells and the presence of intraerythrocytic hemoglobin crystals. A cause for the hematologic changes was not identified. After the animal became clinically normal, siderocytes disappeared from peripheral blood but intraerythrocytic crystals and reticulocytosis persisted.
