Effect of prostate-specific membrane antigen on phospho-ERK,growth and migration of prostate cancer LNCaP cells

AIM: To study the expression of prostate-specific membrane antigen (PSMA) on the level of phospho-ERK, cell growth and migration of prostate cancer LNCaP cells.METHODS: The method of silencing PSMA was established by lentivirus-mediated RNAi in our early experiment. The cells were divided into 3 groups.In experimental group, the expression of PSMA in LNCaP cells was stably blocked by lentivirus-mediated RNAi. In negative control group, the cells were transfected with lentivirus-mediated control RNAi (without any interference to PSMA).The normal LNCaP cells served as blank control. The cells in these 3 groups were cultured in both 2 environments: normal medium and medium with PD98059 (an inhibitor of ERK phosphorylation). The phospho-ERK was detected by Western blotting and immunocytochemistry. Furthermore, the growth and migration of the cells were evaluated by MTT and transwell assays,respectively.RESULTS: In normal medium, the expression of phospho-ERK was attenuated in experimental group (P<0.05) and the quantity of "positive" cells was less than those in other 2 groups (P<0.05). Furthermore, the growth curves of the cells showed that the growth ability in experimental group was significantly decreased (P<0.05, after 48 h) and the migration ability in experimental group was reduced (P<0.05). In the inhibitory medium, the cells in all 3 groups expressed phospho-ERK at a lower level. Moreover, the abilities of growth and migration in these 3 groups were poorly displayed. These inhibitory effects on phosphorylation of ERK were similar to the cells in experimental group cultured in normal medium.CONCLUSION: PSMA may play a role in up-regulation of phospho-ERK and it may take an advantage in growth and migration of prostate cancer LNCaP cells.     

螺虫乙酯对烟粉虱的防治效果评价

采用室内毒力测定与田间试验相结合的方法,研究了螺虫乙酯（Movento）对烟粉虱的防治效果。室内毒力测定结果表明：96 %螺虫乙酯原药对烟粉虱卵和若虫都具有非常高的杀虫活性,LC50分别为18.91 mg?L-1和14.97 mg?L-1,与对照95 %吡虫啉没有显著差异|但对烟粉虱成虫的杀虫效果并不十分理想,LC50达到了453.34 mg?L-1,且显著高于对照95 %吡虫啉。田间试验结果表明：24 %螺虫乙酯悬浮剂（SC）对烟粉虱有良好的防效和持效性,且螺虫乙酯与Biopower（助剂）混用的防效明显优于螺虫乙酯单独使用的效果|螺虫乙酯与0.2 %〔0.24 g?（667 m2）-1〕Biopower混用,药后1～28 d对烟粉虱的防效分别为64.91 %～66.92 %、82.00 %～82.59 %、88.11 %～89.97 %、89.99 %～93.94 %和91.10 %～93.56 %。     

黄土地区深路堑边坡稳定性分析及抗滑桩设计

原状黄土的抗剪强度与黄土的干容重、含水量、粘粒含量等物理力学性质有关.通过对具体参数指标的分析,确定了不同区域内黄土的土性参数,进而提出了深路堑边坡的稳定性分析及打滑桩设计.     

Habitat and food choice of the critically endangered cao vit gibbon (Nomascus nasutus) in China: Implications for conservation

Knowledge of habitat and feeding ecology is essential for developing an effective conservation management plan for threatened primates. Despite having been rediscovered nearly a decade ago, very little is known of the critically endangered cao vit gibbon (Nomascus nasutus) or its habitat. We analysed forest cover, made direct observations and conducted interviews of local elders to better understand forest changes and to determine the forest characters and species important to the survival of the gibbon. Interviews indicated human-induced forest resource exploitation focused on species-specific and larger trees most easily accessed. Gibbon forest habitat comprised four primary forest types. The mean canopy height over the whole site was 10.52 m. Gibbons consumed 81, or nearly half of the tree and liana species recorded in the site; however, only 19 species provide 77.8% of the diet. Six of the 19 food species were logged for different reasons throughout the history of the site. We conclude that effective conservation management of primates with highly limited distributions, focused dietary needs, and in degraded ecosystems will require active forest restoration, such as planting important food species in degraded sites.     

Identification and anti-human glioblastoma activity of tagitinin C from Tithonia diversifolia methanolic extract

The Tithonia diversifolia methanolic extract (TDM), which showed antiproliferative activity against human glioblastoma U373 cells, with an IC50 value of 59.2±3.7 μg mL(-1), was passed through silica gel chromatography and successively eluted with different percentages of EtOAc/hexane. The 10-60% EtOAc/hexane subfractions, which exhibited a comparatively higher antiproliferative activity, were isolated, and then structural identification was proceeded with 1H nuclear magnetic resonance. The isolated compound was tagitinin C, a kind of sesquiterpenoid. The IC50 value was 6.1±0.1 μg mL(-1) in U373 treated with tagitinin C. In flow cytometric analysis and inhibition of pan-caspase, the results showed that the anti-glioblastoma effect was apoptosis-independent. In PARP, p-p38, ULK1, and LC3-II expression, the anti-glioblastoma induced by tagitinin C was likely via autophagy. In the ULK1 siRNA transfection experiment, autophagy blockade counteracted the suppression induced by tagitinin C. The result suggested that tagitinin C induces U373 cell death dependent upon autophagy under certain conditions.     

工业区河涌污染与生态危害评价——以佛山三水大塱涡涌为例

在对工业区大塱涡涌水质及底泥重金属进行调查与分析的基础上,评价了底泥重金属潜在生态危害。结果表明,大塱涡涌水体COD、氨氮、总磷、DO与重金属Cr均超过地表水Ⅴ类水体标准,属于劣Ⅴ类水体;底泥重金属Cu、Zn与Cd平均含量超出土壤环境质量三级标准,且重金属Cu、Zn、Pb平均含量超过农用污泥污染物控制标准,该河段疏浚底泥不宜作为肥料直接施用于农田;大塱涡涌表层底泥重金属的潜在生态危害程度总体为强,几种重金属潜在生态危害平均程度从大到小为：Cd〉Cu〉Pb〉Zn〉Cr。     

烟草花几何建模研究

为实现花冠联合植物的花朵几何建模,以烟草花为例,提出了一种基于球B样条曲线和参数曲面的花朵造型方法。通过对烟草花形态结构的观测分析,分别对雌蕊、雄蕊、花柄、花冠和花萼的几何形状进行了数学描述,提取了模型控制参数;结合颜色渲染色和整个花的拓扑信息,实现了烟草花整个模型的建立。实验结果表明,该方法可以有效地对烟草花进行模拟,可控性强,建立的模型有较高真实感,为其他花冠或花萼联合的植物几何建模提供了方法。     

考虑光照条件的蛋壳蛋心颜色识别方法

研究了机器视觉识别中光照条件对蛋壳蛋心颜色的影响;根据白壳与青壳蛋透光性的差异,对比分析了鸭蛋不同壳色在不同光照强度下的颜色信息,找出了白壳蛋与青壳蛋图像信息差异最大的参数以及对应的光照条件.同时,分析了与蛋黄颜色相关性较大的特征参数及光照条件.实验表明:光照条件的亮度等级为120时,鸭蛋图像的可分度最好,利用形心区域的尺分量来判断蛋壳的颜色,计算量减少,分类准确,易实现;白壳蛋在180亮度及青壳蛋在210亮度等级下,根据形心区域的H分量建立判别模型,预测精度高.     

乌梁素海表层底泥污染特征分析

通过系统取样对乌梁素海表层底泥进行了理化性质分析,并利用MAPGIS软件,研究了表层底泥营养盐空间分布特征,并用底泥肥力评价了表层底泥中营养盐污染程度.结果表明,乌梁素海表层底泥中营养盐的空间分布特征呈现出北高南低,西高东低的差异,底泥中营养物质有机质、总氮和总磷存在明显的富集作用,与国内典型富营养化严重湖泊相比,处于...     

Deg2蛋白酶在拟南芥光损伤D1降解及光保护中的作用

己有研究表明叶绿体内有200种蛋白酶,然而,多数蛋白酶的作用机制尚不清楚,尤其哪些蛋白酶参与了D1蛋白周转。其中Deg2蛋白酶体外实验证明,其参与了光损伤D1蛋白的的初步剪切。为了进一步研究Deg2蛋白酶在植物体内的作用机制,我们筛选了拟南芥De醇蛋白酶功能缺陷型突变体。在120μmol·m-2：·S^-1光照生长条件下,出萨突变体与野生型的生长曲线基本一致;在进一步的高光胁迫（1800μmol·m-2·S^-1）处理及相同的光胁迫处理条件下,无论林可霉素存在与否,突变体PSⅡ的最大光化学效率（Fv／Fm）都和野生型没有区别;利用蛋白免疫印迹实验同样证明了光损伤D1蛋白的降解速度在cfe霞突变体和野生型之间也没有明显区别。我们认为Deg2蛋白酶在光抑制情况下对于光损伤D1蛋白的降解以及PsⅡ的修复不是必需的。
