灵芝原生质体融合子的SRAP分子标记鉴定   总被引:2,自引:0,他引:2  
为验证新型SRAP分子标记技术用于灵芝(Ganoderma lucidum)原生质体融合子鉴定的可行性,通过亲本菌株赤芝05、06对153个SRAP引物组合进行筛选,应用经过初筛和复筛得到的4个清晰稳定、且含有两个亲本各自特异扩增条带的SRAP引物组合,对2个亲本菌株、2个只与亲本05有拮抗的融合子、2个只与亲本06有拮抗的融合子、30个与双亲均有拮抗的融合子进行SRAP扩增.结果表明,2个只与亲本05有拮抗的融合子仅扩增出06的特异条带,2个只与亲本06有拮抗的融合子仅扩增出05的特异条带,30个与双亲均有拮抗的融合子中,有7个融合子同时扩增出05和06的特异条带,证明此7个融合子应是05和06融合产生的菌株,也证明SRAP标记可以用于灵芝原生质体融合子的鉴定.  相似文献

AIM: To explore the protective effect of phytosterol ester (PSE) on aortic aging in rats. METHODS: The female SD rats (12 months old, n=42) were randomly divided into control group, model group and PSE group. During the experiment, the rats in control group, model group and PSE group were treated with basic feed, high-fat diet (HFD) and HFD with 2% PSE (W/W) for 6 months, respectively. The morphological changes of the aorta were observed by HE staining and Masson staining, and the absolute area of smooth muscle cells and collagen fiber in the vascular wall were measured by image analysis. The levels of advanced glycosylation end products (AGEs), malondialdehyde (MDA), superoxide dismutase (SOD) and catalase (CAT) in the plasma were detected. The expression of silent information regulator 1 (SIRT1) and peroxisome proliferator-activated receptor γ (PPARγ) at mRNA and protein levels in the vascular tissue was determined by real time PCR and Western blot, respectively. RESULTS: PSE significantly lowered plasma TC and LDL-C, and increased plasma HDL-C level (P<0.05), but had no effect on plasma TG level. PSE significantly attenuated the thickening of intima and media of aging aortic, and decreased the migration of vascular smooth muscle cells (VSMC) and the amount of VSMC and collagen fiber in the aorta (P<0.05). PSE significantly reduced the contents of AGEs and MDA (P<0.05), but had no effect on the activity of SOD and CAT in the plasma. PSE also down-regulated the expression of PPARγ and up-regulated the expression of SIRT1 (P<0.05). CONCLUSION: PSE is able to attenuate the senescence process in the aorta by reducing the production of reactive oxygen species in plasma, and activating SIRT1, or inhibiting the expression of PPARγ in vascular tissues.  相似文献

AIM: To investigate the influence of Sini decoction (SND) on the proliferation and apoptosis of rabbit abdominal aorta smooth muscle cells after ballon injury and discuss the effect of vascular smooth muscle cell's (VSMCs) proliferation and apoptosis in post-percutaneous coronary intervention (PCI) restenosis (RS) and the feasibility of SND preventing post-PCI RS. METHODS: The animal model of rabbit abdominal aorta ballon injury was set up and the therapertic group was treated with SND. The shape of proliferative and apoptotic cell were investigated by electron microscope. Immunohistochemistry staining was performed using α-actin,PCNA and Cyclin E monoclonal antibodies. In situ Cell Death Detection Kit was used to identify apoptotic cells. Abdomial aorta angiography was operated in the 84th day subgroup and the stenosis degree was evalued by quantitative angiographic analysis. RESULTS: As compared with the control group, the therapeutic group displayed a lower proliferative percentage and a higher apoptosic percentage (P<0.05). Moreover, the apoptosic peek time was on the 14th day after operation,which was longer than the control group. CONCLUSION: SND effectly inhibited the proliferation of VSMCs and iuduced apoptosis in VSMCs.  相似文献

AIM: To investigate the effect of Sini decoction (SND)on the ultrastructure of small intestinal epithelial cells in rats with intestinal ischemia-reperfusion. METHODS: Thirty-two Sprague-Dawley rats of both sexes were randomly divided into 4 groups: (1) Control group in which sham operation was performed; (2) Model group in which intestinal I/R was produced by clamping super mesenteric artery(SMA) for 1 hour and declamping SMA for 3 hours; (3) SND1 group in which SD (0.6 g/200 g rat) was given via stomach tube 3 d before intestinal I/R; (4) SND2 group in which SD (1.2 g/200 g rat)was given via stomach tube 3 d before intestinal I/R. A strip of small intestine was taken from distal end of ileum for electron microscopic examination. The two-dimensional structural parameters and three-dimensional structural parameters of mitochondria were calculated. RESULTS: (1)Morphological changes of small intestine: In control group, epithelial cells were orderly arranged, with normal mitochondria and intestinal villi. In model group, the gaps between epithelial cells widened. There were a lot of apoptotic cells. Microvilli were short and swelled. Mitochondria were swelled obviously with broken ridges. Endoplasmatic reticulum was severely dilated. In SND1 and SND2 groups, microvilli and epithelial cells were orderly arranged relatively, mitochondria was slightly swelled. (2) Structural parameters of mitochondria: In model group, there were the least mitochondria and the swelling of mitochondria was severe. In SND1 and SND2 groups, the mitochondria was more than that of model group and the swelling were slight. CONCLUSION: Sini decoction can protect small intestine from ischemia-reperfusion injury without dose-dependent effect.  相似文献

以草地早熟禾品种——新格莱德成熟种子为供试材料,在含3.0mg/L2,4-二氯苯氧乙酸(2,4-D)、0.5mg/L6-苄氨基嘌呤(6-BA)的MB5培养基中进行胚性愈伤组织诱导的培养。并从生长了7~9个月的胚性愈伤组织中分离出原生质体,将该原生质体置于KM8P培养基(含3.0mg/L2,4-D、0.5mg/L6-BA、100mg/L水解酪蛋白、100mg/L水解乳蛋白、1%蔗糖、0.4mol/L甘露醇)中进行了液体浅层培养。结果表明,新格莱德原生质体在上述KM8P培养基中培养3d后出现第1次细胞分裂,2~3周后形成小细胞团,此时添加低渗培养液2~3次,小细胞团持续分裂并形成愈伤组织。当愈伤组织块长至3~5mm时,转入固体培养基MS+3.0mg/L2,4-D+0.5mg/L6-BA和MS+0.5mg/L萘乙酸(NAA)+5.0mg/L6-BA上进行培养,使其细胞增殖和分化,且逐步形成完整的植株。  相似文献

竹笋夜蛾是我国重要的一类竹子笋期害虫,种类多,形态相似,难以区分,为更为准确地识别主要竹笋夜蛾的种类,本研究通过野外取样和室内饲养,对竹笋基夜蛾Kumasia kumaso、竹笋禾夜蛾Oligia vulgaris、笋连秀夜蛾Apamea repetita conjuncta及竹笋秀夜蛾Apamea apameoides 4种竹笋夜蛾卵、幼虫趾钩、臀棘及成虫翅脉和生殖器形态进行了观察和比较。结果表明,竹笋基夜蛾在卵形态、幼虫体色和雌雄生殖器形态上与其他3种竹笋夜蛾形态差异明显,其余3种夜蛾幼虫体表背线和亚背线形态有差异,趾钩、臀棘、翅脉和雌、雄外生殖器形态差异不显著。本研究明确了我国南方竹区4种竹笋夜蛾的形态差异,为竹笋夜蛾的精准防控提供了参考。  相似文献

为探明不同氮水平下秸秆、生物质炭添加对陇中黄土高原旱作农田土壤酸解有机氮组分的影响,2014年在定西市安定区李家堡镇布设的不同氮水平下秸秆、生物质炭添加定位试验(共9个处理),利用Bremner分级法,对该试验2018年收获后的土壤有机氮组分进行测定与分析。结果表明:在0~30 cm土层(0~5、5~10、10~30 cm土层),各处理酸解总有机氮、酸解氨态氮、酸解氨基酸态氮、酸解未知态氮含量均随土层的加深而降低,酸解氨基糖态氮含量随土层的加深而增加;较之无炭处理(CN0、CN50、CN100处理的均值),生物质炭添加(BN0、BN50、BN100处理的均值)处理可提升酸解总有机氮含量10.12%、9.14%、7.61%(土层由上至下),提升酸解氨态氮含量15.02%、16.25%、17.19%(土层由上至下),提升酸解氨基酸态氮含量13.31%、11.84%、8.74%(土层由上至下),其中BN100处理下对其提升效应最显著;较之无炭处理(CN0、CN50、CN100处理的均值),秸秆添加处理(SN0、SN50、SN100处理的均值)可提升酸解氨基糖态氮含量26.46%、26.51%...  相似文献

通过饼肥与化肥不同比例配施研究得出,河南烟区中等肥力的褐土上以50%芝麻饼肥氮 50%化肥氮处理与单施化肥相比,提高了下、中、上3个部位烟叶饱和脂肪酸相对含量,降低了不饱和脂肪酸含量,表现为豆蔻酸和月桂酸相对含量增加,而亚麻酸和亚油酸含量降低。中部叶、上部叶石油醚提取物含量也得到提高,烟叶糖碱比适宜,化学成分较协调,烟叶品质得到改善,并缩小了不同部位间质量差异。  相似文献
