The susceptibility of bovine udder quarters colonized with Corynebacterium bovis to experimental infection with Staphylococcus aureus or Streptococcus agalactiae. 下载免费PDF全文
Twenty bovine udder quarters colonized with Corynebacterium bovis SR6 and 20 uncolonized quarters were challenged by inoculation of Staphylococcus aureus Newbould 305 (ATCC 29740) into the teat cistern. The percentage of infection in quarters colonized with C. bovis (50%) was significantly lower than that in controls (100%). By similar challenge no significant difference was observed between the percentage of infection with Streptococcus agalactiae ATCC 27956 in 33 quarters colonized with C. bovis (70%) compared to 33 controls (87.9%). A total of 37 quarters colonized with C. bovis and 37 control quarters were challenged with Staph. aureus Newbould 305 (ATCC 29740) and Maxi (ATCC 27543) and Strep. agalactiae (ATCC 27956) by exposure of the teat orifice. The percentage of teat ducts colonized with C. bovis which became infected with either pathogen was not different from that for controls. 相似文献

Fifteen pregnant Holstein cows were freely assigned to 3 experimental groups (5 cows in each group). Cows in group I were inoculated IM and intramammarily (IMm) with Ohio Agricultural Research and Development Center (OARDC) tissue culture-propagated modified-live Nebraska calf diarrhea bovine rotavirus with added adjuvant (OARDC vaccine-immunized cows). Group II cows were given IM injections of a commercial modified-live rotavirus-coronavirus vaccine (commercial vaccine-immunized cows), and the remaining 5 cows were noninoculated controls (group III). Rotavirus antibody in colostrum and milk was mainly associated with immunoglobulin (Ig)G1, and less so with IgG2, IgA, and IgM, as analyzed by the enzyme-linked immunosorbent assay (ELISA), using monospecific anti-bovine IgG1, IgG2, IgM, and IgA sera. In serum, the rotavirus antibody was distributed almost equally between IgG1 and IgG2. The same relationships appeared in both immunized and nonvaccinated cows. All OARDC vaccine-injected cows had virus-neutralization (VN) and ELISA IgG1 rotavirus antibody titers in serum and mammary secretions at significantly increased levels (at least 100-fold; P less than 0.05) compared with the titers in groups II (commercial vaccine-immunized cows) and III (controls). Serum, colostrum, and milk antibody titers from these latter 2 groups did not differ statistically. The ELISA IgG2, IgA, and IgM rotavirus antibody titers also were significantly greater in mammary secretions from OARDC vaccine-immunized cows than in groups II and III cows. There was a high correlation between ELISA IgG1 and VN rotavirus antibody titers for all samples tested (r = 0.97, P less than 0.001), but ELISA IgG1 antibody titers were consistently higher than VN titers. The ELISA IgG1 and VN antibody titers of milk samples collected from cows 30 days after parturition were higher from the OARDC vaccine-immunized cows (ELISA IgG1, geometric mean titer (GMT) = 3,511; VN GMT = 1,689) than were titers from the group II cows (ELISA IgG1 GMT = 39; VN GMT = 33) or group III cows (ELISA IgG1 GMT = 21; VN GMT = 19). These results indicate that IM plus IMm immunization of pregnant cows, using modified-live bovine rotavirus with added adjuvant, may significantly enhance serum, colostrum, and milk rotavirus antibody titers, whereas IM vaccinal inoculation of pregnant cows with a commercial modified-live rotavirus-coronavirus vaccine may not. 相似文献

Evidence is reviewed that indicates Ni is an essential element for the chick, rat, pig, sheep and goat. Although a number of possible functions for Ni have been proposed based on in vitro and in vivo studies, the physiological role of Ni in the mammalian or avian system is presently unknown. Rumen bacterial urease has been shown to be a Ni-dependent enzyme and Ni is a component of factor F430 present in methanogenic bacteria. Nickel can interact or influence the metabolism of a number of minerals. Interactions of Ni and Fe, Zn and Cu are discussed. The requirement for Ni is low (50 to 60 ppb) in chicks fed semipurified diets. Insufficient data are available to estimate the Ni requirement of swine. In ruminants, the Ni requirement appears to be higher than that for other animal species. Nickel supplementation to practical diets has increased gain, feed efficiency and ruminal urease activity in ruminants, but performance results have been inconsistent. Level of crude protein and urea are two factors that influence ruminant responses to dietary Ni. The greatest responses have been observed in ruminants fed low protein diets. Little is known concerning levels, forms and bioavailability of Ni in different feedstuffs. Nickel is homeostatically controlled in the animal's body and high levels of Ni are required to cause toxicity. 相似文献

Identification and measurement of canine factor VIII-related antigen (VIIIR:Ag) has many clinical and research applications, including differential diagnosis of hemophilia A and von Willebrand's disease, use as a marker for specific cell types, and elucidation of the structure of this factor VIII component. We have developed a practical method for producing antibodies to canine VIIIR:Ag that uses 2% agarose filtration for purification and identifies the antigen by correlation with the elution of the peak A280 fraction in the void volume. Antisera for electroimmunoassay (EIA) can be produced in less than four weeks from simple starting materials and with commonly available laboratory equipment. Paraffin sections of the carotid rete-cavernous sinus complex of sheep were studied, using different stains. The carotid rete of sheep was composed of medium-sized arteries with smooth muscle layers that were oriented in different directions. The carotid body cells may have migrated proximally in the adventitia of the intracranial portion of the internal carotid artery as its extracranial portion degenerates early in life. The cavernous sinus shared a common tunica adventitia with surrounding rete branches. At places, the wall of the cavernous sinus had a distinct tunica media interposed between the endothelial cells and the tunica adventitia. An in vitro assessment based on tissue responsiveness to 2 agonists-histamine and carbachol-was made on smooth muscle activity in chickens experimentally poisoned with sesbania. Crude extracts of Sesbania drummondii were prepared and 2 dosage levels, 0.25% and 0.5% of body weight, were used. The birds were dosed orally with the extract for 3 consecutive days, and on the 4th day segments of ileum and lung were collected from each bird. The isometric contractions of each tissue, produced by the addition of histamine or carbachol in graded concentrations, were recorded. The cumulative concentration-effect curves for the tissues to the agonists were constructed and compared with respective control curves. The results indicated the responsiveness of the tissues in the treated groups was significantly decreased, compared with that of tissues in the controls. Responses of both intestinal and parenchymal strips in the chickens given the higher dosage (0.5%) were decreased significantly, whereas in those given the smaller dosage (0.25%), only parenchyma had a significant response. This indicates that the activity of smooth muscles in general was depressed by sesbania. The effect was more evident in the lung than in the intestine. Therefore, an active principle in the extract which affects smooth muscle, rather than causing direct irritation, may exist. This assessment of smooth muscle activity is sensitive and was effective in detecting changes in tissues from sesbania-treated birds which had not shown any clinical signs. The results also support the possibility that smooth muscle involvement could be a primary cause of toxicity in sesbania poisoning. 相似文献

Surgical incisions were made in the skin and superficial digital flexor tendons in horses. A low intensity laser therapy device was used to treat the limbs. After laser therapy was completed, skin and superficial digital flexor tendons from incised laser-irradiated, incised-control, and nonincised-control limbs were compared microscopically. Qualitative differences between laser irradiated and nonirradiated tissues were not found. The laser therapy device used in this study may be of insufficient power to affect wound healing. 相似文献

Antibody responses were compared in guinea-pigs, rabbits and pigs following vaccination with inactivated porcine parvovirus (PPV) vaccines. Mean PPV hemagglutination inhibition (HI) antibody titers of 52, 56 and 36 at 1 week after first vaccination and 896, 640 and 512 at 2 weeks after second vaccination were detected in guinea-pigs, rabbits and pigs, respectively. PPV vaccines prepared with greater concentrations of virus, as determined by hemagglutination (HA) units, and of aluminum hydroxide gel adjuvant, induced higher HI antibody titers in guinea-pigs. Optimal concentrations for inducing consistently high antibody titers consisted of vaccine virus with a HA titer of 256/0.1 ml and gel adjuvant at a final concentration of 50%. A second vaccination at 4 weeks compared to 2 or 3 weeks after first vaccination resulted in higher mean HI titers. These data provide preliminary information on the use of guinea-pigs or rabbits as laboratory animal models for testing the potency of PPV vaccines. 相似文献
