Meiotic recombination in budding yeast requires two RecA-related proteins, Rad51 and Dmc1, both of which form filaments on DNA capable of directing homology search and catalyzing formation of homologous joint molecules (JMs) and strand exchange. With use of a separation-of-function mutant form of Rad51 that retains filament-forming but not JM-forming activity, we show that the JM activity of Rad51 is fully dispensable for meiotic recombination. The corresponding mutation in Dmc1 causes a profound recombination defect, demonstrating Dmc1's JM activity alone is responsible for meiotic recombination. We further provide biochemical evidence that Rad51 acts with Mei5-Sae3 as a Dmc1 accessory factor. Thus, Rad51 is a multifunctional protein that catalyzes recombination directly in mitosis and indirectly, via Dmc1, during meiosis. 相似文献

Enzyme activities in semiarid soils under conservation reserve program,native rangeland,and cropland
There is limited knowledge of biochemical processes in low carbon content soils of semiarid regions under different land use and management. This study investigated several enzyme activities of C, N, P, and S transformations in semiarid soils with different clay (10–21 %) and sand (59–85 %) contents that were under conservation reserve program (CRP), native rangeland (NR), and cropland (CL) under sunflowers (Eriophyllum ambiguum (Gray)), continuous cotton (Gossypium hirsutum L.), or in rotations with wheat (Triticum aestivum L.) or sorghum (Sorghum bicolor L.) in West Texas, USA. Soils under CRP and NR showed higher total C and N contents than cultivated soils under continuous cotton, but soil pH (6.7–8.4) was not affected by the management or land use studied. The activities of β‐glucosidase, β‐glucosaminidase, arylamidase, acid and alkaline phosphatase, phosphodiesterase, and arylsulfatase (mg product (kg soil)–1 h–1) were lower in CL under continuous cotton compared to cotton in rotation with other crops, CRP, and NR. The enzyme activities were also lower when compared to soils from other regions. Linear regression analyses indicated positive correlations between enzyme activities and total C (r values up to 0.96, P < 0.01). There was a positive relationship between enzyme activities and total N, but soil pH showed the opposite trend. Enzyme activities were significantly intercorrelated with r values up to 0.98 (P < 0.001). The specific enzyme activities (mg product (g organic C)–1) were lower in continuous cotton in comparison to the uncultivated soils (i.e., NR and CRP) reflecting differences in organic matter quantity and quality due to cultivation. Among the enzymes studied, the specific activities of β‐glucosidase and arylamidase showed a more pronounced decrease with increasing soil depth. In general, soils under CRP or wheat‐cotton rotations revealed higher enzyme activities than soils under the common agricultural practice for these regions, i.e., continuous cotton under conventional tillage. 相似文献

The effect of recovery from training has not been studied in horses. Therefore, the effect of recovery was examined with exercise of known effect within a conditioning period (CP). A standardized exercise test was performed at the beginning of CP to determine v4, v10, and v180 (horse’s speed, which produced a blood lactate concentration of 4 and 10 mmol/L and a heart rate of 180 beats/min). Six horses were conditioned for three periods of 2 weeks, 5 times per fortnight at their individual v10 for two bouts of 5 minutes on a treadmill. Every 2 weeks of conditioning was followed by 1 week with reduced workload. Standardized exercise test was repeated after each 2 weeks of conditioning and 2 weeks after finishing CP. Exercise speed was individually adapted to the new v10 for every 2 weeks of conditioning. In addition, peak oxygen consumption before, after 3 weeks of conditioning, and at the end of the CP was measured. The mean v4 increased steadily during CP. v180 did not change, whereas peak oxygen consumption increased between the beginning and after 3 weeks of conditioning and leveled off thereafter. In conclusion, reducing the workload for 1 week after 2 weeks of conditioning 5 times per fortnight at v10 for two bouts of 5 minutes allowed for a continuous increase of v4, but the extent of the increase was smaller than in another study with a similar conditioning program but for the recovery week. Uncultivated plants are an important part of agricultural systems and play a key role in the survival of rural marginalized
groups such as women, children, and the poor. Drawing on the gender, environment, and development literature and on the notion
of women’s social location, this paper examines the ways in which gender, ethnicity, and economic status determine women’s
roles in uncultivated plant management in Ixhuapan and Ocozotepec, two indigenous communities of Veracruz, Mexico. The first
is inhabited by Nahua and the second by Popoluca peoples. Information was gathered through group and individual interviews
and a food frequency survey. Results show that the gender ideology prevailing in each community, resulting from distinct ethnic
affiliations and economic contexts, shapes women’s plant management. In Ixhuapan, Nahua women are used to leaving their community
to generate income, while in Ocozotepec men are considered the main breadwinners and are the mediators between Popoluca households
and the larger society. Nahua women gather quelites at the cornfields more often than their men, and more often than their female counterparts in Ocozotepec. They also manage
and sell plants from their homegardens at higher percentages than Popoluca women. However, women in both communities use intensely
the plants of their homegardens and play a key role in biodiversity conservation and cultural permanence.

The availability of iron is known to exert a controlling influence on biological productivity in surface waters over large areas of the ocean and may have been an important factor in the variation of the concentration of atmospheric carbon dioxide over glacial cycles. The effect of iron in the Southern Ocean is particularly important because of its large area and abundant nitrate, yet iron-enhanced growth of phytoplankton may be differentially expressed between waters with high silicic acid in the south and low silicic acid in the north, where diatom growth may be limited by both silicic acid and iron. Two mesoscale experiments, designed to investigate the effects of iron enrichment in regions with high and low concentrations of silicic acid, were performed in the Southern Ocean. These experiments demonstrate iron's pivotal role in controlling carbon uptake and regulating atmospheric partial pressure of carbon dioxide. 相似文献

This study compared co-feeding zooplankton (ZP, mixed copepods and mysids) and formulated diets (FD) supplemented with L-tryptophan (TRP) on the survival and growth of grouper and sea bass fry nursed in brackish water ponds. Grouper (84 fry m?3) and sea bass (150 fry m?3) were reared for 30 days and 60 days, respectively, in net cages within two separate 743 m2 nursery ponds. Five treatments (with three replicates each) were compared (P < 0.05): FD-1 = ZP + basal FD (no added TRP, but containing 0.29% endogenous TRP); FD-2 = ZP + (FD 0.58% TRP); FD-3 = ZP + (FD 1.22% TRP); FD-4 = ZP + (FD 2.50% TRP); and FD-5 = minced fish (Sardinella sp.) + basal FD (no additional TRP). TRP supplementation in grouper diets produced no significant affect on growth but increased survival at rates of 0.58% and above. TRP supplementation at 2.5% (FD-4) produced significantly better sea bass growth than other diets but had no affect on survival. Zooplankton improved both survival and growth in both grouper and sea bass juveniles compared to the minced fish diet, and may be a practical and lower cost alternative to indoor nursing. 相似文献

This article studies the relationship between economic growth and income inequality using panel data for a sample of Latin American countries in the period 1960–2000. Different estimation techniques and specifications are tested. Results indicate that the level of inequality has had a different effect on growth in the region, depending on the level of gross domestic product per capita. Even within a sample of developing countries, the effect of inequality on growth is negative for poorer countries, whereas for richer ones higher inequality may favour economic growth. Overall, our results are not robust to different specification techniques, and coefficients are in some cases only weakly significant, underlying the difficulties in finding a strong and robust relationship between income inequality and economic growth. 相似文献

The USDA‐ARS National Plant Germplasm System maintains a Zimbabwe sorghum collection of 1235 accessions from different provinces. This germplasm has not been extensively employed in US breeding programmes due to the lack of phenotypic and genetic characterization. Therefore, 68 accessions from Zimbabwe were phenotyped, and evaluated for their anthracnose response for two consecutive years, and genetically characterized with 21 simple sequence repeat markers. Phenotypic analysis showed significant differences among accessions with plant height and panicle length being the most variable traits. Likewise, 25 accessions were anthracnose resistant, nine showed variable responses and 34 were susceptible. Genetic analysis identified 174 alleles with an average of 8.3 alleles and 11.8 genotypes per locus and a polymorphic information content of 0.60. These results reflect a moderate genetically diverse germplasm. Neighbour‐joining clustering analysis revealed that the majority of anthracnose‐resistant accessions showed high genetic relatedness; therefore, this germplasm might represent one to six new sources of resistances. Results presented herein show that the Zimbabwe collection contains valuable germplasm for breeding programmes and is an important source of anthracnose resistance. 相似文献

Understanding the mechanisms that maintain protective antibody levels after immunisation is important for vaccine design. In this study, we have determined the kinetics of plasma and memory B cells detectable in the blood of cattle immunised with model T-dependent or T-independent antigens. Immunisation with the T-D antigen resulted in an expansion of TNP-specific plasma cells post-TNP primary and booster immunisations, which was associated with increased titres of TNP-specific IgG antibodies. Although no TNP-specific memory B cells were detected in the T-D group following the primary immunisation, we detected an increase in the number of TNP-specific memory B cells post-TNP boost. In contrast, no TNP-specific plasma or memory B cells were detected after primary or secondary immunisation with the T-I antigen. We then investigated if immunisation with a third party antigen (tetanus toxin fragment C, TTC) would result in a bystander stimulation and increase the number of TNP-specific plasma and memory B cells in the T-D and/or T-I group. TTC immunisation in the T-D group resulted in a small increase in the number of TNP-specific plasma cells post-TTC primary immunisation and boost, and in an increase in the number of TNP-specific memory B cells post-TTC boost. This bystander effect was not observed in the animals previously immunised with the T-I antigen. In conclusion, the present study characterised for the first time the B cell response in cattle to immunisation with T-D and T-I antigens and showed that bystander stimulation of an established T-D B cell memory response may occur in cattle. 相似文献
