An international test performance study (TPS) was organised to generate validation data for three molecular Synchytrium endobioticum tests: van den Boogert et al. (European Journal of Plant Pathology 113, 47–57, 2005), and van Gent-Pelzer et al. (European Journal of Plant Pathology, 126, 129-133, 2010) for the detection of S. endobioticum, and the pathotype 1(D1) identification test described by Bonants et al. (European Journal of Plant Pathology, 143, 495-506, 2015). Two TPS rounds were organised focussing on different test matrices, i.e. round 1: warted potato tissue, and round 2: resting spore suspensions. When using the tests for detection and identification of S. endobioticum in warted potato tissue, no significant differences were observed for diagnostic sensitivity, diagnostic specificity, overall accuracy, analytical sensitivity and robustness. When using the tests for detection and identification of S. endobioticum in resting spore suspensions, the van den Boogert and van Gent-Pelzer tests significantly outperform the Bonants test for diagnostic sensitivity and diagnostic specificity. For overall accuracy and analytical sensitivity, the van Gent-Pelzer significantly outperforms the van den Boogert and Bonants tests and is regarded as the test of choice when identifying S. endobioticum from resting spores. Tests regarded fit for purpose for routine testing of wart material and resting spore suspensions are proposed for the update of EPPO standard PM7/28(1) Synchytrium endobioticum. 相似文献
It is widely documented that a pool of multipotent stem cells located in humans and mice hair follicle outer root sheath (bulge region) is involved in the restoration of the whole follicular unit during each anagen phase. To the authors' knowledge, data regarding the location and characterization of hair follicle stem compartment in dogs have not been reported in the recent relevant literature. In this study, we investigated the haematopoietic stem and progenitor cell antigen CD34 as a marker of putative stem cells located in a bulge-like region of canine hair follicles. The presence of CD34 mRNA and glycoprotein was assessed on formalin-fixed, paraffin-embedded canine skin samples by in situ hybridization technique and by standard immunohistochemistry, respectively. A strong expression of CD34 mRNA and glycoprotein was observed in a well-defined area of the hair follicle isthmic region and appeared uniformly concentrated at the level of the basal layer of the outer root sheath. These findings provide compelling support to the hypothesis that in dogs, a subpopulation of basal keratinocytes located in the hair follicle isthmic region and characterized by the selective expression of CD34 is potentially associated with the stem cell compartment of this skin appendage. 相似文献
In this work the feasibility of extracting cellulose from cotton, sisal and flax fibers, corn stover and rice husk by means of usual chemical procedures such as acid hydrolysis, chlorination, alkaline extraction, and bleaching was analyzed. Cellulose nanowhiskers from these sources, and from commercial cellulose, were produced by the acid hydrolysis of the obtained celluloses. The final products were characterized by means of Thermogravimetric Analysis (TGA), Infrared Spectroscopy (FTIR), X-ray Diffraction (XRD), Scanning Electronic Microscopy (SEM) and Atomic Force Microscopy (AFM). The chemical procedure used to obtain cellulose nanowhiskers was effective in all cases but differences on the thermal stability, chemical composition, crystallinity and morphology were found due to the dissimilar nature of the different sources. Thus, this work demonstrates that the morphology and physical properties of cellulose nanowhiskers synthesized by the same conditions are strongly dependent on their source. 相似文献
Asparagus fly (Plioreocepta poeciloptera (Schrank, 1776)) is a serious pest in German asparagus (Asparagus officinalis L.) production. To evaluate the effects of different routine on-farm soil tillage measures on the number of flies emerging the following spring, asparagus fields in Lower Saxony, Germany, were investigated. Soil samples were taken before and after tillage in autumn 2017 and autumn 2018. Investigations were also conducted in both years on the effect that the soil depth at which asparagus fly pupae were buried had on the emergence of adult flies.
This study revealed that the number of emerging flies was not reduced by mulching, but was significantly reduced by subsequent tillage and/or tillage and dam formation. The emergence rate of adult flies was significantly reduced the deeper the pupae had been buried the previous autumn. The effects also depended on the year. The highest mean emergence rate observed was 68% and 45% for pupae buried at a depth of 10?cm and 20?cm, respectively. In conclusion, the key mechanism causing a decrease in asparagus fly population the following spring through routine on-farm tillage could be the burial of pupae when forming dams. Routine on-farm soil tillage can be regarded as a physical measure for controlling asparagus fly and is therefore an essential tool in the integrated pest management of asparagus production.
The stem bark of Acacia oxyphylla Graham ex Bentham is used as an anthelmintic by the natives of Mizoram (North-East India). Therefore, the aim of the study was to assess the effect of the active compound isolated from A. oxyphylla on the tegument of adult Raillietina echinobothrida and Ascaridia galli. The test parasites R. echinobothrida and A. galli were incubated in physiological buffered saline containing 0.0005, 0.001, 0.05, 0.1 and 1mg/ml of the isolated compound. The alterations in the tegument of the parasites post paralysis were examined using electron microscopes. The compound reduced the cestode's motility soon after incubation, but did not induce paralysis in the nematodes till about 11-14 h at highest concentration. The compound caused extensive digestion of cestode tegument as evident by electron microscopy. Disorganization of muscle bundles, loss of cell-cell contact, extreme vacuolization and oedema were some of the changes observed. Loss of cellular organelles combined with distortion of those present was markedly noted throughout the parasite tissue. Deformation and disorganization of epicuticle, disruption of mitochondrial and nuclear membrane were also observed in nematode exposed to the active compound of the plant. Substantial structural deformities in the treated parasites are indicative of an efficient vermicidal activity of the isolated compound against cestodes and nematodes. 相似文献
