Drosophila suzukii (Diptera: Drosophilidae) is an invasive alien species devastating soft fruit crops in newly invaded territories. Little is known about the importance and potential of long-distance dispersal at a regional scale. The goal of this work is to investigate D. suzukii dispersal ability during different times of the season, and along an elevational gradient in a mountain valley in Trentino Province, Italy. We employed a mark–release–recapture strategy using protein markers. Flies were recaptured using fruit-baited traps. The protein-marked flies were positively identified using ELISA procedure. Additional microsatellite analyses were performed on D. suzukii collected during autumn at different elevations to characterize the population structure. Results suggest that a portion of the local D. suzukii population moves from low to high elevations during spring and summer and travels back to low elevations in autumn. Genetic analysis further revealed that samples collected during autumn at different elevations belong to the same population. These results show that D. suzukii are able to fly up to about 9000 m away from the marking point and that seasonal breezes likely facilitate long-distance movement. We suggest that these migrations have multiple functions for D. suzukii, including conferring the ability to exploit gradual changes of temperature, food, and ovipositional resources in spring and autumn, as well as to assist in the search for suitable overwintering sites in late autumn. Our findings help to unveil the complex ecology of D. suzukii in Italian mountainous regions and provide important clues for improving the efficacy of integrated pest management control techniques to combat this pest.  相似文献

Biofumigation is a biocontrol method that uses volatile compounds to combat soil pathogens. We investigated a biofumigation process based on the green manure of Brassicaceae. These plants contain the glucosinolate-myrosinase system which releases inhibitory compounds such as isothiocyanate into the soil. However, the biocontrol effectiveness is often lower than expected, possibly due to microbial transformation of the isothiocyanates. In order to identify the possible function of microorganisms, their interaction with glucosinolates and glucosinolate-derived products was investigated. We report the ability of a soil isolate of Aspergillus flavus to grow in liquid culture and convert 2-propenyl and 2-phenylethyl glucosinolate and their desulfo-derivatives, to nitriles. This finding would suggest the existence of an arylsulfatase and a β-thio-glucosidase, different from myrosinase, which could direct glucosinolate conversion in soil towards nitriles rather than isothiocyanates. If confirmed in soil, our observations could at least partially explain the low concentrations of isothiocyanates after biofumigation.  相似文献

In the last decades, it has been demonstrated that marine organisms are a substantial source of bioactive compounds with possible biotechnological applications. Marine sponges, in particular those belonging to the class of Demospongiae, have been considered among the most interesting invertebrates for their biotechnological potential. In this review, particular attention is devoted to natural compounds/extracts isolated from Demospongiae and their associated microorganisms with important biological activities for pharmacological applications such as antiviral, anticancer, antifouling, antimicrobial, antiplasmodial, antifungal and antioxidant. The data here presented show that this class of sponges is an exciting source of compounds, which are worth developing into new drugs, such as avarol, a hydroquinone isolated from the marine sponge Disidea avara, which is used as an antitumor, antimicrobial and antiviral drug.  相似文献

Sea urchin gonads are exploited both for gastronomic and scientific purposes; thus, the production of large and mature gonads is essential. Natural populations of the Mediterranean Sea urchin Paracentrotus lividus are subject to increasing fishing efforts, due to continuous intensification of consumptions. Aquaculture practices may represent an answer, but the availability of artificial feeds accelerating the production of high‐quality gonads in terms of size, taste, colour, firmness, maturity and viability of gametes is critical to boost the productions. The accessibility of cheap and effective feeds promoting the fattening of gonads and the quality of gametes is still a bottleneck slowing down the expansion of echinoculture practices. This investigation is aimed at enabling the development of this strategic sector, by comparing the dietetic effects of fresh foods and a commercial feed for aquaculture, to four newly formulated feeds. The protein contents of diets were strongly related to the GSIs. The abundance of fatty acids appeared inversely related to the viability of embryos and abnormalities of larvae. The features of an ideal diet for this sea urchin were defined, based on the results of experimental trials, and the need for increasing levels of plant‐derived proteins during the grow‐out period was demonstrated.  相似文献

Heavy chain antibodies (HCAbs), devoid of the light chains and the CH(1) domain, are present in the serum of camelids. IgG(2) and IgG(3) are HCAbs; whereas IgG(1) has the conventional structure. In order to study the immunological properties of llama HCAbs, from which to date little is known, llamas (Lama glama) HCAbs cDNA were cloned, sequenced and compared with other mammalian Igs. The sequence analysis showed that llama HCAbs cDNA organization is similar to other mammalian Igs and the presence of conserved binding motifs to Protein A, Protein G, FcγRI, FcγRIII and C1q in HCAbs were observed. In a previous work, different IgG isotypes purified by Protein A and Protein G chromatography, were assayed for their ability to fix complement. Both IgG(1) and the total serum were able to fix complement, whereas IgG(2) and IgG(3) fixed complement even in the absence of antigen (anti-complementary activity). Therefore, in this work we performed the complement activating activity of the different IgG isotypes purified under physiological conditions using Sephadex G-150 and their ability to induce hemagglutination. Llamas were immunized with sheep red blood cells (RBC) stroma and the different isotypes were purified from sera. Whole serum and IgG(1) could activate complement; however, HCAbs (IgG(2)+IgG(3)) could not, despite the presence of the C1q binding motif in their primary sequence. Unlike IgG(1), the fraction corresponding to IgG(2)+IgG(3) did not display hemagglutinating activity. Our findings suggest that HCAbs cannot crosslink efficiently with different antigens and that the C1q binding site might be hindered by the proximity of the variable domains.  相似文献

It has been reported that various cultivars of fruits and vegetables may present a different pattern for the contained allergens. Here, we report on the different content in allergenic proteins for different peach (Prunus persica) cultivars, sampled during two consecutive harvest seasons. Fruits from six cultivars of peaches were harvested fully ripe, and the proteins extracted from whole or chemically peeled fruits were analyzed by SDS-PAGE and immunoblotting. All the protein extracts from whole fruit contained a 9 kDa protein. This protein proved to be absent in the extracts taken from chemically peeled fruit. In four cultivars, this protein corresponds to the allergen Pru p3, a lipid transfer protein that causes the oral allergy syndrome (OAS) in sensitized people. In the following year, fruits from four of the six cultivars of peaches studied previously were harvested at different times, at one and two weeks before the commercial ripening time and when fully ripe, to ascertain whether the presence of the 9 kDa allergen might be related to the ripening process. Two cultivars out of four produced an intense allergenic band corresponding to a 9 kDa protein already two weeks before the commercial ripening date, while the others showed a progressive increment of the 9 kDa allergen during ripening.  相似文献

(5R)-5-Vinyl-1,3-oxazolidine-2-thione, (2S)-1-cyano-2-hydroxy-3-butene, and two diastereoisomeric erythro-(2S)- and threo-(2S)-1-cyano-2-hydroxy-3,4-epithiobutanes were prepared in pure form starting from (2S)-2-hydroxybut-3-enyl glucosinolate (epiprogoitrin). This glucosinolate was isolated in almost pure form using ripe seeds of Crambe abyssinica and then hydrolyzed under different conditions. The hydrolysis was carried out using either myrosinase immobilized on nylon, to produce (5R)-5-vinyl-1,3-oxazolidine-2-thione, or the endogenous myrosinase contained in defatted crambe meals, to produce the other epiprogoitrin-derived products. After purification and physicochemical characterization, all four myrosinase degradation products were tested for their biological activity. A bioassay on Lactuca sativa was chosen as a simple test to determine their apparent action on living tissues. (5R)-5-Vinyl-1,3-oxazolidine-2-thione negatively affected mainly root growth, whereas (2S)-1-cyano-2-hydroxy-3-butene affected the early phase of germination, and both (2S)-1-cyano-2-hydroxy-3,4-epithiobutane diastereoisomers appeared to negatively affect both germination and root growth at doses 5-10 times lower than those of (2S)-1-cyano-2-hydroxy-3-butene or (5R)-5-vinyl-1,3-oxazolidine-2-thione.  相似文献

Cyanogenic plants have some potential as biocidal green manure crops in limiting several soilborne pests and pathogens. Sorghum (Sorghum bicolor (L.) Moench) and Sudangrass (Sorghum bicolor subsp. sudanense (P.) Stapf), in fact, contain the cyanogenic glucoside p-hydroxy-(S)-mandelonitrile-β-D-glucoside (dhurrin) as a substrate of its secondary defensive system able to release hydrogen cyanide following tissue lesions due to biotic or abiotic factors. Given that dhurrin content is correlated with the biofumigant efficacy of the plants, a high dhurrin content could be a positive character for utilization of sorghum and Sudangrass as biocidal green manure plants. For chemical characterization of the available germplasm, a simple, safe, and accurate method is necessary. In this paper, a new method for dhurrin analysis, based on methanol extraction and high-performance liquid chromatography, is reported and discussed. The feasibility of this analytical procedure was tested by evaluating dhurrin level in roots and stems during cultivation of four different sorghum and Sudangrass varieties in agronomic trials performed in 2008 in the Po valley (Italy). The dhurrin content ranged from 0.16 ± 0.04 to 7.14 ± 0.32 mg g(-1) on dried matter (DM) in stems and from 1.38 ± 0.02 to 6.57 ± 0.09 mg g(-1) on DM in roots, showing statistical differences among the tested germplasms that could be linked to the efficacy of their utilization as biofumigant plants. The method also opens new perspectives for the characterization of sorgum plants as fodder, for which the presence of dhurrin is considered to be negative for its well-known toxicity.  相似文献
